Abstract:Many taxa in the Acrididae have controversial phylogenetic positions. A typical example of such controversies is the phylogenetic positions of the genera Caryandoides, Paratoacris, Fer and Longchuanacris, as well as some other related taxa, which were placed in Oxyinae by some authors, but were considered members of the subfamily Catantopinae by others. In this study, the complete mitogenomes of nine species were sequenced using next-generation sequencing, the characteristics of the newly sequenced mitogenomes are presented briefly, and the phylogeny of the Oxyinae and Catantopinae are reconstructed using a selected dataset of mitogenome sequences under maximum likelihood and Bayesian inference frameworks. The results show that the four controversial genera were consistently assigned to the subfamily Oxyinae rather than Catantopinae in all phylogenetic trees deduced from different datasets under different frameworks, and this finding is entirely consistent with their morphological characters. Therefore, it is more appropriate to place them in Oxyinae rather than Catantopinae. In addition, the results from our analysis also confirm the membership of the genus Apalacris in Coptacrinae rather than Catantopinae, and indicate the uncertainty in the phylogenetic position of the genus Traulia, and a more in-depth study is necessary to resolve the relationship of Traulia with other catantopine groups or Coptacrinae.

The Complete Mitogenomes of Three Grasshopper Species with Special Notes on the Phylogenetic Positions of Some Related Genera.

Abstract:Clarifying phylogenetic position and reconstructing robust phylogeny of groups using various evidences are an eternal theme for taxonomy and systematics. In this study, the complete mitogenomes of Longzhouacris mirabilis, Ranacris albicornis, and Conophyma zhaosuensis were sequenced using next-generation sequencing (NGS), and the characteristics of the mitogenomes are presented briefly. The mitogenomes of the three species are all circular molecules with total lengths of 16,164 bp, 15,720 bp, and 16,190 bp, respectively. The gene structures and orders, as well as the characteristics of the mitogenomes, are similar to those of other published mitogenomes in Caelifera. The phylogeny of the main subfamilies of Acrididae with prosternal process was reconstructed using a selected dataset of mitogenome sequences under maximum likelihood (ML) and Bayesian inference (BI) frameworks. The results showed that the genus Emeiacris consistently fell into the subfamily Melanoplinae rather than Oxyinae, and the genus Choroedocus had the closest relationship with Shirackiacris of the subfamily Eyprepocnemidinae in both phylogenetic trees deduced from mitogenome protein coding genes (PCGs). This finding is entirely consistent with the morphological characters, which indicate that Emeiacris belongs to Melanoplinae and Choroedocus belongs to Eyprepocnemidinae. In addition, the genera Conophymacris and Xiangelilacris, as well as Ranacris and Menglacris, are two pairs of the closest relatives, but their phylogenetic positions need further study to clarify.

Phylogenetic Reconstruction of the Family Acrypteridae (orthoptera: Acridoidea) Based on Mitochondrial Cytochrome B Gene.

Abstract:Sequences from the mitochondrial cytochrome b gene (Cyt b) were determined for 25 species from the superfamily Acridoidae and the homologous sequences of 19 species of grasshoppers were downloaded from the GenBank data library. The purpose was to develop a molecular phylogeny of the Acrypteridae, and to interpret the phylogenetic position of the family within the superfamily Acridoidea. Phylogeny was reconstructed by Maximum-parsimony (MP) and Bayesian criteria using Yunnanites coriacea and Tagasta marginella as outgroups. The alignment length of the fragments was 384 bp after excluding ambiguous sites, including 167 parsimony informative sites. In the fragments, the percentages of A + T and G + C were 70.7% and 29.3%, respectively. The monophyly of Arcypteridae is not supported by phylogenetic trees. Within the Arcypteridae, neither Arcypterinae nor Ceracrinae is supported as a monophyletic group. The current genus Chorthippus is not a monophyletic group, and should be a polyphyletic group. The present results are significantly different from the classification scheme of Arcypteridae, which is based on morphology.
Molecular phylogenetic analysis of five subfamilies of the Acrididae(Orthoptera:Acridoidea) based on the mitochondrial cytochrome b and cytochrome c oxidase subunit I gene sequences

