Nevertheless, the detailed roles of lncRNA SUCLG2-AS1 was rarely known and remained to be elucidated. METHODS: The differential expressed lncRNAs were identified by TCGA and GEO database. The expression level of SUCLG2-AS1 was measured by Real-Time PCR (RT-PCR) in 59 patients with clear cell renal cell carcinoma (ccRCC) and different RCC cell lines. EdU assay, CCK-8 assay and transwell assay were performed to investigate the biological effects of SUCLG2-AS1 on RCC cells proliferation, migration and invasion. RNA-seq analysis, RNA-FISH and western blot were performed to explore the molecular mechanisms underlying the functions of SUCLG2-AS1. RESULTS: In present study, we found that SUCLG2-AS1 was down-regulated in ccRCC tissues through analyzing TCGA database. Moreover, decreased SUCLG2-AS1 expression level was correlated to higher tumor stage, advanced histological grade and poorer prognosis. Low expression of SUCLG2-AS1 was also confirmed in ccRCC clinic samples and RCC cell lines. In vitro, overexpression of SUCLG2-AS1 inhibited cell proliferation, migration and invasion in different RCC cell lines. In vivo, we confirmed that SUCLG2-AS1 overexpression could significantly inhibit the growth of RCC. Mechanistically, we unveiled that SUCLG2-AS1 was mainly localized in the cytoplasm. Furthermore, overexpression of SUCLG2-AS1 suppressed the expression of MAPK pathway related proteins in RCC. CONCLUSIONS: In conclusion, our findings demonstrated that SUCLG2-AS1 functions as a tumor suppressor gene, which may provide a candidate prognostic biomarker and treatment target for patients with ccRCC.
