Abstract:Pesticide residues have raised considerable concern about environmental health and food safety. Despite a great advance in enzymatic sensors for pesticide detection, the intrinsic fragility of native enzyme and possible fake results due to single mode signal have hindered its wide application. Here, a novel dual-mode sensor is reported for organophosphorus pesticide detection by using metal-organic framework (MOF) nanozyme NH2-CuBDC as sensing element. The intrinsic peroxidase-mimicking activity and fluorescence property of NH2-CuBDC enable both colorimetric and fluorescent detection of chlorpyrifos. Compared with previously reported chlorpyrifos sensors, our sensor exhibits outstanding sensitivity, and the limits of detection (LOD, S/N = 3) in colorimetric and fluorescent modes are 1.57 ng/mL and 2.33 ng/mL, respectively. No obvious interferences from other substances were measured and chlorpyrifos analysis in real samples presented good reliability, showing practical potential. This work is anticipated to provide new insights to develop multifunctional nanozymes and integrated multi-mode sensing platforms.

Abstract:The great threat of pesticide residues to the environment and human health has drawn widespread interest to explore approaches for pesticide monitoring. Compared to commonly developed single-signal pesticide assays, multi-mode detection with inherent self-validation and self-correction is expected to offer more reliable and anti-interference results. However, how to realize multi-mode analysis of pesticides still remains challenging. Herein, we propose a dual-mode fluorescence and colorimetric method for pesticide determination by integrating stimulus-responsive luminescence with oxidase-mimetic activity into cerium-based coordination polymer nanoparticles (CPNs(Ⅳ)). The CPNs(Ⅳ) exhibit good oxidase-like activity of catalyzing the colorless 3,3′,5,5′-tetramethylbenzidine (TMB) oxidation to its blue oxide, offering a visible color signal; by employing acid phosphatase (ACP) to hydrolyze ascorbic acid 2-phosphate (AAP), the generated ascorbic acid (AA) can chemically reduce the CPNs(Ⅳ) to CPNs(Ⅲ), which exhibit a remarkable fluorescence signal but lose the oxidase-mimicking ability to trigger the TMB chromogenic reaction; when pesticides exist, the enzymatic activity of ACP is restrained and the hydrolysis of AAP to AA is blocked, leading to the recovery of the catalytic TMB chromogenic reaction but the suppression of the fluorescence signal of CPNs(Ⅲ). According to this principle, by taking malathion as a pesticide model, dual-mode 'off-on-off' fluorescence and 'on-off-on' colorimetric detection of the pesticide with good sensitivity was realized. Excellent interference-tolerance and reliability were verified by applying it to analyze the target in real sample matrices. With good performance and practicability, the proposed dual-mode approach shows great potential in the facile and reliable monitoring of pesticide residues.

Abstract:Semiconductor nanocrystals (NCs) possess unique photoluminescent properties which can be used to design fluorescence probes for chemo/biosensing applications. Several have recently emerged that offer excellent turn-on or ratiometric fluorescence chemosensory protocols by sophisticated procedures, but it has been challenging to realize all of these advantages in a single construct. Herein, we develop an intrinsic dual-emitting Mn-doped ZnS nanocrystal-based probe that achieves this goal with turn-on and ratiometric fluorescence response for the determination of organophosphate (diethylphosphorothioate, DEP). The probe relies on the modification of dopamine dithiocarbamate on the surface of NCs and the modulation of dual emission through a photoinduced electron transfer process, which makes use of red fluorescence of Mn(2+) ions doped in the NCs as specific recognition for the target analyte and blue defect emission of the NCs as stable internal reference. In presence of DEP, the red emission of the probe is thus enhanced by switching off the electron transfer pathway, while the blue emission is almost unchanged. With the addition of different amounts DEP, the two emission intensity ratios gradually vary and display color changes from dark-blue to purple to red. Thus, this method generates turn-on and ratiometric fluorescence signals for quantitative and visual detection of the analyte. Significantly, the dual-emitting probe has been used to fabricate paper-based test strips for visual detection of DEP residues, which validate the method for its rapid, on-site, and visual identification.

