Abstract:Collagen fibrils hierarchically assemble from microscale to macroscale, which endows the natural composite bone with good mechanical properties and remodeling functions. Revealing the intrafibrillar growth process of hydroxyapatite nanocrystals of collagen will guide the research of bone repair or collagen-based composites. Herein, we investigated the mineralization of multiscale collagen matrices and strongly proved the intrafibrillar hydroxyapatite nanocrystals in the collagen fibrils. The hydroxyapatite nanocrystals were deposited within collagen fibrils with co-orientation along the (002) crystal plane, which is the longitude of the fibril. The whole growth process was captured by TEM to demonstrated the five stages of the intrafibrillar growth process of hydroxyapatite nanocrystals. The infiltration and transformation of amorphous calcium phosphate in isolated collagen fibrils are both demonstrated. The intrafibrillar growth process of hydroxyapatite nanocrystals in collagen film was also investigated, showing that the growth area of collagen films increased linearly with time and the growth process. By studying the in situ mineralization under different reaction conditions, the kinetic equation of the mineralized area of collagen film under each condition was obtained, and the optimal hydroxyapatite mineralized solution was proved to be a solution with polyacrylic acid of 50 μG/mL and a pH of 7.5. Our work provides more detailed information of the growth process of HAP nanocrystals during the mineralization of collagen at different scales and would contribute to future research on the formation process of more minerals in collagen.

Abstract:Collagen is the critical organic component of bone matrix, which is the template for bone biomineralization. Phosphorylation modification of collagen plays an important role in the process of biomineralization in vivo, but its mechanism on in vitro biomimetic mineralization of bone matrix remains unclear at the molecular level. Sodium tripolyphosphate is used to phosphorylate collagen in this study and new phosphate groups appear on collagen fibrils after phosphorylation modification. The chelating amount of calcium is improved linearly with increasing the phosphorylation degree of collagen fibrils, which demonstrates that the introduced phosphate groups serve as new nucleation sites and participate in the formation of apatite minerals inside the collagen fibers. Stabilized nanosized amorphous calcium phosphate by polyacrylic acid can also permeate into collagen fibers and further transform into another layer of hydroxyapatite minerals. Both layers of apatite minerals eventually induce the formation of hierarchical intrafibrillar mineralization structure within the phosphorylated collagen fibers. The present research enriches the previous biomineralization mechanism of bone matrix, provides a facile strategy for biomimetic mineralization of collagen, and offers the basis for future investigation of the advanced bone substitute materials.

Abstract:Contemporary models of intrafibrillar mineralization mechanisms are established using collagen fibrils as templates without considering the contribution from collagen-bound apatite nucleation inhibitors. However, collagen matrices destined for mineralization in vertebrates contain bound matrix proteins for intrafibrillar mineralization. Negatively charged, high-molecular weight polycarboxylic acid is cross-linked to reconstituted collagen to create a model for examining the contribution of collagen-ligand interaction to intrafibrillar mineralization. Cryogenic electron microscopy and molecular dynamics simulation show that, after cross-linking to collagen, the bound polyelectrolyte caches prenucleation cluster singlets into chain-like aggregates along the fibrillar surface to increase the pool of mineralization precursors available for intrafibrillar mineralization. Higher-quality mineralized scaffolds with better biomechanical properties are achieved compared with mineralization of unmodified scaffolds in polyelectrolyte-stabilized mineralization solution. Collagen-ligand interaction provides insights on the genesis of heterogeneously mineralized tissues and the potential causes of ectopic calcification in nonmineralized body tissues.

Abstract:Molecular dynamics (MD) simulations have been used to study calcium phosphate formation regulated by collagen in solution. In this study, a collagen-like peptide molecule has been mixed with different types of ions for simulations of the early stage of biomineralization. We have compared one system containing calcium ions, and hydrogen phosphate ions to the other system containing calcium ions, phosphate ions and hydroxy ions in a water box containing a peptide molecule (1CAG) in the center. After the simulations, the radical distance function and coordination number profiles show that calcium ions and phosphate ions attract each other and form stable clusters in both systems studied. The hydroxy ions are attracted by calcium ions and the distance between them is close to that in hydroxyapatite, which is considered to play important roles in the formation of hydroxyapatite. The current results provide a useful approach to study the scenario of mineralizing collagen using MD methods.

