Abstract:A Pt prodrug polyphenol and gadolinium ion loaded cancer theranostics nanoplatform based on a mild acidic pH and thermal sensitive polymer was designed for photoacoustic (PA)/magnetic resonance (MR)/positron emission tomography (PET) multimodal imaging-guided chemo-photothermal combination therapy. The Pt drug release can be controlled by tumour-specific acidic pH and heat generated by external NIR irradiation. The nanoparticles were stable under normal physiological environments and released the drug under the acidic pH of tumours and NIR laser irradiation, which can reduce the side effects of the drug to normal organs. Moreover, the MR signal can be significantly enhanced (approximate to 3-fold increase in T-1 relaxivity) under the acidic tumour microenvironment, which is favorable for cancer diagnosis. The nanoparticles exhibited excellent tumour accumulation and led to complete tumour eradication with low power NIR laser irradiation. This promising approach provides a new avenue for imaging-guided combination therapy.
A Nanoparticle-Based Strategy for the Imaging of a Broad Range of Tumours by Nonlinear Amplification of Microenvironment Signals.

Abstract:Stimuli-responsive nanomaterials are increasingly important in a variety of applications such as biosensing, molecular imaging, drug delivery and tissue engineering. For cancer detection, a paramount challenge still exists in the search for methods that can illuminate tumours universally regardless of their genotypes and phenotypes. Here we capitalized on the acidic, angiogenic tumour microenvironment to achieve the detection of tumour tissues in a wide variety of mouse cancer models. This was accomplished using ultra pH-sensitive fluorescent nanoprobes that have tunable, exponential fluorescence activation on encountering subtle, physiologically relevant pH transitions. These nanoprobes were silent in the circulation, and then strongly activated (>300-fold) in response to the neovasculature or to the low extracellular pH in tumours. Thus, we have established non-toxic, fluorescent nanoreporters that can nonlinearly amplify tumour microenvironmental signals, permitting the identification of tumour tissue independently of histological type or driver mutation, and detection of acute treatment responses much more rapidly than conventional imaging approaches.

Abstract:Optical imaging is promising in tumor diagnosis due to its high sensitivity, no radioactive irradiation and low running cost. Compared to small molecular probes, fluorescence nanoprobes demonstrate a tunable circulation lifetime, an up-regulated intratumoral accumulation and enhanced sensitivity by labeling multiple imaging reporters on a single nanoparticle. Acidic extracellular fluid is a universal phenomenon of solid tumors. Therefore, a nanoprobe that responds to the acidic microenvironment is promising in visualizing a tumor in vivo, regardless of the tumor type or even developmental stage. Moreover, the fluorescence "activation" in the tumor but not the normal tissues will greatly increase the "target to background" signal ratio, which benefits the visualization of small volume tumors with high sensitivity. In this review, we summarize the recent developments in pH responsive fluorescence nanoprobes for tumor visualization.

Abstract:Visualizing tumors in their early stages is crucial to increase the survival rate of cancer patients. Optical imaging, as a developing imaging modality, proves to be promising in visualizing tumors non-invasively in vivo due to its high sensitivity, no radioactive irradiation and low running cost. The development of fluorescence probes with high tumor targeting specificity will accelerate the translation of optical imaging from the bench to bedside. Compared to the small molecular imaging probes, the tumor targeting nanoprobes demonstrate the advantages, such as the tunable circulation lifetime, the up-regulated tumor accumulation via the enhanced permeability and retention (EPR) effect, and improved targeting specificity/sensitivity by labeling multiple targeting/imaging domains on a single nanoparticle. Tumor microenvironment (TME) indicators such as acidosis, hypoxia and high interstitial pressure are universal phenomena of solid tumors. Therefore, fluorescent nanoprobes that respond to TME, especially the physiologically acidic pH environment, are promising in visualizing tumor and metastases regardless of the tumor types or even developmental stages. The signal of these pH "responsive" nanoprobes stays "silent" at the normal tissues, but it enhances significantly in the tumor acidic microenvironment. Therefore the signal ratio between the tumor and surrounding normal tissues can increase remarkably, which benefits the visualization of early stage/small volume tumors with high sensitivity. Here, we have summarized the recent advances in the development of fluorescence nanoprobes for tumor visualization by sensing the tumor acidic microenvironment.

