Abstract:Sulfur-containing compounds are commonly present in both natural and synthetic bioactive molecules. The ability to construct C-S bonds using inexpensive and easily accessible raw materials under mild conditions is an area of significant interest in organic synthesis. In this study, we introduce a novel photo-induced iron-catalyzed strategy that facilitates the transformation of alkyl and heteroaryl carboxylic acids into their respective thioether products. Furthermore, our method also enables the site-selective synthesis of thioethers and sulfoxides from unreactive hydrocarbons under a reaction atmosphere of N-2 or air. This procedure is operationally simple and readily scalable and provides access to high-value sulfur-containing compounds from simple raw materials in one step.

Abstract:A fragmentation-alkynylation, -alkenylation and -alkylation cascade via photoinduced iron-catalyzed ligand-to-metal charge transfer (LMCT) is described. This reaction enables the preparation of a series of valuable internal alkynes and olefins, demonstrating a new synthetic pathway for the activation of iminyl radicals from oxime acids. This method exhibits a lot of advantages, including mild conditions, simple operation, and the use of cheap and easy-to-obtain iron catalysts. An iron-catalyzed fragmentation alkynylation, alkenylation and alkylation enabled by photoinduced ligand-to-metal charge transfer (LMCT)processis herein reported.

Abstract:We report an alkoxy radical process for the C-C bond cleavage and functionalization of unstrained tertiary and secondary cyclic alcohols. In the absence of a chlorine atom, the readily available iron catalysts [Fe(OBu-t)3 or Fe(acac)3/t-BuONa] facilitate alkoxy radical formation via the direct ligand-to-metal charge transfer of Fe alkoxide and further enable the ring opening and amination of cyclic alcohols. The remote amino carbonyl compounds could be obtained with a broad scope in up to excellent yields under the mildly redox-neutral system. Light-driven electron transfer, alkoxy radical formation, and subsequent C-C bond cleavage via β-scission were the keys to the transformation.

Abstract:We report here a photoinduced iron-catalyzed fluoroalkylation-peroxidation of activated and/or unactivated alkenes with fluoroalkyl carboxylic acids and hydroperoxide. The ligand-to-iron charge transfer strategy effectively overcomes the high redox potential of the fluoroalkyl carboxylic acids, facilitating the difunctionalization reaction to occur smoothly under mild reaction conditions. The late-stage functionalization of drug and natural product derivatives was also demonstrated.

Abstract:Catalytic C(sp3)-H functionalization has provided enormous opportunities to construct organic molecules, facilitating the derivatization of complex pharmaceutical compounds. Within this framework, direct hydrogen atom transfer (HAT) photocatalysis becomes an appealing approach to this goal. However, the viable substrates utilized in these protocols are limited, and the site selectivity shows preference to activated and thermodynamically favored C(sp3)-H bonds. Herein, we describe the development of undirected iron-catalyzed C(sp3)-H borylation, thiolation, and sulfinylation reactions enabled by the photoinduced ligand-to-metal charge transfer (LMCT) process. These reactions exhibit remarkably broad substrate scope (>150 examples in total), and most importantly, all of these three reactions show unconventional regioselectivity, with the occurrence of C(sp3)-H borylation, thiolation, and sulfinylation preferentially at the distal methyl position. The procedures are operationally simple and readily scalable and provide access to high-value products from simple hydrocarbons in one step. Mechanistic studies and control experiments indicate that the afforded site selectivity is not only relevant to the HAT species but also largely affected by the use of boron- and sulfone-based radical acceptors.

Abstract:Directly utilizing abundant and inexpensive sources of aliphatic carboxylic acids is highly attractive for the synthesis of CF2H- and CH3-containing compounds. Herein, we report a versatile photoinduced iron-catalyzed platform for hydrodifluoromethylation and hydromethylation of alkenes without the need of additional stoichiometric oxidants. These two transformations are accomplished by a visible-light-induced ligand-to-iron charge transfer open-shell activation system by using industrial chemical difluoroacetic and acetic acids as precursors and postulated to operate via a difluoromethyl and methyl radical-mediated decarboxylation mechanism. Experimental results show that the strategy is also applicable to the difunctionalization of unactivated alkenes. Meanwhile, this strategy provides a viable route for the synthesis of pharmaceutically active molecules containing deuterated methyl units. The protocol exhibits several features, including mild reaction conditions, divergent radical precursors, operational simplicity and scalability, good chemo- and regioselectivity, scalability supported by continuous flow technology, and the ability to provide high-value building blocks for applications in one step.