Abstract:In this work, a convenient ratiometric fluorescent platform was designed to measure organophosphorus pesticides (OPs) based on acetylcholinesterase (AChE), acetylthiocholine (ATCh), manganese dioxide nanosheets (MnO 2 ), near-infrared carbon dots (RCDs) and o-phenylenediamine (OPD). In this platform, a direct oxidation of OPD by MnO 2 generated the luminescent product 2,3-diaminophenolazine (DAP) through intrinsic oxidase activity, while RCDs served as a fluorescent reference indicator. In the presence of AChE and ATCh, the enzymatic hydrolysate thiocholine (TCh) would reduce MnO 2 nanosheets to Mn 2+ , leading to the quenching of DAP fluorescence. On the other hand, OPs can inhibit the catabolism of ATCh by AChE thus acting as a recognizer of OPs. According to these reactions, OPs were quantitatively analyzed by the intensity ratio of fluorescence emitted from RCDs and DAP ( F 560 / F 676 ). The constructed platform can detect OPs with the range of 0.2–0.6 μM with a detection limit of 4.3 nM.

Abstract:Organophosphorus pesticide residues pose significant risks to human health, but conventional analytical methods are costly and complex. To address this, we developed a new environmentally friendly technique using a simple hydrothermal method to create a ratiometric fluorescent composite (RFC) with two distinct signals: blue and orange. This RFC allowed for rapid detection and quantitative analysis of organophosphorus pesticides. The composite incorporated a cerium-based metal–organic framework (Ce-UiO-66), which could degrade methyl paraoxon (MP) due to the two oxidation states of cerium. The resulting degradation product, p-nitrophenol (p-NP), specifically quenched the blue fluorescence of the RFC via the inner filter effect (IFE), while the orange fluorescence remained unaffected, ensuring consistency under microenvironmental and spectrometer conditions. This innovative approach was sensitive, with a detection limit of 0.55 μM, selective, and repeatable, offering a new strategy for pesticide analysis and safety evaluation of edible and medicinal plant and water samples.

Abstract:In this work, a fluorescence and phosphorescence dual-signal readout platform for multi-channel detections of carbaryl, thiram and chlorpyrifos was constructed by confining carbon dots (CDs) in SiO2 nanoparticles. The CDs were synthesized by a simple hydrothermal treatment of fresh pomegranate seeds, L-aspartic acid and L-tryptophan (PAT-CDs). PAT-CDs embedded in SiO2 nanoparticles (PAT-CDs@SiO2) emitted blue fluorescence and very long lifetime green phosphorescence in aqueous solution. Accordingly, one detection channel for carbaryl on the PAT-CDs@SiO2 platform was developed with a ratiometric approach applying dual-signal readouts from two emission wavelengths. Moreover, two detection channels for thiram and chlorpyrifos with a fluorescence and phosphorescence dual-signal readout strategy were built on the PAT-CDs@SiO2 platform. Finally, the proposed multi-channel detection method recovered carbaryl, thiram and chlorpyrifos in tap and pork samples in the range of 90% and 110%. Therefore, multi-channel detection functions can be potentially integrated onto CDs based platforms due to their high performances and possible multi-signal readouts.

Abstract:In this study, based on the oxidase activity and photothermal effect of manganese dioxide nanosheets (MnO2 NSs), with thiamine (TH) as the fluorescence response signal and tris (2,2'-bipyridyl) ruthenium (II) hexahydrate as the reference signal, an enzyme-regulated ratiometric fluorescence and photothermal dual-mode probe was constructed for the quantitative detection of organophosphorus pesticide (OPs) residues. OPs reduced the production of the reductive product thiocholine by inhibiting the activity of acetylcholinesterase, thereby regulating the residual amount of MnO2 NSs. With the increase of OPs concentration, the color of the probe solution gradually transitioned from red to blue, and the temperature gradually increased. Using dichlorvos and chlorpyrifos as pesticide models, the developed probes exhibited sensitive responses to OPs in a wide linear range of 0.1-8000 ng/mL. The detection limits of dichlorvos and chlorpyrifos in fluorescence mode were 1.13 × 10-3 ng/mL and 0.86 ng/mL, respectively. The corresponding detection limits in photothermal mode were 1.01 ng/mL and 1.02 ng/mL, respectively. The proposed probe displayed excellent anti-interference and reliability in the analysis of OPs residues in real samples. The dual-mode probe with self-verification function is expected to provide more accurate and robust detection results than the single-mode probe, and has a wider application prospect.

