Abstract:<p>Carbon dots (CDots) are strongly absorptive over the visible spectrum, with the effective photon-harvesting driving rich excited state processes and properties. In this work, spectroscopic probing of these processes and properties is coupled with the evaluation of the photoinduced bactericidal function of CDots, aimed toward making correlations among the findings from the former and those from the latter on the inactivation of bacterial pathogens. Within the general mechanistic framework for CDots, the observed effective and efficient antibacterial activities of the CDots under visible light are attributed to major contributions by the initially photo-generated electrons and holes that are trapped at passivated surface defect sites of the dots, in addition to the traditional reactive oxygen species (ROS) produced in the emissive excited states from the recombination of the redox pairs. Such major contributions to the inactivation of the bacteria by the separated redox species in CDots can not be quenched by ROS scavengers commonly used in the study of photodynamic effects with classical molecular photosensitizers, thus making light-activated CDots uniquely potent antimicrobial agents. The characteristic features of photoexcited CDots and their related mechanistic implications are discussed in reference to the similar behaviors and mechanistic model of conventional semiconductor quantum dots.</p>

The dominant role of surface functionalization in carbon dots' photo-activated antibacterial activity

Abstract:Background: Carbon dots (CDots) have recently been demonstrated their effective visible light-activated antimicrobial activities toward bacteria. This study was to evaluate and understand the roles of the surface functionalities in governing the antimicrobial activity of CDots. Methods: Using the laboratory model bacteria Bacillus subtilis, the photo-activated antimicrobial activities of three groups of CDots with specifically selected different surface functionalization moieties were evaluated and compared. The first group consisting of CDots with surface functionalization by 2,2-(ethylenedioxy)bis(ethylamine) (EDA) vs. 3-ethoxypropylamine (EPA), was evaluated to determine the effect of different terminal groups/charges on their photo-activated antibacterial activities. The second group consisting of CDots functionalized with oligomeric polyethylenimine (PEI) and those prepared by the carbonization of PEI - citric acid mixture, was to evaluate the effects of dot surface charges vs. fluorescent quantum yields on their antimicrobial activities. The third group consisting of CDots functionalized with PEI of 1,200 vs. 600 in average molecular weight was evaluated for the effect of molecular weight of surface passivation molecular on their antimicrobial activities. Results: The results indicated the EDA-CDots in the first group was more effective and was attributed to the positive charges from the protonation of the amino groups (-NH2) being more favorable to interactions with bacterial cells. The evaluation of the second group CDots suggested the same surface charge effect dominating the antibacterial performance over the fluorescent quantum yields. The evaluation of the third group CDots functionalized with PEI of 1,200 vs. 600 in average molecular weight, indicated the latter was significantly more effective. Conclusions: The results from this study highlighted the dominant role of surface functionalities in governing CDots' light activated antimicrobial activity and should have significant implications to the further design and development of CDots as a new class of visible light-activated antibacterial agents.

Abstract:Carbon–semiconductor hybrid quantum dots are classical carbon dots with core carbon nanoparticles doped with a selected nanoscale semiconductor. Specifically, on those with the nanoscale TiO2 doping, denoted as CTiO2-Dots, their synthesis and thorough characterization were reported previously. In this work, the CTiO2-Dots were evaluated for their visible light-activated antibacterial function, with the results showing the effective killing of not only Gram-positive but also the generally more resistant Gram-negative bacteria. The hybrid dots are clearly more potent antibacterial agents than their neat carbon dot counterparts. Mechanistically, the higher antibacterial performance of the CTiO2-Dots is attributed to their superior photoexcited state properties, which are reflected by the observed much brighter fluorescence emissions. Also considered and discussed is the possibility of additional contributions to the antibacterial activities due to the photosensitization of the nanoscale TiO2 by its doped core carbon nanoparticles.