Abstract:The homologus sequences of the mitochondrial cytochrome b (Cytb) and cytochrome c oxidase subunit I (COI) genes were sequenced in 17 species representing five subfamilies (belonging to family Acrididae):Melanoplinae,Catantopinae, Cyrtacanthacridinae,Oedipedinae and Gomphocerinae from China.The concatenated sequence from both genes was 1 998 bp in length,consisting of 1 266 bp and 732 bp for COI and Cytb respectively.In the concatenated sequence, A + T content was about 72.13%,and C + G only 27.87%.There are 889 sites conserved,1 109 sites were variable,and 838 sites were parsimony-informative in the variable sites.Phylogenetic trees were reconstructed with maximum parsimony,neighbor-joining,and maximum likelihood using Erianthus versicolor and Erianthus sp.of the Eumastacoidea as outgroups.Our results showed that the monophyly of the subfamilies Gomphocerinae and Catantopinae were not supported here.The phylogenetic trees did not entirely agree with the international taxonomic system of grasshoppers.The subfamily Oedipodinae clustered as one clade,which was supported as a monophyletic group.Based on the close relationships among four subfamilies,Gomphocerinae,Catantopinae,Cyrtacanthacridinae and Melanoplinae,we suggest that the four subfamilies should be considered to merge into one subfamily.Simultaneously,we found it was not very dependable to infer the phylogenetic relationships among subfamilies within the Acrididae based on the concatenated sequence from Cytb and COI genes.

Abstract:Phylogenetic relationships of Acridoidea were examined using mitochondrial cytochrome oxidase subunit sequences (COI, COII and COIII, total 2970bp). Fourteen grasshopper species of thirteen genera from seven families were sequenced to obtain mitochondrial genes data, along with twenty-two grasshopper species were obtained from the GenBank nucleotide database. The purpose of this study is to infer the phylogenetic relationships among families within Acridoidea and testing the monophyly of Acridoidea and each families of it. Phylogenic trees were reconstructed using Maximum Likelihood (ML) and Maximum Parsimony (MP) methods with Tettigonioidea and Gryllotalpoidea as outgroups. The putative initiation codon for COI is CCG in thirteen studied species and ATC in Bryodema luctuosum luctuosum. The 2970 bp concatenated sequences included 1431 conserved sites, 1539 variable sites, and 1216 parsimony-informative sites, the nucleotide compositions were significantly biased toward A and T (68.8%). The resulted phylogenetic trees supported the monophyly of Acridoidea, but did not entirely agree with the traditional morphology-based taxonomic system of grasshoppers within Acridoidea. The monophyly of three families of Acrididae, Catantopidae and Arcypteridae were not supported; Gomphoceridae and Arcypteridae were recovered together as a monophyletic group because of closer phylogenetic relationships; Pyrgomorphidae and Chrotogonidae have the same closer relationships; Pneumoridae, Pyrgomorphidae and Chrotogonidae were the most basal groups; while the taxonomic status of Pamphagidae, which was revealed as a monophyletic group, was not clear in this analysis. Moreover, the results indicate that a phylogeny inferred from the combination of several genes is more reliable than that from only a single gene sequence, and the third codon positions of protein coding genes can improve the topology and node supports of the phylogenetic trees.
A Phylogenetic Analysis of Six Genera of Acrididae and Monophyly of Acrididae in China Using 16S Rdna Sequences (orthoptera, Acridoidea)

