Cardiotonic Pills®Post‐treatment Ameliorates Ischemia‐Reperfusion Induced Myocardial Microcirculatory Disturbances and Myocardial Lesions

Abstract:Cardiotonic Pills® (CP) is a compound preparation of Chinese medicine widely used for treatment of cardiovascular diseases in China. We investigated the ameliorating effects of CP post‐treatment on ischemia‐reperfusion (I/R) induced myocardial microcirculatory disturbances and myocardial lesions. Anesthetized SD rats were subjected to 30 min of left anterior descending coronary artery occlusion and followed by reperfusion for 6 days. In some animals, CP (0.1 g/kg, 0.4 g/kg or 0.8 g/kg) was administrated once a day starting from 3 hrs of reperfusion until day 6. CP post‐treatment significantly attenuated I/R‐induced myocardial microcirculatory disturbance, including the decrease in red blood cell velocity and coronary blood flow, and the increase in albumin leakage. In addition, noticeable changes were observed at day 6 after reperfusion in myocardial infarct size, myocardial fibrosis size, myocardial ultrastructure, the number of myeloperoxidase‐ positive cells, and the expression of transforming growth factor β1 in myocardial tissue, which were significantly ameliorated by post‐treatment with CP. These results suggested that post‐treatment with CP ameliorates the myocardial microcirculatory disturbances, myocardial infarct, and myocardial fibrosis induced by I/R.