Abstract:In this study, a nanozyme (ZIF-Co-Cys) with high oxidase-like catalytic activity was prepared, and a ratiometric fluorescent/photothermal dual-mode probe was constructed for organophosphorus pesticides (OPs) detection based on the competitive effect of ZIF-Co-Cys and the enzymatic reaction product of acid phosphatase (ACP) on o-phenylenediamine and the inhibition effect of OPs on ACP activity. Using dimethyl dichloroviny phosphate (DDVP) as the model, both the fluorescence intensity ratio and the temperature change of the probe solution exhibited an excellent correlation with OPs concentration. The detection limits were 1.64 ng/mL and 0.084 ng/mL, respectively. Additionally, the detection of DDVP residues in real samples verified the outstanding anti-interference and accuracy of the probe. This work not only provided a complementary dual-mode method for the accurate and rapid detection of OPs residues in complex samples, but also supplied a new insight into the design of a multi-mode sensing platform based on the cascade reaction of nanozyme.

Abstract:Pesticides are enormously important to industry requirements and in agriculture for improving yields. Parathion is widely used to control pests in vegetables, fruits, and flowers. However, excessive use of parathion poses a threat to food safety, the environment, and human health. Due to low cost, ease to use, and high selectivity and sensitivity, fluorescent nanoprobe is a candidate for parathion detection. The blue fluorescent carbon dots were prepared by hydrothermal method with rutinum and o-phenylenediamine as precursors. The Rut-CDs were purified by dialysis, thin layer chromatography, and chromatographic column. Excellent linear ranges of 0-7.5 mg/L and 12.5-62.5 mg/L parathion were obtained with a low detection limit of 0.11 mg/mL. In addition, the quenching mechanism of parathion to the fluorescent of Rut-CDs was illuminated. Furthermore, the nanoprobe was efficiently applied to determine the parathion content in Chinese cabbage, cantaloupe, and cowpea samples. It shows great potential to detect parathion.

Abstract:In this study, we used purine hydrazone derivatives and coumarin aldehyde to synthesize a novel fluorescent sensor (EDTP) by Schiff base reaction, which exhibited significant selective fluorescence quenching of Cu2+, and a distinct change from brilliant yellow to red is present along with the solution color. The detection limit of EDTP for Cu2+ was 109.52 nM. Job's plot experiment, density flooding theory (DFT) and 1H NMR titration experiments revealed the possible binding mechanism of EDTP to Cu2+, the probe EDTP could achieve highly detection of Cu2+ through forming a 1:1 complex. Additionally, this new fluorescent sensor EDTP-Cu2+ can be further applied in the rapid and selective detection of pesticide residues in solutions. When the EDTP-Cu2+ system was subsequently exposed to organophosphorus pesticides (glyphosate and glufosinate-ammonium), it was observed that the fluorescence was recovered and accompanied by a red to yellow color change. This may be attributed to the strong chelation of glyphosate and glufosinate-ammonium with Cu2+, leading to the dissociation of the EDTP-Cu2+ system and thus triggering the fluorescence recovery effect. The detection limits of the EDTP-Cu2+ system is 2.48 nM for glyphosate and 17.23 nM for glufosinate-ammonium, respectively. Finally, the developed sensor system has been successfully utilized image glyphosate and glufosinate-ammonium fluorescence in living cells. Purine fluorescence probes are a potential fluorescent probe for the detection of metal ions and pesticides due to their good characteristics. This study opens up a new way for the detection of fluorescent probes in pesticides.