Abstract:Nanoparticle-based diagnosis-therapy integrative systems represent an emerging approach to cancer treatment. However, the diagnostic sensitivity, treatment efficacy, and bioavailability of nanoparticles as well as the heterogeneity and drug resistance of tumors pose tremendous challenges for clinical implementation. We herein report on the fabrication of tumor pH-sensitive magnetic nanogrenades (termed PMNs) composed of self-assembled iron oxide nanoparticles and pH-responsive ligands. These PMNs can readily target tumors via surface-charge switching triggered by the acidic tumor microenvironment, and are further disassembled into a highly active state in acidic subcellular compartments that "turns on" MR contrast, fluorescence and photodynamic therapeutic activity. We successfully visualized small tumors implanted in mice via unique pH-responsive T1MR contrast and fluorescence, demonstrating early stage diagnosis of tumors without using any targeting agents. Furthermore, pH-triggered generation of singlet oxygen enabled pH-dependent photodynamic therapy to selectively kill cancer cells. In particular, we demonstrated the superior therapeutic efficacy of PMNs in highly heterogeneous drug-resistant tumors, showing a great potential for clinical applications.

Abstract:A tumor-responsive nanoprobe based on a conjugated polyelectrolyte and gold nanoparticle (AuNP) hybrid was designed to response to the low pH extracellular microenvironment in tumor with light-up fluorescence. AuNPs with positive surface charges were prepared by direct reducing Au salt with sodium borohydride and stabilized by cystamine. A pH triggered charge-reversible polymer and a water-soluble cationic conjugated polyelectrolyte (CPE) were sequentially deposited onto the AuNP surface through electrostatic interaction. The obtained hybrid probe is monodispersed with an average diameter of 68.3 nm by dynamic light scattering measurement. In physiological conditions (pH ≈ 7.4), the hybrid probe is almost non-fluorescent due to the super-quenching of CPE by AuNPs via energy/charge transfer and efficient exciton migration along the polymer backbone. When exposed to acidic extracellular microenvironments in tumor (pH(e) ≈ 6.5), the acid-labile amides hydrolyze into primary amines. The generated amine groups result in strong electrostatic repulsion between CPE and AuNPs, leading to recovered probe fluorescence. The fluorescence turn-on is further utilized for tumor extracellular acidic microenvironment imaging. In addition, under in vivo conditions, the nanosized hybrid probe exhibits specific accumulation in tumor tissue with light-up fluorescence, which provides new opportunities for easy tumor imaging and identification.
A Transistor-Like Ph Nanoprobe for Tumour Detection and Image-Guided Surgery

Abstract:It is challenging to detect a broad range of malignant tumours at high resolution, because of profound genetic and histological differences in cancerous tissue. Here, we report the design and performance of a fluorescent nanoprobe with transistor-like responses (transition pH = 6.9) for the detection of deregulated pH, which drives many of the invasive properties of cancer. The nanoprobe amplifies the fluorescence signal in the tumour over that in the surrounding normal tissues, resulting in a discretized, binary output signal with a spatial resolution smaller than 1 mm. The nanoprobe allowed us to image a broad range of tumours in mouse models using a variety of clinical cameras. We were able to perform real-time tumour-acidosis-guided detection and surgery of occult nodules (<1 mm 3 ) in mice bearing head and neck or breast tumours, significantly lengthening mice survivability. We also show that the pH nanoprobe can be used as a reporter in a fast, quantitative assay to screen for tumour-acidosis inhibitors. The binary delineation of pH achieved by the nanoprobe promises to improve the accuracy of cancer detection, surveillance and therapy.

