Three-Dimensional Organ Size Control by Ptp61F

Abstract:Precise organ size control is fundamental for all metazoans, but it remains largely unexplored at a molecular level how organ size is controlled in a three-dimensional way. Here, through a RNAi screen, we identify Drosophila Ptp61F as a pivotal regulator of organ size that integrates Hippo pathway, TOR (target of rapamycin) pathway, and actomyosin machinery. In pathological condition, loss of Ptp61F synergizes with RasV12 to induce Hippo and TOR dependent tumorigenesis. Physiologically, depletion of Ptp61F increases body size, drives neoplastic tumor formation and intestinal stem cell proliferation and renewal. We further show that Ptp61F regulates cell contractility and myosin activation and controls three-dimensional cell shape by reducing cell height and horizontal cell size. Mechanistically, Ptp61F regulates Hippo, Tor signaling and cell contractility via forming a complex with Expanded (Ex). Additionally, Ptp61F increases endosomal localization of Ex and Yki, and Ptp61F/Ex complex interacts with Ap-2α. Strikingly, we demonstrate that PTPN2, the conserved human ortholog of Ptp61F, can functionally substitute for Ptp61F in Drosophila. Our work uncovers Ptp61F as an essential determinant that controls three-dimensional organ size under both physiological and pathological conditions.