Abstract:In Xia's system, the family Acrididae has long been recognized as a natural, monophyletic group, which contain six subfamilies in China. However, there has been much dispute over its monophyly. To investigate its monophyly and phylogenetic relationships among six genera Carsula, Phlaeoba, Mongolotettix, Gelastorhinus, Acrida, and Gonista within this family, we sequenced 450 bp fragments of 16S rRNA gene sequences of 14 recognized species and downloaded the sequences of another grasshopper Acrida turrita and two Tetrigidae species from GenBank. Phylogenetic trees were reconstructed with maximum parsimony, neighbor-joining, and maximum likelihood using the Tetrigidae species as outgroup. The monophyly of Acrididae was not supported in all analyses. The phylogenetic trees did not agree with the Chinese traditional classifications, but they just agreed well with Kevan's, which is popular in western countries. This prompt that the Chinese system must be earnestly improved and consummated. However, monophyly of genus Acrida was strongly supported. Leptacrinae was closely related to the species of the Chinese Catantopidae, not to Acrididae. Although gene sequences obtained from Acrida rufipes and Acrida shanghaica were identical, we could not conclude that they are the same species [Acta Zoologica Sinica 52 (2): 302 - 308, 2006].
Characterization of the mitochondrial genomics and phylogeny of Orthoptera (Insecta: Arthropoda)

Abstract:To date,complete mitogenomes from 167 taxa within Orthoptera have been collected in GenBank database,which included extant representing 9 caeliferan superfamilies (22 caeliferan families with 99 taxa) and 7 ensiferan superfamilies (12 ensiferan families with 68 taxa).Based on these sequences,the basic characterization of orthopteran mitogenomes were analyzed,and we summarized the application in phylogenetic analysis of orthopteran mitogenomes.Moreover,a robust phylogeny of orthopteran were also established based on complete mitogenomes.The main results are as follows:(1) There are 8 patterns of gene arrangements in orthopteran mitogenomes,trnK-trnD only exists in caeliferan,trnN-trnS-trnE only exists in Grylloidea and trnM-trnI-(-trnQ) only exists in Pseudophyllinae;(2) The nucleotide composition in orthopteran mitogenomes showed obvious AT composition bias;(3) Different protein-coding genes have different evolutionary rates in Orthoptera;(4) The monophyly of Orthoptera,caeliferan and ensiferan was supported;(5) We failed to recover monophyly of Stenopelmatoidea;(6) The monophyly of superfamilies in caeliferan were all supported,and the relationships among 9 superfamilies of caeliferan were (Tridactyloidea + (Tetrigoidea + (Proscopioidea + (Eumastacoidea + (Tanaoceroidea + (Pneumoroidea + Trigonopterygoidea) + (Pyrgomorphoidea + Acridoidea)))))).

Abstract:To enrich the genomic database of Catantopinae (Orthoptera: Acrididae), mitogenomes of three species from different genera, Traulia nigritibialis (15,701 bp), Choroedocus capensis (16,293 bp) and Stenocatantops splendens (15,574 bp), were characterized and compared with those of other grasshoppers in the subfamily. All 13 proteincoding genes (PCGs) were initiated by ATN codons except COI with ACC (C. capensis and S. splendens) and ND6 with TTG (S. splendens). All transfer RNA (tRNA) genes had a typical clover-leaf structure, except tRNA(ser(AGN)) in which the base pairs of the dihydrouridine (DHU) arm were reduced. The phylogenetic relationships were constructed among 22 species from four subfamiles of Acrididae by classical classifications based on two datasets of their mitogenomes using both Bayesian Inference (BI) and Maximum Likelihood (ML). The phylogenetic analysis confirmed the monophyly of the three other subfamilies, but did not provided support of the monophyly of Catantopinae.
Molecular Phylogeny of Some Subfamilies of Catantopidae (orthoptera: Caelifera: Acridoidea) in China Based on Partial Sequence of Mitochondrial COⅡ Gene

