Single-Cell RNA Sequencing Reveals PD-L1+ Fibroblasts As a Determinant of Immune Exhaustion in Pancreatic Cancer Microenvironment

Abstract:Compelling evidence has suggested the pivotal role of stroma cells in carcinogenesis and metastasis in pancreatic cancer. Interrogation on the cell diversity is essential to delineate the tumor microenvironment (TME) and the cross-talks between stroma cells and tumor. In the current study, we provide a comprehensive single-cell map of the pancreatic cancer to date. Interestingly, cancer-associated fibroblasts (CAFs) display previously unidentified heterogeneity containing five subpopulations, potentially leading to varied degrees of tissue fibrosis. Strikingly, we define a new population of CAFs named “regulatory CAFs” (regCAFs) considering its functions of suppressing immune response and promoting T cell exhaustion. Our analyses highlight the connection between infiltrated T cells and CAFs by different chemokine and chemokine receptor pairs and provide novel insights into the cellular crosstalk in pancreatic TME at single-cell resolution. This study will further our understanding of immune landscape in pancreatic cancer with implications for therapeutic development.
