A DNA Based Sensor Array for Label-Free Fluorescent Pattern Identification of Metal Contaminated Water

Abstract:Development of convenient method for monitoring toxic metal pollution is of practically significant for the environment and public health. Pattern-based fluorescent sensor array has attracted great attention owing to their potential to differentiate and detect multiple metal ions simultaneously with only few probes. However, it remains a challenge to improve the time, cost and synthesis simplicity to construct the pattern recognition elements. In this work, a fluorescent library consisted of lanthanide/antenna/DNA complex was facilely built by variation both commercially-available antenna and DNA sequence. Through selection of complex from the library, a novel sensor array that can provide fluorescent-on or fluorescent-off based cross-reactive responses to a series of metal ions to realize pattern recognition ability has been developed. The combination of metal ions binding property and lanthanide fluorescence sensitization process empowers the present sensor ensemble with good discrimination ability, which could well differentiate 11 metal ions, including Hg 2+ , Cr 3+ , Fe 3+ , Pb 2+ , Cu 2+ , Cr 6+ , Zn 2+ , Co 2+ , Al 3+ , Cd 2+ , and Ag + . Moreover, this sensing array could be further applied for discriminating of clean and single or multiple metal ions polluted drinking water, indicating their application potential for water quality monitoring.