Abstract:Graphical and Lay Summary Shining carbon dots (CDs) produced by a single‐step hydrothermal method can afford enhanced antibacterial activity against both Gram‐positive and Gram‐negative bacteria. Mechanistic insights reveal that the CDs generate singlet oxygen (1O2) under light irradiation, contributing to an augmented ROS level, facilitating disruption of biofilms and inhibition of biofilm formation. In spite of tremendous efforts dedicated to addressing bacterial infections and biofilm formation, the post‐antibiotic ear continues to witness a gap between the established materials and an easily accessible yet biocompatible antibacterial reagent. Here we show carbon dots (CDs) synthesized via a single hydrothermal process can afford promising antibacterial activity that can be further enhanced by exposure to light. By using citric acid and polyethyleneimine as the precursors, the photoluminescence CDs can be produced within a one‐pot, one‐step hydrothermal reaction in only 2 h. The CDs demonstrate robust antibacterial properties against both Gram‐positive and Gram‐negative bacteria and, notably, a considerable enhancement of antibacterial effect can be observed upon photo‐irradiation. Mechanistic insights reveal that the CDs generate singlet oxygen (1O2) when exposed to light, leading to an augmented reactive oxygen species level. The approach for disruption of biofilms and inhibition of biofilm formation by using the CDs has also been established. Our findings present a potential solution to combat antibacterial resistance and offer a path to reduce dependence on traditional antibiotics.

Abstract:Owing to merging antibiotic resistance, the synthesis of novel antibacterial agents becomes increasingly important. Carbon dots (CDs) belong to a new class of carbon-based nanomaterials that have shown great potential in combating emerging and drug-resistant infections due to their unique optical properties, such as excellent biocompatibility and easy surface modification. Herein, light-treated CDs (LCDs) with enhanced antibacterial activity and low drug resistance were synthesized by using a simple one-pot hydrothermal method. LCDs can generate reactive oxygen species (ROS) under the light-emitting diode light, which kill two pathogenic bacteria, including methicillin-resistant Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli). In addition, in vitro experiments show that the liquid crystal displays excellent biocompatibility. Due to the visible photo-oxidative activity, LCDs can be used as photodynamic therapy agents, providing efficient bactericidal activity. This study provides a simple way to modify CDs that lead to new properties.

Abstract:Bacterial contamination poses a serious and global threats to public health. Due to broad-band antimicrobial capacity and unspecific tissue interaction, antibacterial nanomaterials offer promising alternative and green strategies to resist bacterial contamination. Herein, an enhanced metal-free carbon-dots nanozymes, i.e., m-aminophenol based phosphoric acid doping carbon dots (P-CDs), with excellent photoresponse enzyme-like properties are designed through a simple one-pot hydrothermal method for highly effective antimicrobial. Developed P-CDs exhibited superior photoresponse oxidase-like activity, which may attribute to the higher crystallinity conferred by P-doping. Interestingly, the generation of superoxide radicals (O2 •−) under photocatalytic for such P-CDs exhibited excellent antimicrobical effects with a high maximum reaction rate (33.3 × 10-8 M/s). Notably, P-CDs effectively inhibited the growth of both Gram-positive and Gram-negative bacteria under a 1 W white LED irradiation, especially against Gram-positive Staphylococcus aureus (S. aureus). Plate counting experiments confirmed that 75 μg/mL P-CDs could kill 98 % of E. coli when illuminated for 60 min, however, 50 μg/mL P-CDs could inhibit 100 % of S. aureus for the same illumination time. Surely, our metal-free carbon dots nanozyme provides a strong and viable alternative for the management of bacterial contamination in food security and environmental preservation, as well as a theoretical basis for P-doping to promote photocatalytic activities of carbon-dots.

Abstract:The misuse of many types of broad-spectrum antibiotics leads to increased antimicrobial resistance. As a result, the development of a novel antibacterial agent is essential. Photodynamic antimicrobial chemotherapy (PACT) is becoming more popular due to its advantages in eliminating drug-resistant strains and providing broad-spectrum antibacterial resistance. Carbon dots (CDs), zero-dimensional nanomaterials with diameters smaller than 10 nm, offer a green and cost-effective alternative to PACT photosensitizers. This article reviewed the synthesis methods of antibacterial CDs as well as the recent progress of CDs and their nanocomposites in photodynamic sterilization, focusing on maximizing the bactericidal impact of CDs photosensitizers. This review establishes the base for future CDs development in the PACT field.