Abstract:Nanoscale replication of the hierarchical organization of minerals in biogenic mineralized tissues is believed to contribute to the better mechanical properties of biomimetic collagen scaffolds. Here, an intrafibrillar nanocarbonated apatite assembly is reported, which has a bone-like hierarchy, and which improves the mechanical and biological properties of the collagen matrix derived from fibril-apatite aggregates. A modified biomimetic approach is used, which based on the combination of poly(acrylic acid) as sequestration and sodium tripolyphosphate as templating matrix-protein analogs. With this modified dual-analog-based biomimetic approach, the hierarchical association between collagen and the mineral phase is discerned at the molecular and nanoscale levels during the process of intrafibrillar collagen mineralization. It is demonstrated by nanomechanical testing, that intrafibrillarly mineralized collagen features a significantly increased Young's modulus of 13.7 +/- 2.6 GPa, compared with pure collagen (2.2 +/- 1.7 GPa) and extrafibrillarly-mineralized collagen (7.1 +/- 1.9 GPa). Furthermore, the hierarchy of the nanocarbonated apatite assembly within the collagen fibril is critical to the collagen matrix's ability to confer key biological properties, specifically cell proliferation, differentiation, focal adhesion, and cytoskeletal arrangement. The availability of the mineralized collagen matrix with improved nanomechanics and cytocompatibility may eventually result in novel biomaterials for bone grafting and tissue-engineering applications.

Abstract:Type I collagen, as the critical organic component of bone matrix, plays a template role in mineralization. Gamma-ray irradiation is used to prepare collagen with different crosslinking degrees without introducing foreign substances for mineralization. Significant improvement in crosslinking degree of collagen can be inferred by enzymolysis and differential scanning calorimetry analysis. Scanning electron microscopy observation reveals that, as crosslinking degree increases, the pore structure of collagen becomes more compact. Fourier transform infrared spectroscopy analysis shows the intact retention of classical triple-helical structure of collagen after crosslinking and the following mineralization. Atomic force microscopy characterization, calcium and phosphorus assay kit tests and thermogravimetry measurements indicate that the more compact the collagen structure, the more hydroxyapatite can attach on collagen fibers. These findings reveal the high crosslinking degree of collagen is favorable for the improvement of mineralization ability by providing a more stable and compact network structure. Such crosslinked collagen may provide a more potential way for the preparation of bone substitute materials.

Abstract:During bone formation, collagen fibrils mineralize with carbonated hydroxyapatite, leading to a hybrid material with excellent properties. Other minerals are also known to nucleate within collagen in vitro. For a series of strontium- and calcium-based minerals, we observed that their precipitation leads to a contraction of collagen fibrils, reaching stresses as large as several megapascals. The magnitude of the stress depends on the type and amount of mineral. Using in-operando synchrotron x-ray scattering, we analyzed the kinetics of mineral deposition. Whereas no contraction occurs when the mineral deposits outside fibrils only, intrafibrillar mineralization generates fibril contraction. This chemomechanical effect occurs with collagen fully immersed in water and generates a mineral-collagen composite with tensile fibers, reminiscent of the principle of reinforced concrete.