Abstract:Imaging tumors in their early stages is crucial to increase the surviving rate of cancer patients. Currently most fluorescence probes visualize the neoplasia by targeting the tumor-associated receptor over-expressed on the cancer cell membrane. However, the expression level of these receptors in vivo is hard to predict, which limits their clinical translation. Furthermore, the signal output of these receptor-targeting probes usually stays at a high level, which leads to a strong background signal in normal tissue due to non-specific binding. In contrast to receptors, characteristics of the tumor microenvironment - such as acidosis - are pervasive in almost all solid tumors and can be easily accessed. In this work, a novel biodegradable nanoprobe InNP1 that demonstrates pH-activated near-infrared (NIR) fluorescence in both human glioblastoma U87MG cancer cells in vitro and the subcutaneous U87MG tumor xenografts in vivo is developed. Bio-distribution, in vivo optical imaging, and autoradiography studies demonstrate that the pH-activated NI R fluorescence is the dominant factor responsible for the high tumor/normal tissue (T/N) ratio of InNP1 in vivo. Overall, the work provides a nanoprobe prototype to visualize the solid tumor in vivo with high sensitivity and minimal systemic toxicity by sensing the tumor acidic microenvironment.

Abstract:Acidity-stimulated platforms have the potential to improve the specificity of cancer imaging and therapy within the tumor microenvironment (pH 6.5–6.8). However, avoiding non-specific activation in normal cells (lysosomal or endosomal activation at pH < 5.4) remains a significant challenge. Herein, we developed core-satellite semiconducting polymer nanoparticles @ cobalt hydroxide oxide (SPNs@CoOOH), which is a novel companion diagnostic system tailored to tumor microenvironments. We propose a novel accelerator/brake strategy that regulates their pH-responsive range. This approach allows for efficient and precise treatment of tumors through the adaptation of the optimal pH (pH 6.6) while minimizing non-specific activation within normal tissue and reducing the risk of “off-targeted” toxicity. The specific chemical reaction between 1O2 and thiophene-based SPNs allows for the chemiluminescence from SPNs@CoOOH to be well correlated with the generation of 1O2 under various conditions (e.g., different pH, concentration, or time), thereby showing good relevance with cancer inhibition rates in vivo.

Abstract:Stimulus-responsive fluorescence imaging modality shows great promise for detection of tumor due to the advantages of high sensitivity, simplicity and noninvasiveness. However, some non-cancer regions including nodules and inflammation may also exhibit a stimulus-related characteristic, which cause the problem of nonspecific responsiveness and then cause "false positive" results for tumor recognition. Herein, hypoxia and acidic pH, two typical features strongly associated with tumor invasion, progression and metastasis in tumor microenvironment (TME), are chosen as dual stimuli to fabricate "dual lock-and-key" fluorescent nanoprobe for highly specific and precise imaging of tumor cells. Mesoporous silica coated gold nanorods (AuNR@mSiO2) are employed as nanocarrier and nanoquencher to load the pH-sensitive fluorescent reporter (Rho-TP). Azobenzene (azo) which can be reduced to amines by the highly expressed azoreductase under hypoxic conditions, is elected as the effective gatekeeper for AuNR@mSiO2 by forming complex with β-cyclodextrin polymer via host-guest interaction (azo/β-CDP). By elaborately combining the hypoxia-responsive gatekeeper and pH-responsive fluorescent signal reporter into one nanoprobe, sensitive and specific imaging of tumor cells can be realized. The fabricated dual lock-and-key fluorescent nanoprobe successfully further apply in tumor-bearing mice model, which indicate potential of early diagnosis and assessment of cancer treatment.

Abstract:A pH-responsive light-up nanoparticle probe with aggregation-induced emission (AIE) features was designed and synthesized. The probe carries negative charges and shows very weak fluorescence under physiological conditions. In a tumor acidic extracellular micro-environment, the nanoparticle probe can switch to positive surface charge and thus significantly light up cancer cells, allowing for targeted imaging and selective suppression of cancer cells. As AIE nanoparticles are known for high fluorescence in the aggregate state, this study represents the first example of light-up AIE nanoparticle probe design.