Abstract:A total of 22 species （representing 10 subfamilies and 16 genera） and an outgroup species, Filchnerella sunanensis, were sampled in this study. A 585 bp sequence of COⅡ gene was amplified using PCR, and then sequenced directly through cycling sequencing method. Several statistical approaches were used to test the phylogenetic signal in the DNA sequence data sets and three partition data sets, which were defined based on different codon positions. Phylogenetic analyses by the Neighbor Joining （N J）, maximum parsimony （MP）, maximum likelihood （ML） and Bayesian inference （BI） were employed to reconstruct the phylogeny. The results indicated that the COⅡ gene sequences of 22 data sets contained strong phylogenetic signal. It can species were highly A + T biased. All of the different be found from the consensus tree that：（1） the relationships among Calliptaminae, Cyrtacanthacridinae, Eyprepocnemisinae and Catantopinae are close. （2） The relationship between Caryandinae and Oxyinae is also close. It is so reasonable to include the subfamily Caryandinae into the subfamily Oxyinae, with Spathosterninae as sister group. （3） Melanoplinae should merge with Podisminae into one subfamily. （4） The relationships of species and genera within Coptacrinae, as well as its relationships with other subfamilies were not resolved. （5） The relationship between Melanoplinae and Podisminae is close, and both are distant to other subfamilies here studied. The results prove that COⅡ gene is a useful marker to infer phylogenetic relationships among species and genera within a subfamily.

A key to all Asian species (except for the eparmata species complex) of this subgenus was provided. All currently available DNA barcode (partial mitochondrial cytochrome c oxidase subunit I (COI) gene) sequences of this subgenus (217 sequences of 54 species) are employed in a molecular analysis using different species delimitation methods. The results indicate that approximately 68.5% (37 of 54 spp.) of Phortica (s. str.) species could be clearly distinguished from closely related morphospecies or cryptic species.
Complete Mitochondrial Genome of Atractomorpha Sagittaris (orthoptera: Pyrgomorphidae) and Its Phylogenetic Analysis for Acrididea

Abstract:We sequenced the complete mitogenome of Atractomorpha sagittaris (Orthoptera: Pyrgomorphidae) and compared with other Acrididea species. The circular genome was 15,763 bp in size and contained the entire set of 37 genes usually present in orthopteran mitogenomes. The AT-skew of this mitogenome was slightly positive (0.17) and the nucleotide composition biased toward A + T nucleotides (73.38%). All of the 13 PCGs initiated with canonical start codon (ATN), except for COI initiating with ACT. In addition, analyses of pairwise genetic distances between individual PCGs in Acrididea species showed that ND1 was the least conserved gene, while COI the most conserved. Furthermore, we reconstructed phylogenetic trees among species from six superfamiles in Acrididea using Bayesian Interference and Maximum Likelihood methods. Our phylogenetic analyses offered support of monophyly of the superfamilies, and reconfirmed some of species groups including A. sagittaris in Pyrgomorphidae.

Abstract:The complete mitochondrial genome of Tonkinacris sinensis is 15,627 bp long and contains13 protein-coding genes (PCGs), 22 tRNA genes, 2 rRNA genes and one A + T-rich region. The gene order and orientation are identical to those of other Orthoptera species, containing the rearrangement of trnD and trnK. Intriguingly, a tRNA(Ser)-like gene exists on the N strand between the trnSUCN and nad1 genes. The length of this gene is 110 bp, and it has a typical clover-leaf structure, an anticodon, and a high cove score (23.49). On its clover-leaf structure, on the anticodon arm, there is a 41 bp intron with an unknown function. Here, phylogenetic analysis was conducted based on 13 PCGs of 30 species from 9 subfamilies of Acrididae to understand their phylogenetic relationships. According to the phylogenetic tree, the relationship among the 9 subfamilies within Acrididae was as follows: (Spathosterninae + (Oxyinae + (Catantopinae + (Calliptaminae + (Cyrtacanthacridinae + (Melanoplinae + (Gomphocerinae + (Oedipodinae + Acridinae)))))))). Abstract:The phylogenetic relationships of 26 species from nine subfamilies Catantopidae were reconstructed using combined sequence data set of Cyt b,16S rDNA and 28S rDNA.The results suggest that:1) the monophyly of all subfamilies except for Melanoplinae,Podisminae and Coptocrinae can be supported;2) melanoplinae should merge with Podisminae and form one subfamily,and both are distant to other subfamilies;3) Traulia should be moved from the subfamily Coptocrinae;4) the Caryandinae and Oxyinae are sister groups;5) the Calliptaninae,Eypropocnemidinae,Catantopinae,Cyrtacanthacridinae and the genera Traulia are closely related.