Abstract:Introduction: Natural bone has a complex hierarchical nanostructure composed of well-organized collagen fibrils embedded with apatite crystallites. Bone tissue engineering requires scaffolds with structural properties and functionality similar to the natural bone. Inspired by bone, a collagen-apatite (Col-A1) nanocomposite was fabricated with bonelike subfibrillar nanostructures using a modified bottom-up biomimetic approach and has a potential role in the healing of large bone defects in unresolved apical periodontitis. Methods: The bone regeneration potential of the Col-Ap nanocomposite was investigated by comparing it with inorganic beta-tricalcium phosphate and organic pure collagen using a critical-sized rodent mandibular defect model. Micro computed tomographic imaging and histologic staining were used to evaluate new bone formation in vivo. Results: When compared with the beta-tricalcium phosphate and collagen scaffolds, the Col-Ap nanocomposite scaffold exhibited superior regeneration properties characterized by profuse deposition of new bony structures and vascularization at the defect center. Immunohistochemistry showed that the transcription factor osterix and vascular endothelial growth factor receptor 1 were highly expressed in the Col-Ap group. The results indicate that the Col-Ap nano composite activates more bone-forming cells and stimulates, more vascular tissue ingrowth. Furthermore, the Col-Ap nanocomposite induces extracellular matrix secretion and mineralization of rat bone marrow stem cells. The increased expression of transforming growth factor beta 1 may contribute to the formation of a mineralized extracellular matrix. Conclusions: The present study lays the foundation for the development of ColAp nanocomposite based bone grafts for future clinical applications in bone regeneration of large periapical lesions after apical curettage or apicoectomy.

Abstract:The bone and dentin mainly consist of type-I collagen fibrils mineralized by hydroxyapatite (HAP) nanocrystals. In vitro biomimetic models based on self-assembled collagen fibrils have been widely used in studying the mineralization mechanism of type-I collagen. In this chapter, the protocol we used to build a biomimetic model for the mechanistic study of type-I collagen mineralization is described. Type-I collagen extracted from rat tail tendon or horse tendon is self-assembled into fibrils and mineralized by HAP in vitro. The mineralization process is monitored by cryoTEM in combination with two-dimensional (2D) and three-dimensional (3D) stochastic optical reconstruction microscopy (STORM), which enables in situ and high-resolution visualization of the process.
Study of the nucleation sites in collagen mineralization

Abstract:By comparing IR spectra of collagen, collagen/CaCl2 and collagen/calcium phosphate, it was found that intensities of amide I, II and III bands were significantly decreased after mineralization. Band amide I shifted to wards lower wave number. This shift indicated chemical interaction between carboxyl groups and Ca ions formed in tie mineralization. The result indicated that there was another nucleation site, i.e. carbonyl (dropO-O) on collagen, besides the previous reported nucleation site of carboxyl (-OOOH) on collagen. The decrease of amide I peak intensity was mainly due to blockage of C-O stretch. Comparing the IR spectra of demineralized collagen and pure collagen, amide I, II and III peaks intensity and position were almost the same. When the mineral dissolved, amide peaks rebounded, which indicated that the crystals nucleated on these sites enwrapped the groups and blocked the groups vibration. The decreases of peak intensity of amide I, II and III were different due to their different vibration modes. The result confirmed that the carbonyl group was one kind of the nucleation site during collagen mineralization. The relationship between the process of collagen mineralization and variety of amide bonds was also studied.
Investigation on the Microstructural Diversity of a Three-Dimensional Porous Hydroxyapatite/Wollastonite Skeleton via Biomineralization in Simulated Body Fluids

Abstract:The occurrence of fractures has emerged as one of the most prevalent injuries in the human body. In bone reconstruction surgery, after the implantation of porous hydroxyapatite materials, there is an initial infiltration of body fluids into the porous implant, followed by biomineralization-mediated apatite crystal formation and the subsequent ingrowth of bone cells. Despite extensive research efforts in this field, previous investigations have primarily focused on the formation of apatite crystals on exposed surfaces, with limited literature available regarding the formation of apatite crystals within the internal microstructures of bone implants. Herein, we demonstrate the occurrence of dynamic biomineralization within a three-dimensional porous hydroxyapatite/wollastonite (HA/WS) skeleton, leading to the abundant formation of nano-sized apatite crystals across diverse internal environments. Our findings reveal that these apatite nanocrystals demonstrate distinct rates of nucleation, packing densities, and crystal forms in comparison to those formed on the surface. Therefore, the objective of this study was to elucidate the temporal evolution of biomineralization processes by investigating the microstructures of nanocrystals on the internal surfaces of HA/WS three-dimensional porous materials at distinct stages of biomineralization and subsequently explore the biological activity exhibited by HA/WS when combined with cell investigation into apatite crystal biomineralization mechanisms at the nanoscale, aiming to comprehend natural bone formation processes and develop efficacious biomimetic implants for tissue engineering applications. The simultaneous examination of bone cell attachment and its interaction with ongoing internal nanocrystal formation will provide valuable insights for designing optimal scaffolds conducive to bone cell growth, which is imperative in tissue engineering endeavors.
Highly Aligned Hierarchical Intrafibrillar Mineralization of Collagen Induced by Periodic Fluid Shear Stress.