Abstract:Polymer-based fluorescent nanomaterials have proven to universally image various tumors based on their extremely sharp responsiveness to pH change. Such a property has never been realized in supramolecular systems. We herein design a small molecule (DPP-thiophene-4) that is composed of a diketopyrrolopyrrole (DPP) core and two alkyl chains terminated with quaternary ammonium. DPP-thiophene-4 can self-assemble into a nonfluorescent nanoassembly when the pH is >7.0 but reversibly disassembles back to fluorescent monomers when the pH is <6.8. Meanwhile, its fluorescence emission increases by 10-fold within a 0.2 pH unit change. Such a fluorogenic nanoassembly can precisely differentiate a number of malignant tumors among normal tissues in vivo due to the slight acidity within tumor microenvironments. Further the nanoassembly shows satisfactory biocompatibility and an effective clearance from the body. Overall, this supramolecular fluorogenic nanoassembly exhibits an immense potential for realizing broad range tumor diagnosis.
Rational Design of pH-Activated Upconversion Luminescent Nanoprobes for Bioimaging of Tumor Acidic Microenvironment and the Enhancement of Photothermal Therapy

Abstract:The development of effective and safe tumor photothermal therapeutic strategies has attracted considerable attention. Herein, we synthesized tumor microenvironment (TME)-activatable self-assembling organic nanotheranostics (NRhD-PEG-X NPs (X = 1, 2, 3, and 4)) for precise tumor targeting and upconversion image-guided photothermal therapy (PTT). The amphiphilic polymer NRhD-PEG-X consisted of upconversion luminescent probes (NRhD) modified with polyethylene glycol (PEG) of various lengths. The continuous external irradiation-free photothermal NRhD-PEG-4 NPs with pKa 6.70 displayed high sensitivity and selectivity to protons, resulting in the turn-on upconversion luminescence and enhanced photothermal properties in the acidic TME without asynchronous therapy and side effects. This nanotheranostic offers acidic activatability, tumor targetability, and PTT enhancement, thus allowing autofluorescence-free upconversion luminescent imaging-guided precision PTT. Our strategy affords a paradigm to develop activatable theranostic nanoplatforms for precision medicine. Statement of significance As a hyperthermia-based treatment, activatable photothermal therapy (PTT) is highly significant in tumor treatment. Herein, we develop acidic tumor microenvironment-activatable nanotheranostics for upconversion luminescent imaging-guided diagnosis and precision tumor-targeted PTT. PEGylation of upconversion dyes not only could self-assemble to yield organic nanoparticles in water, but it could also significantly improve biocompatibility, stability, and circulation time and tune significantly the pKa value of nanoparticles. In an acidic tumor microenvironment, NRhD-PEG-4 NPs with pKa 6.70 show high sensitivity to release NRhDH + -PEG-4 NPs, which exhibit good upconversion luminescence and enhanced photothermal effect. Therefore, upconversion luminescence imaging-guided precision PTT has high potential to enhance cancer diagnostic and therapeutic efficiency.

Abstract:Molecular imaging probes for biomarker-based diagnosis typically target, with limited sensitivity, a single molecular process or event in a complex biological system. Here, we show that the macromolecular near-infrared poly(ethylene glycol)-conjugated iridium (iii) complex can be designed to successively respond to tumour acidity and hypoxia while amplifying detection sensitivity via signal propagation. We used the probe to detect, by near-infrared imaging, primary tumours and metastatic tumour nodules as small as 1 mm in mice, and to measure the in vivo metabolic rate of cancer cells. We anticipate that probes for imaging coupled biological events with amplified detection sensitivity will offer opportunities for enhanced molecular diagnostics and image-guided biomedical applications.

