Chlorpyrifos Induces Mitochondrial Apoptosis in Porcine Trophectoderm Cells Via Mapk Signaling Pathway

Abstract:Chlorpyrifos (CPF) is an organophosphorus pesticide that is widely used in agricultural production and residential environments worldwide. In this study, we investigated the harmful effects and toxicological mechanism of CPF on porcine trophectoderm (pTr) cells. The findings revealed that CPF significantly decreased pTr cell viability and increased intracellular lactate dehydrogenase (LDH) release. Moreover, after CPF treatment, pTr cells showed significant apoptosis by flow cytometry and western bolt analysis. The potential of the mitochondrial membrane was also measured. The mitochondrial membrane potential of pTr cells treated with CPF was markedly depolarized compared to the control group, according to JC-1 staining and flow cytometry analyses. Furthermore, CPF can also induce the increase of ROS and decrease the tight junction (TJ) protein expression in pTr cells. Finally, we further verified that CPF-induced mitochondrial apoptosis in pTr cells is mediated via MAPK signaling pathway by measuring the changes of MAPK signaling pathway proteins and adding inhibitors U0126, SP600125 and SB203580. These results will provide theoretical basis and potential treatment for reproductive toxicity induced by CPF.