Abstract:Periodic fluid shear stress (FSS) is one of the main mechanical microenvironments in mineralization of bone matrix. To elucidate the mechanism of periodic FSS in collagen mineralization, a mechanical loading induced mineralization system is developed and compared with traditional polyacrylic acid (PAA) induced mineralization. Fourier transform infrared (FTIR) spectroscopy, calcium-to-phosphorus molar ratio and transmission electron microscopy (TEM) demonstrate that both periodic FSS and PAA can control the size of amorphous calcium phosphate (ACP) to avoid aggregation and help the formation of intrafibrillar mineralization. Differently, periodic FSS under a proper cycle and range can accelerate the conversion of ACP to apatite crystals and alleviate the reduced transformation caused by PAA. Under the action of template analogues, periodic FSS can also promote the formation of highly oriented hierarchical intrafibrillar mineralized (HIM) collagen. These findings are helpful for understanding the mechanism of collagen mineralization in natural bone matrix and contribute to the design of novel bone substitute materials with hierarchical structures.
Phosphorylation of collagen fibrils enhances intrafibrillar mineralization and dentin remineralization

Abstract:The hierarchical assembly of nanoapatite within a type I collagen matrix was achieved through biomimetic mineralization in vitro, cooperatively regulated by non-collagenous proteins and small biomolecules. Here, we demonstrated that IP6 could significantly promote intrafibrillar mineralization in two- and three-dimensional collagen models through binding to collagen fibrils via hydrogen bonds (the interaction energy ~10.21 kJ mol-1), as confirmed by the FTIR spectra and isothermal experimental results. In addition, we find that the IP6 associated with dental collagen fibrils can also enhance the remineralization of calcium-depleted dentin and restore its mechanical properties similar to the natural dentin within 4 days. The promoting effect is mainly due to the chemical modification of IP6, which alters the interfacial physicochemical properties of collagen fibrils, strengthening the interaction of calcium phosphate minerals and mineral ions with collagen fibrils. This strategy of interfacial regulation to accelerate the mineralization of collagen fibrils is essential for dental repair and the development of a clinical product for the remineralization of hard tissue.

Abstract:Bones are biocomposites with hierarchical structure that require controlled mineral deposition during their self-assembly to form tissues with unique mechanical properties. Type I collagen proteins, acidic extracellular matrix proteins, play a critical role in mineral formation and many researches on artificial bones have been made inspired by nature using type I collagen derived from animal tissues. Here we report that recombinant human-like type I collagen, an acidic protein, can direct growth of hydroxyapatite (HA) nanocrystals in vitro in the form of self-assembly of nano-fibrils of mineralized collagen resembling extracellular matrix. The mineralized collagen fibrils aligned parallel to each other to form mineralized collagen fibers. HA nanocrystals grew on the surface of these collagen fibrils with the c-axis of nanocrystals of HA orienting along the longitudinal axis of the fibrils. These artificial analogs of bone have a potential clinical application in bone repair.