Abstract:The early detection of cancer shows great promise for the control and prevention of cancer. For early detection, one of the challenges that still exists is searching for methods that can illuminate tumors with high sensitivity. Here, acidity and hypoxia, two typical features that exist universally in a solid tumor microenvironment, were focused on to attain synergistic imaging with an enhanced signal-to-noise ratio. This was realized using an iridium(iii) based optical probe (Ir-1) that could sense acidity and hypoxia simultaneously and synergistically. Through the synergistic sensing of acidic pH and hypoxia, stronger emission signals or larger lifetime changes can be obtained than if a single factor (acidity or hypoxia) is used to induce variations. Furthermore, its potential for biological applications was confirmed by employing Ir-1 for phosphorescence synergistic intensity and lifetime imaging of acidity and hypoxia in live monolayer cells and 3D multicellular spheroids.

Abstract:To address the limitations of traditional photothermal therapy (PTT)/ photodynamic therapy (PDT) and real-time cancer metastasis detection, a pH-responsive nanoplatform (NP) with dual-modality imaging capability was rationally designed. Herein, 1 H,1 H-undecafluorohexylamine (PFC), served as both an oxygen carrier and a 19 F magnetic resonance imaging (MRI) probe, and photosensitizer indocyanine green (ICG) were grafted onto the pH-responsive peptide hexahistidine (H 6 ) to form H 6 -PFC-ICG (HPI). Subsequently, the heat shock protein 90 inhibitor, gambogic acid (GA), was incorporated into hyaluronic acid (HA) modified HPI (HHPI), yielding the ultimate HHPI@GA NPs. Upon self-assembly, HHPI@GA NPs passively accumulated in tumor tissues, facilitating oxygen release and HA-mediated cell uptake. Once phagocytosed by lysosomes, protonation of H 6 was triggered due to the low pH, resulting in the release of GA. With near-infrared laser irradiation, GA-mediated decreased HSP90 expression and PFC-mediated increased ROS generation amplified the PTT/PDT effect of HHPI@GA, leading to excellent in vitro and in vivo anticancer efficacies. Additionally, the fluorescence and 19 F MRI dual-imaging capabilities of HHPI@GA NPs enabled effective real-time primary cancer and lung metastasis monitoring. This work offers a novel approach for enhanced cancer phototherapy, as well as precise cancer diagnosis.

Abstract:Upconversion nanoparticles (UCNPs) emerged as promising near-infrared (NIR) light-triggered nanotransducers for photodynamic therapy (PDT). However, the traditionally used 980 nm excitation source could cause an overheating effect on biological tissues, and the single photosensitizer (PS) loading could not efficiently utilize multiradiation UC luminescence, resulting in a limited efficiency of PDT in tumor tissues with hypoxia characteristics. Herein, 808 nm light-responsive Nd-sensitized 2@MnO2 core–shell NPs were designed as light nanotransducers with efficient UC emission at 550 and 650 nm for PDT and downshifting luminescence at 1525 nm for second NIR (NIR-II) imaging. UC emission was fully utilized by loading dual PSs, rose bengal (RB), and zinc phthalocyanine (ZnPc), thus significantly improving the reactive oxide species (ROS) generation efficiency. Moreover, a manganese dioxide (MnO2) shell with ultrasensitive biodegradability in an acidic tumor microenvironment (TME) can generate an amount of oxygen molecules, alleviating the symptoms of hypoxia and then improving the efficacy of PDT. Meanwhile, the biodegraded Mn2+ ions can further strengthen T1-weighted magnetic resonance imaging (MRI). Electrical and fluorescence in situ monitoring of tumor microenvironment-based pH-responsive polymer dot coated surface