Abstract:Carbon dots (CDs) are sub-10 nm carbon particles with notable photoluminescence and photoelectrochemical properties, finding diverse applications in optoelectronics, chemistry, and medicine. Their unique physicochemical properties give rise to antimicrobial actions, being realized through complex mechanisms. Discovering the latter was the aim of this review. The primary interaction of CDs with negatively charged bacterial cells is ensured by electrostatic interaction with that because of CDs' surface positive charge. Hydrophobic forces further contribute to this interaction. Modification of CDs with different alkyl chains enhances their antibacterial effect by balancing positive charge and hydrophobicity, facilitating membrane penetration and causing damage to bacterial cells. Another powerful antibacterial mechanism is the ability of photoexcited CDs to generate reactive oxygen species under visible light, effectively destroying critical biomolecules and inducing cell death. Additionally, the photothermal conversion properties of CDs, allowing them to raise local temperatures upon near-infrared light excitation, result in DNA damage and protein denaturation within bacteria, forming the basis for photothermal therapy. Following infiltration of bacterial walls and membranes, CDs can bind to DNA and RNA in bacteria and fungi through noncovalent interactions, inducing structural changes in DNA and affecting RNA. These multifaceted mechanisms underscore the potential of CDs as versatile antibacterial agents with applications across various biomedical fields.
Silver peroxide-incorporated carbon dots with high photothermal performance for combating bacteria

Abstract:The widespread use of antibiotics often increases bacterial resistance. Herein, we reported a silver peroxide-incorporated carbon dots (defined as Ag2O2-CDs) with high photothermal conversion efficiency viain situoxidation process. The prepared Ag2O2-CDs exhibited ultra-small size of 2.0 nm and hybrid phase structure. Meanwhile, the Ag2O2-CDs were of a similar optical performance comparing with traditional carbon dots (CDs). Importantly, the incorporation of Ag2O2into CDs significantly enhanced photothermal conversion efficiency from 3.8% to 28.5%. By combining silver ion toxicity and photothermal ablation, the Ag2O2-CDs were capable of destroying gram-positive and gram-negative bacterium effectively. These findings demonstrated that the Ag2O2-CDs could be served as a potential antibacterial agent for clinical applications.

Abstract:Biocompatible metal-free carbon dots (CDs) with good photo-induced strong oxidation capacity in aqueous solutions are scarce for high-performance photocatalytic antibacterial and tumor therapy. In this work, we achieved effective visible light-induced cell death and antibacterial performance based on biocompatible metal-free CDs. The visible-light-induced reducing ability of the surface electron-withdrawing structure of the CDs allowed for the remaining photo-induced holes with high oxidation capacity to oxidize water molecules and generate hydroxyl radicals. Antibiotic-resistant bacteria were effectively inhibited by the CDs under xenon lamp irradiation with 450 nm long pass filter. Moreover, CD-based tumor photocatalytic therapy in mice was achieved using a xenon lamp with 450 nm long pass filter (0.3 W cm −2 ).

Abstract:Given the ongoing proliferation of bacterial resistance, particularly the rise of “superbugs”, it is imperative to create novel, effective, non-toxic, and non-resistant antibacterial materials and technologies for use in medical settings. Carbon dots derived from dopamine hydrochloride and citric acid (DA-CDs) were effectively produced through a two-step method, displaying photodynamic therapy (PDT) characteristics with low-drug resistance. Only a concentration of 0.125μgmL-1 of DA-CDs can effectively break down established S. aureus biofilms when exposed to blue light (450~500nm), resulting in a biofilm destruction rate of 96.2%. And after 28 passages, the sensitivity of S. aureus and E. coli to DA-CDs when irradiated with blue light stays the same. The ultra-high negative zeta potential of DA-CDs (-35 mV) and excess intracellular reactive oxygen species (ROS) induced by DA-CDs accounted for their antibacterial and antibiofilm effects. Furthermore, DA-CDs exhibit outstanding biocompatibility without causing harm to red blood cells (RBCs), 3T3 Cells, bone marrow mesenchymal stem cells (BMSCs) and human umbilical vein endothelial cells (HUVECs). The DA-CDs from this study can effectively kill harmful bacteria and destroy established biofilms using PDT, providing a new alternative to antibiotics and a valuable tool for exploring new treatments for biofilm-related infections.

Abstract:After splenectomy the immunologic impairment and the susceptibility to infection is increased, but the most important impetus for splenic preservation has been the observation of the overwhelming postsplenectomy sepsis. Successful results with surgical repair of traumatized spleen and several different techniques have been reported. Instead of suture of splenorrhaphy we used in our experiences a new biological adhesive-system, consisting in thrombin, highly concentrated native human fibrinogen and clotting factor XIII. The highly adhesive properties, the excellent tissue compatibility and the haemostyptic effects recommend this system for using in preservation of the injured spleen as well as possible.