Abstract:Mineralization of fibrillar collagen with biomimetic process-directing agents has enabled scientists to gain insight into the potential mechanisms involved in intrafibrillar mineralization. Here, by using polycation- and polyanion-directed intrafibrillar mineralization, we challenge the popular paradigm that electrostatic attraction is solely responsible for polyelectrolyte-directed intrafibrillar mineralization. As there is no difference when a polycationic or a polyanionic electrolyte is used to direct collagen mineralization, we argue that additional types of long-range non-electrostatic interaction are responsible for intrafibrillar mineralization. Molecular dynamics simulations of collagen structures in the presence of extrafibrillar polyelectrolytes show that the outward movement of ions and intrafibrillar water through the collagen surface occurs irrespective of the charges of polyelectrolytes, resulting in the experimentally verifiable contraction of the collagen structures. The need to balance electroneutrality and osmotic equilibrium simultaneously to establish Gibbs–Donnan equilibrium in a polyelectrolyte-directed mineralization system establishes a new model for collagen intrafibrillar mineralization that supplements existing collagen mineralization mechanisms. A new model for collagen intrafibrillar mineralization shows the need for a balance between osmotic equilibrium and electroneutrality to establish Gibbs–Donnan equilibrium.

Abstract:Biomineralization is an important process of bone tissue generation. Calcium (Ca) and phosphate (P) ions aggregate and nucleate under the regulation of biomolecules at the initial mineralization stage. Due to the complexity of the physiological environment, the movement behavior and mineralization mechanism of calcium and phosphate ions, as well as the effect of biomolecules on them, are not clear. In this study, computer simulations and experimental verification were applied to investigate the characteristics of the initial biomineralization from the view of ion aggregation and nucleation. The results prove that P ions play a more important role in mineralization than Ca ions. The guanidyl group and surrounding carboxyl terminal groups are a potential excellent nucleation domain on proteins. The interval distribution of acidic/basic residues on protein is more conductive to the formation of large Ca and P ions clusters. The involvement of protein could increase the probability of hydroxyapatite phase precipitation, especially in the presence of a helical conformation. Abstract:Herein, the hierarchical assembly of nanoapatite within a type I collagen matrix was achieved through biomimetic mineralization in vitro using phosphorylated chitosan (Pchi) to promote mineralization. The mineralization kinetics, nanostructure of the collagen matrix, and mineral content were evaluated by Fourier-transform infrared spectroscopy, transmission electron microscopy, and scanning electron microscopy. In addition, differential thermal analysis was used to quantify the extent of mineralization. The results showed that mineralized collagen fibrils were well encapsulated by newly formed nanoapatite mineral nanocrystals. In addition, the use of Pchi was found to significantly shorten the self-assembly process, achieving remineralization of calcium-depleted dentin within 96 h. Chemical modification of collagen fibers with Pchi accelerated the rate of crystallization due to the excellent ion chelating properties of chitosan derivatives and their ability to cause a high degree of conglutination. The modification process created new nucleation sites on the fibers onto which amorphous calcium phosphate could attach within the collagen I matrix. This novel biomimetic mineralization technique provides an efficient method to produce an advanced mineralized matrix and may present an important strategy for future bone tissue engineering applications.
Visualization of Collagen-Mineral Arrangement Using Atom Probe Tomography

Abstract:Bone is a functional material comprised of mainly two phases: an organic collagenous phase and an inorganic mineral phase. Collagen-mineral arrangement has implications for bone function, aging, and disease. However, theories on collagen-mineral arrangement have been confined to studies with low spatial and/or compositional resolution resulting in an extensive debate over the location of mineral with respect to collagen. Herein, a strategy is developed to extract a single mineralized collagen fibril from bone and analyze its composition and structure atom-by-atom with 3D sub-nanometer accuracy and compositional clarity using atom probe tomography (APT). It is shown for the first time a method to probe fibril-level mineralization and collagen-mineral arrangement from an in vivo system with both the spatial and compositional precision required to comment on nanoscale collagen-mineral arrangement. APT of leporine bone shows distinct and helical collagen fibrils with mineralized deposits both encapsulating and incorporated into the collagenous structures. This study demonstrates a novel fibril-level detection method that can be used to probe the composition of bone and contribute new insights to the structure and organization of mineralized materials such as bones and teeth.