Abstract:Multiple dopants contained carbon dots (CDs) with enhanced selectivity have aroused extensive attentions. In this work, three heteroatoms (nitrogen, sulfur, boron) and flavonoid moiety derived from the extracts of Ginkgo biloba leaves co-incorporated CDs were synthesized based on the hydrothermal strategy for novel carbon materials investigation and sensing applications. Nitrogen and flavonoid moiety co-doped CD (N-CD-Fla), nitrogen, sulfur, and flavonoid moiety co-doped CD (NS-CD-Fla), nitrogen, sulfur, boron and flavonoid moiety co-doped CD (NSB-CD-Fla) were prepared, respectively. Three CDs had diversities in nanoparticle sizes, chemical structures, and luminescence properties. NS-CD-Fla exhibited blue luminescence and had higher quantum yield (QY) than N-CD-Fla (yellow-green) and NSB-CD-Fla (blue-green), which can be attributed to its higher N content and N, S synergistic enhancement effect. The luminescence of the three CDs was speculated to be related with the electron transition of CN bonds derived from grafting amines groups onto the flavonoid-related moieties. N-CD-Fla, NS-CD-Fla, and NSB-CD-Fla were further proved to be sensitive to fenitrothion (Fen), dithianon (Dit), dinoseb (Din) and their structural analogues, they were employed as superior chemosensors for the three pesticides sensitive and selective detection. The application potentials of the novel chemosensors based analytical methods were further evaluated by real sample analysis.

Abstract:The residues of organophosphorus pesticides (OPs) are increasing environmental pollution and public health concerns. Thus, the development of simple, convenient and sensitive method for detection of OPs is crucial. Herein, a multifunctional Fe-based MOF with fluorescence, catalytic and adsorption, is synthesized by a simple one-pot hydrothermal method. The ratiometric fluorescence sensor for detection of OPs is constructed by using only one multifunctional sensing material. The NH2-MIL-101(Fe) is able catalyze the o-phenylenediamine (OPD) into 2,3-diaminophenazine (DAP) in the presence of H2O2. The generated DAP can significantly quench the intrinsic fluorescence of NH2-MIL-101(Fe) by the fluorescence resonance energy transfer (FRET) and internal filtration effect (IFE), while producing a new measurable fluorescence. Without immobilization or molecular imprinting, pyrophosphate ion (PPi) can inhibit the peroxidase-like activity of the NH2-MIL-101(Fe) by chelating with Fe3+/Fe2+ redox couple. Moreover, PPi can also be hydrolyzed by alkaline phosphatase (ALP), the presence of OPs inhibits the activity of ALP, resulting in the increase of extra PPi preservation and signal changes of ratiometric fluorescence, the interactions of ALP with different OPs are explored by molecular docking, the OPs (e.g., glyphosate) interact with crucial amino acid residues (Asp, Ser, Ala, Lys and Arg) are indicated. The proposed sensor exhibits excellent detection performance for OPs with the detection limit of 18.7 nM, which provides a promising strategy for detection of OPs.

Abstract:<p>Organophosphorus compounds are toxic organic phosphorus molecules including nerve agents and pesticides. Organophosphorus pesticides play a significant role in improving large-scale modern agriculture. Nerve agents are neurotoxic chemicals. Therefore, there is a demand for assays to measure organophosphorus analogs in a rapid, sensitive, and convenient way. Here, colorimetric and fluorescence sensors can be an effective approach for sensing these substances. They have been extensively explored due to their portability and high sensitivity. This review summarizes recent development in colorimetric and fluorescent materials including small organic fluorophores, nanomaterials, complexes, and polymers since 2015. We describe advances in sensing a variety of organophosphorus analogs. We also discuss recent challenges and design strategies in developing colorimetric and fluorescent assays with high performance and specific sensing.</p>

