Methane Production, Substrates Transformation and Microbial Community in High-Solid Anaerobic Digestion: Insight into C/N Ratio and Biodegradability

Abstract:In order to investigate the properties of methanogenesis with different types of substrates during high solid anaerobic digestion (HS-AD), experiments aimed at C/N ratio and biodegradability were carried out. The refractory (wheat straw and white meat) and easily degrade (wheat flour and peptone) substrates with the C/N ratio of 82.72~2.82 were digested, respectively. Methane production and intermediate metabolites were monitored to evaluate the variation of substrates during HS-AD, mathematical model and microbial community analysis were carried out to explore the mechanism of methane production performance. Results showed that under high C/N ratio (>50:1), the difference between C/N ratio and biodegradability have little influence on the methane yield (MY). With the C/N ratio increasing from 2.82 to 3.38, the MY increased by 2.11 times. The compositional advantages of refractory substance make it more superior in HS-AD. Mathematic fitting results confirming that the high C/N ratio easily degrade substrate and low C/N ratio refractory substance had higher methane production rate. In addition, high C/N ratio substrates had the highest proportion of propionic acid in total VFAs during HS-AD, which reduces the methanogenic performance of the system. The archaea and bacteria in system of low C/N ratio refractory substrate had established a direct interspecies electron transfer (DIET) pathway, which improve the methane production efficiency. The results of the study have brought a new insight of the impact of the C/N ratio and biodegradability of the substrates on HS-AD.
