An Integrated Analysis of Metagenomics and Targeted and Untargeted Metabolomics of Skeletal Muscle Mass Adjusted for BMI among Chinese Peri-/Post-Menopausal Women

Abstract:Emerging evidence has suggested the presence of a gut-muscle axis, by which the gut microbiota can produce compounds which enter the systemic circulation and ultimately affect muscle. Here we performed metagenomic sequencing, and targeted and untargeted metabolomics profiling in 498 Chinese peri-/post-menopausal women to examine the relationships among the gut microbiota, serum metabolites, and muscle mass. We identified eight bacteria species, two host metabolites, and two SCFAs that were significantly associated with muscle mass. Among them, Oscillibacter sp.KLE-1728, Ruminococcus albus, and acetic acid were positively correlated with muscle mass, while palmitic acid was negatively correlated with muscle mass. Ruminococcus is reported to produce acetic acid, which increases muscle mass. We further found that Oscillibacter sp.KLE-1728 was negatively correlated with palmitoleic acid, both of which are involved in fatty acid biosynthesis metabolic pathways. Additionally, phenylacetylglutamine (PAGln) was positively correlated with Oscillibacter sp.KLE-1728 and Ruminococcus albus, all of which are involved in phenylalanine metabolism.