Abstract:Water-soluble non-conjugated polymer dots (PDs) have been synthesized using hyperbranched polyethyleneimine (PEI) and dihydroxybenzaldehyde (DHB) for the first time via the Schiff base reaction at room temperature. The yielded non-conjugated PDs of PEI-DHB could display the well-defined spheric structure and good water solubility. In contrast to the common PDs otherwise showing blue emission, the PEI-DHB PDs could give out strong green fluorescence in aqueous media. Especially, the fluorescence of the PEI-DHB PDs could be specifically quenched by MnO2 nanosheets through the inner filter effects and further restored by the thiocholine that could reduce MnO2 nanosheets into Mn2+. Herein, thiocholine could be produced in hydrolysis reaction of acetylthiocholine catalyzed by the acetylcholinesterase (AChE), of which the catalytic activity could be irreversibly inhibitted by the introduction of organophosphates. A highly selective fluorimetric method was thereby been developed for the detection of organophosphorus pesticides using dimethyl-dichloro-vinyl phosphate as a model. The linear concentrations ranges from 0.050 to 2.5 μM. Importantly, the non-conjugated PDs probes with strong green fluorescence and high water solubility may promise the extensive applications in the environmental, food, and clinical analysis fields.

Abstract:Pesticide residues have seriously threatened human health and affected the sustainable development of the environment. It is pivotal to establish an efficient detection platform for pesticide assays. A novel ratiometric fluorescent aptasensor for sensitive determination of pesticide has been successfully established. Carbon dots (CDs) are used as the reference emitter, while the T-rich single-stranded DNA templated copper nanoparticles (CuNPs) provide the increasing signal. More precisely, CDs/magnetic beads (MBs) combined with split aptamer are used in this detection platform to present a label-free and enzyme-free isocarbophos (ICP) detection with high sensitivity and specificity. In the presence of ICP, a sandwich-typed complex is formed which attaches a T-rich DNA. After magnetic separation and addition of Cu2+/ascorbic acid (AA), the CuNPs are generated and the strong red luminescent appears. Without ICP, just blue emission from CDs exits. The developed ratiometric aptasensor offers an excellent ICP detection in the linear range of 3–200 nM with a detection limit of 0.81 nM. It provides new ways for the assay of pesticides in agricultural products or environmental samples.

Abstract:The toxicological influence and pervasive manifestation of hazardous pesticides posture adverse consequences on humans and a number of environmental matrices, directly through bioaccumulation or indirectly via the food chain. Much emphasis has been paid to reevaluating how to manage pesticide usage practices without affecting or damaging the environment due to the persistent existence of pesticide residues over acceptable levels in numerous forms. Therefore, the studies related to the creation of trustworthy techniques for pesticide monitoring are hotspot across the globe. It has been suggested that carbon dots (C-dots), which have outstanding biocompatibility strong photostability and low toxicity, could replace other materials in the construction of several optical sensors for the detection of pesticide. Based on the complex nature and health hazards of the pesticides, this manuscript comprehensively summarizes the overview of recent developments of C-dots based optical sensors for pesticide detection with special emphasis on colorimetric and photoluminescent sensors. This section is further nourished by highlighting the sensing mechanisms, structural design through integration of types of recognition elements such as, nanoparticles, enzymes, aptamers, antibodies and some of the molecularly imprinted polymers. Beyond evaluating how well these suggested optical sensors operate in practical applications, detailed discussions regarding current monitoring gaps and difficulties as well as potential future directions has also been presented. It is believed that this review would offer significant new insights for the creation of novel C-dots based pesticide monitoring systems with a large variety of applications.