Abstract:The diversity of bacteria and their ability to acquire drug resistance lead to many challenges in traditional antibacterial methods. Photothermal therapies that convert light energy into localized physical heat to kill target microorganisms do not induce resistance and provide an alternative for antibacterial treatment. However, many photothermal materials cannot specifically target bacteria, which can lead to thermal damage to normal tissues, thus seriously affecting their biological applications. Here, we designed and synthesized bacteria-affinitive photothermal carbon dots (BAPTCDs) targeting MurD ligase that catalyzes the synthesis of peptidoglycan (PG) in bacteria. BAPTCDs presented specific recognition ability and excellent photothermal properties. BAPTCDs can bind to bacteria very tightly due to their chiral structure and inhibit enzyme activity by competing with D-glutamic acid to bind to MurD ligases, thus inhibiting the synthesis of bacterial walls. It also improves the accuracy of bacteria treatment by laser irradiation. Through the synergy of biochemical and physical effects, the material offers outstanding antibacterial effects and potentially contributes to tackling the spread of antibiotic resistance and facilitation of antibiotic stewardship.

Abstract:Carbon dots (CDs) with an average diameter of 6.3 nm were synthesized from the medicinal seed extract of Syzygium cumini L. using one-pot hydrothermal synthesis. The prepared CDs exhibited excitation-dependent emission characteristics with photoluminescence (PL) emission maxima at an excitation of 340 nm. The CDs at 500 μg/mL displayed antimicrobial activities against four common pathogens. Both Staphylococcus aureus and S. epidermidis were completely eradicated by CDs within 12 h, compared to 24 h for Escherichia coli and Klebsiella pneumonia . The release of various oxygen species (ROS) was postulated to play a critical role in bacterial eradication. The CDs decorated on cotton fabric by ultrasonication also displayed good antibacterial activities against the above bacteria. The finding opens a plausible use of CDs in biomedical textiles with potent antimicrobial properties against both Gram-negative and Gram-positive bacteria.

Abstract:Fluorescent carbon dots (CDs) with oxygen-containing functional groups were prepared by a hydrothermal method. Three kinds of typical oxygen functional groups, i.e. hydroxyl, carbonyl, and carboxylic acid groups, were selectively deactivated to independently assess their photocatalytic activities on the surface of CDs. The presented results indicate that the carbonyl group on CDs is their photocatalytic active sites. Hereby, the high photocatalytic activities of CDs could be produced by enhancing carbonyl group concentration on their surface.

Abstract:Frequent bacterial/fungal infections and occurrence of antibiotic resistance pose increasing threats to the public and thus require the development of new antibacterial/antifungal agents and strategies. Carbon dots (CDs) have been well demonstrated to be promising and potent antimicrobial nanomaterials and serve as potential alternatives to conventional antibiotics. In recent years, great efforts have been made by many researchers to develop new carbon dot-based antimicrobial agents to combat microbial infections. Here, as an update to our previous relevant review (C 2019, 5, 33), we summarize the recent achievements in the utilization of CDs for microbial inactivation. We review four kinds of antimicrobial CDs including nitrogen-doped CDs, metal-containing CDs, antibiotic-conjugated CDs, and photoresponsive CDs in terms of their starting materials, synthetic route, surface functionalization, antimicrobial ability, and the related antimicrobial mechanism if available. In addition, we summarize the emerging applications of CD-related antimicrobial materials in medical and industry fields. Finally, we discuss the existing challenges of antimicrobial CDs and the future research directions that are worth exploring. We believe that this review provides a comprehensive overview of the recent advances in antimicrobial CDs and may inspire the development of new CDs with desirable antimicrobial activities.

Abstract:Using organic pollutants as natural electron transfer mediators and carbon dots (CDs) as photosensitizers for integrated monitoring and purification of contaminants is a new game-changing idea. We herein first reported a novel dual-functional platform of detection and degradation based on the photoinduced electron transfer (PET) process between tetracycline (TC) and CDs. On the one hand, CDs can be used as fluorescence probes for TC detection with good sensitivity and selectivity. On the other hand, they display an excellent photodegradation ability for TC without any external photosensitizer or cocatalyst. Studies suggested that the PET process is beneficial for the transfer of electrons from TC to CDs, which further accelerates electron transfer and suppresses the recombination of electron-hole pairs, thereby improving the fluorescence quenching efficiency and photocatalytic activity. We believe this study can offer referable directions and theoretical guidance for the development of effective environmental pollution control and remediation technologies using CDs.