Abstract:A boronate-ester structure forming a pH-responsive polymer dot (Plu-PD) coated biosensor between carbonized-sp2 rich dopamine-alginate [PD(Alg)] and boronic acid-grafted Pluronic (BA-Pluronic) was developed for the electrochemical and fluorescence detection of cancer cells. The reduced fluorescence (FL) resulting from fluorescence resonance energy transfer (FRET) mediated by π-π interactions within Plu-PD was successfully reinvigorated through the specific cleavage of the boronate-ester bond, triggered by the acidic conditions prevailing in the cancer microenvironment. The anomalous variations in extracellular pH levels observed in cancer (pH ∼6.8), as opposed to the normal cellular pH range of approximately 7.4, serve as robust indicators for discerning cancer cells from their healthy counterparts. Moreover, the Plu-PD coated surface demonstrated remarkable adaptability in modulating its surface structure, concurrently exhibiting tunable electroconductivity under reduced pH conditions, thereby imparting selective responsiveness to cancer cells. The pH-modulated conductivity change was validated by a reduction in resistance from 211 ± 9.7 kΩ at pH 7.4 to 73.9 ± 9.4 kΩ and 61.5 ± 11.5 kΩ at pH 6.8 and 6.0, respectively. The controllable electrochemical characteristics were corroborated through in vitro treatment of cancer cells (HeLa, B16F10, and SNU-C2A) via LED experiments and wireless output analysis. In contrast, identical treatments yielded a limited response in normal cell line (CHO-K1). Notably, the Plu-PD coated surface can be seamlessly integrated with a wireless system to facilitate real-time monitoring of the sensing performance in the presence of cancer and normal cells, enabling rapid and accurate cancer diagnosis using a smartphone.
Redox-Activated Light-Up Nanomicelle for Precise Imaging-Guided Cancer Therapy and Real-Time Pharmacokinetic Monitoring

Abstract:Simultaneous tumor imaging, therapy, and pharmacokinetic monitoring can offer a safe and effective strategy for cancer therapy. This work describes the design of a fluorescence light-up nanomicelle that can afford precise imaging-guided drug delivery and pharmacokinetic monitoring in a real-time fashion for cancer chemotherapy. The nanomicelle, which contains a boron dipyrromethene based fluorescent probe as the hydrophobic core and a redox-triggered detachable poly(ethylene glycol) (PEG) shell, can accumulate at the tumor site via enhanced permeation and retention effect. The PEG detachment induced by tumoral and intracellular glutathione can destabilize the nanomicelle, leading to fluorescence light up and simultaneous drug release. Importantly, the fluorescence intensities generated by the nanomicelles in different organs are well-correlated with released drug concentrations in both temporal and spatial manners, suggesting its precise role for imaging-guided drug delivery and pharmacokinetic monitoring in vivo. The tumor growth can be effectively inhibited by the docetaxel-loaded nanomicelle formulation, and the nanomicelles are monitored to be excreted via hepatobiliary routes. This nanomicelle for precise imaging-guided chemotherapy provides a safe and robust theranostic strategy for the evaluation of cancer nanomedicine.
Fluorescent nano- and microparticles for sensing cellular microenvironment: past, present and future applications

Abstract:The tumor microenvironment (TME) features distinct hallmarks, including acidosis, hypoxia, reactive oxygen species (ROS) generation, and altered ion fluxes, which are crucial targets for early cancer biomarker detection, tumor diagnosis, and therapeutic strategies. A variety of imaging and sensing techniques have been developed and employed in both research and clinical settings to visualize and monitor cellular and TME dynamics. Among these, ratiometric fluorescence-based sensors have emerged as powerful analytical tools, providing precise and sensitive insights into the TME and enabling real-time detection and tracking of dynamic changes. In this comprehensive review, we discuss the latest advancements in ratiometric fluorescent probes designed for optical mapping of pH, oxygen, ROS, ions, and biomarkers within the TME. We elucidate their structural designs and sensing mechanisms, as well as their applications in in vitro and in vivo detection. Furthermore, we explore integrated sensing platforms that reveal the spatiotemporal behavior of complex tumor cultures, highlighting the potential of high-resolution imaging techniques combined with computational methods. This review aims to provide a solid foundation for understanding the current state of the art and the future potential of fluorescent nano- and microparticles in the field of cellular microenvironment sensing.