One-Pot Preparation of Ratiometric Fluorescent Molecularly Imprinted Polymer Nanosensor for Sensitive and Selective Detection of 2,4-Dichlorophenoxyacetic Acid

Abstract:The development of fluorescent molecular imprinting sensors for direct, rapid, and sensitive detection of small organic molecules in aqueous systems has always presented a significant challenge in the field of detection. In this study, we successfully prepared a hydrophilic colloidal molecular imprinted polymer (MIP) with 2,4-dichlorophenoxyacetic acid (2,4-D) using a one-pot approach that incorporated polyglycerol methacrylate (PGMMA-TTC), a hydrophilic macromolecular chain transfer agent, to mediate reversible addition-fragmentation chain transfer precipitation polymerization (RAFTPP). To simplify the polymerization process while achieving ratiometric fluorescence detection, red fluorescent CdTe quantum dots (QDs) and green fluorescent nitrobenzodiazole (NBD) were introduced as fluorophores (with NBD serving as an enhancer to the template and QDs being inert). This strategy effectively eliminated background noise and significantly improved detection accuracy. Uniform-sized MIP microspheres with high surface hydrophilicity and incorporated ratiometric fluorescent labels were successfully synthesized. In aqueous systems, the hydrophilic ratio fluorescent MIP exhibited a linear response range from 0 to 25 μM for the template molecule 2,4-D with a detection limit of 0.13 μM. These results demonstrate that the ratiometric fluorescent MIP possesses excellent recognition characteristics and selectivity towards 2,4-D, thus, making it suitable for selective detection of trace amounts of pesticide 2,4-D in aqueous systems.
Sensitive colorimetric and fluorescence dual-mode detection of thiophanate-methyl based on spherical Fe3O4/GONRs composite nanozyme

Abstract:Pesticide detection based on nanozyme is largely limited in terms of the variety of pesticides. Herein, a spherical and well-dispersed Fe3O4/graphene oxide nanoribbons (Fe3O4/GONRs) composite nanozyme was applied to firstly develop an enzyme-free and sensitive colorimetric and fluorescence dual-mode detection of thiophanate-methyl (TM). The synthesized Fe3O4/GONRs possess excellent dual enzyme-like activities (peroxidase and catalase) and can catalyze H2O2 to oxidize 3,3',5,5'-tetramethylbenzidine (TMB) into oxidized TMB (oxTMB). We found that Fe3O4/GONRs can adsorb TM through the synergistic effect of multiple forces, thereby inhibiting the catalytic activities of nanozyme. This inhibition can modulate the transformation of TMB to oxTMB, producing dual responses of absorbance decrease (oxTMB) and fluorescence enhancement (TMB). The limits of detection (LODs) of TM were 28.1 ng/mL (colorimetric) and 8.81 ng/mL (fluorescence), respectively. Moreover, the developed method with the recoveries of 94.8-100.8% also exhibited a good potential application in the detection of pesticides residues in water and food samples.
Integrating Peroxidase-Mimicking Activity with Photoluminescence into One Framework Structure for High-Performance Ratiometric Fluorescent Pesticide Sensing

Abstract:We develop a robust ratiometric fluorescent nanosensor based on bifunctional Fe-based metal-organic frameworks (NH2-MIL-101(Fe)) for high-performance pesticide determination. The NH2-MIL-101(Fe) plays dual roles: the 2-aminoterephthalic acid ligand endows the framework with a photoluminescence at 428 nm, and the Fe node exhibits peroxidase-mimetic catalytic activity to oxidize the o-phenylenediamine substrate to diaminophenazine (DAP) with a fluorescence at 556 nm, which in turn suppresses the intrinsic signal (428 nm) of NH2-MIL-101(Fe) due to inner filter effect. When acetylcholinesterase is introduced for hydrolyzing acetylthiocholine chloride to thiocholine, the product with certain reducibility can inhibit the o-phenylenediamine oxidation, resulting in the DAP signal suppression (556 nm) and the NH2-MIL-101(Fe) one recovery (428 nm). Since pesticide residues effectively inhibit the activity of acetylcholinesterase, their presence can increase the formation of DAP and its fluorescence at 556 nm, thus the inherent signal of NH2-MIL-101(Fe) at 428 nm is suppressed again. By taking carbaryl as a pesticide model, the ratiometric nanosensor exhibits good sensitivity for carbaryl sensing in the scope of 2-100 ng/mL with a 1.45 ng/mL detection limit. No notable interferences from other species are observed for target detection. Moreover, our nanosensor is confirmed to be reliable for carbaryl analysis in real samples.