Through adjusting the structure of alkoxyl radical precursors, functionalized lactones and keto-amides were synthesized with good yields and excellent functional group tolerance under redox-neutral conditions. Notably, this protocol enables the integration of lactone fragments with many amine drugs and drug fragments.

Abstract:Light-induced ligand-to-metal charge transfer (LMCT) has been utilized as a powerful strategy in various organic reactions. First-row transition metals, especially iron complexes, show good applications in this process. Fe(III)-Cl and Fe(III)-OR species are two key intermediates involved in the LMCT of iron complexes. This review highlights studies on LMCT of Fe(III)-OR species, including carboxylate-iron and alkoxy-iron species, in organic transformations. Reaction conditions, substrate scope and related mechanisms are discussed.

Abstract:Organophosphine compounds are of high significance in organic synthesis, organometallic chemistry, and materials science. The high demand for the incorporation of phosphine moieties into molecular structures has witnessed significant progress. However, synthetic methods of C(sp(3))-H phosphorylation on unactivated hydrocarbons still remain scarce. Herein, we describe the development of an iron-catalyzed C(sp(3))-H phosphorylation reaction enabled by a photoinduced ligand-to-metal charge transfer (LMCT) process. The reaction exhibits remarkably broad substrate scope (>66 examples), including various alkanes, halides, ketones, esters, nitriles, ethers, thioethers, and silanes as viable substrates. Notably, unconventional site selectivity of C-H phosphorylation is achieved, with the occurrence of phosphorylation preferentially at sterically unhindered C-H bonds. The procedure is operationally simple and readily scalable under continuous-flow, and provides access to high-value organophosphines from simple hydrocarbons in one step. Preliminary studies on the reaction mechanism and site selectivity have also been carried out.

Abstract:A base‐and‐additive‐free photo‐induced radical‐mediated cascade cyclization reaction of 1,6‐enyne compounds was presented. In this study, C(sp3)−H hydrocarbons, including various alkanes and ethers, were activated via Fe‐catalyzed photo‐induced conditions, providing a bunch of γ‐lactam products via hydrogen atom transfer and radical addition processes. Control experiments indicated that the reaction proceeded through photo‐induced ligand‐to‐metal charge transfer (LMCT) process. Gram scale experiment was also completed by using continuous‐flow‐photocatalytic‐device.

Abstract:We report a photoinduced iron/copper dual-catalytic strategy for the radical decarboxylation functionalization of aliphatic carboxylic acids. The photoinduced ligand-to-iron charge transfer process under light was initially occurred to generate an unstabilized alkyl radical, and the copper catalyst delivered the radical and enabled the subsequent coupling reactions to form C–N or C–C bonds. By merging iron-catalyzed decarboxylation with copper catalysis, this system allows the smooth conversion of a wide range of aliphatic carboxylic acids for the amination, decarboxylative dehydrogenation, and alkylation efficiently. A wide variety of complex drug or natural molecules is applicable, suggesting that this strategy would facilitate rapid compound library synthesis and benefit to the discovery of pharmaceutical agents.

Abstract:A general catalytic protocol for the halodecarboxylation of aliphatic carboxylic acids via ligand-to-metal-charge-transfer (LMCT) is presented. This conversion is capable of transforming primary, secondary, and tertiary alkyl acids into corresponding bromo-, iodo- and chloroalkanes using water as a solvent under an air atmosphere. The broad range of substrate scope, the use of a green solvent, and the ability to derivatize drug molecules and natural products offer a new prospect for decarboxylative halogenation reactions.

Abstract:Photoinduced ligand-to-metal charge transfer (LMCT) has emerged as an effective strategy for synthesizing organic molecules in a sustainable manner. However, the majority of existing reports on selective C(sp 3 )–H bond functionalization via photoinduced LMCT focus on the use of late transition metals or rare-earth metals for radical additions or cross-couplings. In contrast, the utilization of photoinduced LMCT with 3d early transition metals poses a significant challenge. Herein, we describe an unprecedented approach to allylic C(sp 3 )–H addition to aldehydes, employing chromium (Cr) complexes as catalysts through visible-light-induced LMCT. By investigating the reaction pathway through various mechanistic studies, including radical trapping, kinetic isotope effect (KIE) analysis, and transient absorption spectroscopy, valuable insights have been gained. The proposed mechanism suggests the intermediacy of bromine radicals through homolysis of the Cr–Br bond. Notably, this protocol expands our understanding of the photochemical properties of earth-abundant Cr complexes.
