Acidic Tumor Microenvironment Sensitive GAD1 Inhibits Gliobalstomas Migration and Invasion by Β-Catenin Translocation

Abstract:Background: Glutamate decarboxylase 1 (GAD1) is found in the GABAergic neurons of the central nervous system. Little is known about the relevance of GAD1 in gliomas. The aim of this study is to explore the expression status of GAD1 and its functional mechanisms in gliomas. Methods: We evaluated the GAD1 mRNA and protein expressions according to the level of The Cancer Genome Atlas (TCGA), as well as the clinical samples by immunoblotting analyses and the immunohistochemistry. The critical functions of acidic tumor microenvironment was analyzed by the immunoblotting analyses. To assess the critical functions of GAD1 in cellular invasiveness and migration, the U87 and U251 cells were treated with the shRNA. In addition, The potential mechanisms between GAD1 and β-catenin translocation was examined by immunoblotting analyses. Results: The GAD1 expression was inversely correlated with malignancy grade at both transcriptional and protein levels based on the data from The Cancer Genome Atlas (TCGA), which was consistent with the clinical sample. The acidic tumor microenvironment reduced the expression of GAD1 and increased the invasion and migration ability of U87 and U251 glioma cells, which may be achieved by promoting the phosphorylation of Src TYR 527. The functional analysis revealed that downregulation of GAD1 promotesd the invasion and migration ability in glioma cells, and the promotion of nuclear localization of β-catenin was also observed in GAD1 knockdown. Conclusions: Our data shows that the acidic tumor microenvironment inhibits the expression of GAD1 and promotes migration and invasion in glioma cells. GAD1 is correlated with cellular invasiveness and migration by regulating β-catenin translocation. GAD1 might work as a potential therapeutic target for treatment.