Abstract:The complete 15,831 bp nucleotide sequence of the mitochondrial genome from Elimaea cheni (Phaneropterinae) was determined. The putative initiation codon for cox1 was TTA. The phylogeny of Orthoptera based on different mtDNA datasets were analyzed with maximum likelihood (ML) and Bayesian inference (BI). When all 37 genes (mtDNA) were analyzed simultaneously, the monophyly of Caelifera and Ensifera were recovered in the context of our taxon sampling. The phylogeny of Orthoptera was largely consistent with previous phylogenetic hypotheses. Rhaphidophoridae to be a sister group of Tettigoniidae, and the relationships among four subfamilies of Tettigoniidae were (Phaneropterinae + (Conocephalinae + (Bradyporinae + Tettigoniinae))). Pyrgomorphidae was the most basal group of Caelifera. The relationships among six acridid subfamilies were (Oedipodinae + (Acridinae + (Gomphocerinae + (Oxyinae + (Calliptaminae + Cyrtacanthacridinae))))). However, we did not recover a monophyletic Grylloidea. Myrmecophilidae clustered into one clade with Gryllotalpidae instead of with Gryllidae. ML and BI analyses of all protein coding genes (using all nucleotide sequence data or excluding the third codon position, and amino acid sequences) revealed a topology identical to that of the entire mtDNA genome dataset. However, 22 tRNAs genes excluding the DHU loop and TψC loop (TRNA), and two rRNA genes (RRNA) perform poorly when analyzed as single dataset. Our results suggest that the best phylogenetic inferences were ML and BI methods based on total mtDNA. Excluding tRNA genes, rRNA genes and the third codon position of protein coding genes from dataset and converting nucleotide sequences to amino acid sequences do not positively affect phylogenetic reconstruction.

Abstract:The complete 15,599-bp mitogenome ofAcrida cinereawas determined and compared with that of the other 20 orthopterans. It displays characteristic gene content, genome organization, nucleotide composition, and codon usage found in other Caelifera mitogenomes. Comparison of 21 orthopteran sequences revealed that the tRNAs encoded by the H-strand appear more conserved than those by the L-stand. All tRNAs form the typical clover-leaf structure excepttrnS(agn), and most of the size variation among tRNAs stemmed from the length variation in the arm and loop of TΨC and the loop of DHU. The derived secondary structure models of therrnSandrrnLfrom 21 orthoptera species closely resemble those from other insects on CRW except a considerably enlarged loop of helix 1399 ofrrnSin Caelifera, which is a potentially autapomorphy of Caelifera. In the A+T-rich region, tandem repeats are not only conserved in the closely related mitogenome but also share some conserved motifs in the same subfamily. A stem-loop structure, 16 bp or longer, is likely to be involved in replication initiation in Caelifera and Grylloidea. A long T-stretch (>17 bp) with conserved stem-loop structure next torrnSon the H-strand, bounded by a purine at either end, exists in the three species from Tettigoniidae.
Comparative Analysis of Mitogenomes among Three Species of Grasshoppers (orthoptera: Acridoidea: Gomphocerinae) and Their Phylogenetic Implications

