CircGSAP Alleviates the Dysfunction of Pulmonary Microvascular Endothelial Cells via BMPR2 Signalling Pathway

Abstract:Background: We recently found that circular RNA- gamma -secretase-activating protein (circGSAP) down-regulation in peripheral blood mononuclear cells was associated with the occurrence and outcomes of idiopathic pulmonary arterial hypertension (IPAH). We aimed to demonstrate that the underlying mechanisms of circGSAP regulate pulmonary microvascular endothelial cells (PMECs) dysfunction by absorbing microRNA to inhibit target genes in IPAH.Methods: Quantitative real-time polymerase chain reaction, immunofluorescence staining, Cell Counting Kit-8, Calcein-AM/PI staining and Transwell and dual-luciferase reporter assays were used to clarify the roles of circGSAP.Findings: CircGSAP levels were decreased in the plasma of patients with IPAH and were associated with poor outcomes. In monocrotaline-induced PH rats, circGSAP overexpression decreased mortality, and improved pulmonary vascular remodelling and right ventricular remodelling. CircGSAP overexpression inhibited hypoxia-induced PMECs proliferation, migration and mortality by absorbing miR-27a-3p. BMPR2 was identified as a miR-27a-3p target gene. BMPR2 levels were up-regulated in PMECs treated with circGSAP plasmid and down-regulated in hypoxia-treated PMECs. The effects of small interfering RNA (siRNA) circGSAP on PMECs proliferation, migration and mortality were mitigated via a miR-27a-3p inhibitor.