Abstract:Whole mitochondrial genomes have been widely used in phylogenetic analysis, population genetics and biogeography studies. This study sequenced and characterized three complete mitochondrial genomes (Dasyhippus peipingensis, Myrmeleotettix palpalis, Aeropedellus prominemarginis) and determined their phylogenetic position in Acrididae. The length of the mitochondrial genomes ranged from 15,621-15,629 bp and composed of 13 PCGs, 2 rRNA, 22 tRNA genes and an AT control region. The arrangement and structure of the mitochondrial genomes were similar to those of other invertebrates. Comparative genomics revealed that the three mitochondrial genomes were highly conserved in terms of gene size, structure, and codon usage, all PCGs were purified selections with an ATN start codon and a TAN stop codon. All tRNAs could be folded into the typical clover-leaf structure, except tRNA Ser (AGN) that lacked a dihydrouridine (DHU) arm. Phylogenetic analysis based on 13 PCGs of 34 Acrididae species and seven outgroup species revealed that differences in the shape of antennae within the family Acrididae should be given less weight as a taxonomic character for higher-level classification. Moreover, the divergence time estimates indicates that in Gomphocerinae, the species with clubbed antennae were formed within the nearest 18 Mya, and Pacris xizangensis is more ancient.

Abstract:To date, 12 species group taxa (including subspecies) of the grasshopper genus Ceracris are generally recognized, of which nine species occur in China. The phylogeny of Ceracrine grasshoppers is unknown. Using partial DNA sequence data from mitochondrial 16S and 12S rRNA genes, we reconstruct the phylogeny of some Chinese grasshoppers belonging to the subfamily Ceracrinae. Both Maximum Parsimony and Neighbor-Joining methods were employed. Our phylogeny suggests that the examined taxa of Ceracris are 1) monophyletic; 2) composed of two sister taxa; 3) does not include Rammeacris kiangsu, and agree with the results of current morphological taxonomy research; and 4) Ceracris versicolor is closely related to Ceracris fasciata contrary to morphological suggestions.

Abstract:Background and aims: In Xia's taxonomic revision, Gomphocerus rufus (Linnaeus, 1758), Chorthippus chinensis and Phlaeoba albonema belong to the families Gomphoceridae, Arcypteridae and Acrididae, respectively; whereas in Otte's taxonomic analysis of Orthoptera, all three species belong to the subfamily Gomphocerinae, family Acrididae.Materials and methods: We determined the mitochondrial genomes (mitogenomes) of G. rufus, compared these with 10 other caeliferan mitogenomes, and performed phylogenetic analyses in order to clarify the relationships of the three families in Xia's taxonomic revision and which study is more accurate in defining the relationships of the three families. Furthermore, the mitogenome of Primnoa arctica (Zhang and Jin, 1985) was determined. This is the first mitogenome of the subfamily Catantopinae, superfamily Acridoidea. Through the comparison of mitogenomes from six subfamilies of the superfamily Acridoidea and one species of Pyrgomorphoidea, we hope to summarize a general law on the composition of the caeliferan mitogenome.Results: The two molecules contain the same set of mitochondrial genes for 22 tRNAs, 2 rRNAs, 13 proteins, and a non-coding, AT-rich region. The base composition, gene order, and codon usage of the two genomes conform to those reported for other caeliferan species. Both genomes possess the rearrangement of tRNA(Lys) and tRNA(Asp). Compared with their ancestral mitogenome, this is a significant difference between the mitogenome of the suborders Caelifera and Ensifera or other Metazoa. A stem-loop structure that is similar to a previously presumed one (that probably involved in replication initiation) was found at the A+T-rich region of each mitogenome. In the phylogenetic analyses, the species from suborders Caelifera and Ensifera cluster, respectively, as monophyletic groups, and the two suborders cluster as sister groups.Conclusion: Within Caelifera, the subfamily Gomphocerinae appears to be a paraphyletic group in the analyses of the protein-coding gene (PCG) dataset and a monophyletic group in the analyses of rRNA dataset. The subfamily Oedipodinae is a monophyletic group; superfamily Acridoidea is a monophyletic group, and Pyrgomorphoidea is a sister to Acridoidea. Regarding Xia's revision, the families Gomphoceridae and Arcypteridae are closely related, and the family Acrididae is a paraphyletic group. Overall, the phylogenetic analyses in our study are in accordance with Otte's taxonomic analysis.
