Abstract:Rhodium-catalyzed transannulation of 1,2,3-triazoles and ring-opening reactions of epoxides is described. A number of 3,4-dihydro-2H-1,4-oxazines are obtained in moderate yields probably involving generation of alpha-imino rhodium(II) carbene species.

Abstract:A rhodium-catalyzed denitrogenative formal (3 + 3) transannulation of 1,2,3-thiadiazoles with alk-2-enals is achieved, producing 2,3-dihydrothiopyran-4-ones in moderate to excellent yields. An inverse KIE of 0.49 is obtained, suggesting the reversibility of the oxidative addition of thioacyl Rh(i) carbenes to alk-2-enals. The late-stage structural modifications of steroid compounds are realized. Moreover, our studies show that thioacyl carbenes have different reactivities to those of α-oxo and α-imino carbenes, and highlight the importance of heteroatoms in deciding the reactivities of heterovinyl carbenes.

Abstract:Allenoates, a type of perpendicularly "strained" surrogate of alkynes, readily transannulate, site-specifically and regio-selectively, with 1,2,3-thiadiazoles in the presence of catalytic Rh(i)/bisphosphine, producing tetrasubstituted thiophenes in a redox-neutral manner. 1,2,3-Thiadiazoles act as masked 1,3-dipoles with nucleophilic sulfur and electrophilic carbon termini.

Abstract:1,2,3-Thiadiazoles serve as masked S-electrophilic thia-1,3-dipoles. Under rhodium/racemic BINAP catalysis, they undergo denitrogenative (3 + 2) umpolung transannulations with aryl isothiocyanates with inverse regioselectivity and excellent stereoselectivity, yielding N-aryl 3H-1,2-dithiol-(Z)-3-imines in a redox-neutral, step-efficient, and functionality-tolerant manner. Abstract:Herein, we report unprecedented Ni-catalyzed chemodivergent annulations to produce the 3,4-dihydrocoumarins and 2H-chromenes that are important attractive building blocks in many biologically active compounds. By the introduction of dcype ligand, a range of 3,4-dihydrocoumarins can be afforded with good to excellent yield and wide functional group tolerance via chemoselective intramolecular tan-dem isomerization/esterification. In the absence of ligand, a useful synthesis of 2H-chromenes has been developed through a nickel-catalyzed direct O-allylic substitution with good to excellent efficiency.(c) 2023 Elsevier Inc. Abstract:Comprehensive SummaryA rhodium‐catalyzed denitrogenative carbonylation of 1,2,3‐thiadiazole with alkynes to construct (benzo)thiochromenones is developed. Both terminal and internal alkynes were compatible in this [3+2+1] cycloaddition, affording a broad range of poly‐substituted thiochromenones. Unsymmetrical alkyne substrates exhibited good regioselectivity. It is noteworthy that the present strategy could be used to construct both monocyclic and benzo‐thiochromenones, and it is a novel example of denitrogenative transannulation with CO insertion.

Abstract:Novel cyclic fluorinated sulfoximines featuring an E-monofluoroalkenyl benzoisothiazole 1-oxide moiety as single diastereomers can be facilely accessed by Rh(III)-catalyzed redox-neutral [4+1] annulation of sulfoximines with α,α-difluoromethylene alkynes. The reaction proceeds with sequential selective cleavage of both a C-H and a C-F bond, thus endowing with high step and atom economy. Through a combined experimental and computational mechanistic study, the origin of annulative chemoselectivity, unconventional E-selectivity, and excellent diastereoselectivity have been revealed accordingly.

Abstract:1,2,3-Thiadiazoles are precursors of thioacylcarbenes and thioketenes and show various reactions with unsaturated compounds. The rhodium-catalyzed reactions of 1,2,3-thiadiazoles and alk-2-enenitriles, including alkyl, aryl, and heteroaryl acrylonitriles, have been investigated, showing chemospecific reactivity to generate 3-vinylisothiazole derivatives via the (3+2) transannulation between the cyano group of alk-2-enenitriles and generated thioacylcarbenes. (Z)-Cinnamonitriles are less active than (E)-cinnamonitriles due to the strong coordination of the (Z)-C=C bond and the Rh catalyst. The current reaction provides a method for the synthesis of 3-vinylisothiazole derivatives.

Abstract:A rhodium(III)-catalyzed cross-coupling of benzyl thioethers and aryl carboxylic acids through the two directing groups is reported. Useful structures with diverse substituents were efficiently synthesized in one step with the cleavage of four bonds (CH, CS, OH) and the formation of two bonds (CC, CO). The formed structure is the privileged core in natural products and bioactive molecules. This work highlights the power of using two different directing groups to enhance the selectivity of a double CH activation, the first of such examples in cross-oxidative coupling.

Abstract:A novel rhodium-catalyzed highly selective N-2-alkylation of benzotriazoles with diazo compounds/enynones is achieved, providing N-2-alkylated benzotriazoles in good to excellent yields and with excellent N-2 selectivities. Importantly, different to traditional carbene insertion into X - H (X = N, O etc) bonds, DFT calculations disclose that this selective N-2-alkylation probably proceeds through a formal 1,3- rather than 1,2-H shift to give the final products.

Abstract:Catalytic stereoselective desymmetrization of prochiral compounds to valuable chiral molecules is of interest in asymmetric synthesis. Here, we report the development of an efficient biocatalytic desymmetrization strategy for the stereodivergent synthesis of various chiral epoxides and oxazolidinones by the identification and engineering of halohydrin dehalogenases. The use of stereocomplementary halohydrin dehalogenases for the desymmetrization of 2-substituted-1,3-dichloro-2-propanols generates chiral epoxides in up to 95% yield and 99% ee by a single-step dehalogenation reaction, as well as chiral oxazolidinones in up to 88% yield and >99% ee via a dehalogenation and cyanate-mediated epoxide ring-opening cascade reaction. We also propose a probable mechanism for stereoselectivity control in the halohydrin dehalogenase-catalyzed desymmetrization reaction.

Abstract:An organocatalyzed asymmetric dearomative [3+2] annulation reaction of electron-deficient 2-nitrobenzo heteroarenes including 2-nitrobenzofurans, 2-nitrobenzothiophenes and 2-nitroindoles with 3-isothiocyanato oxindoles was developed. With a multiple hydrogen-bonding bifunctional thiourea as the catalyst, the reaction displays broad substrate scope and provides an access to structurally diverse polycyclic spirooxindoles containing three contiguous stereogenic centers in high yields with good diastereo- and enantioselectivities (up to 99% yield, 94:6 dr, and 97% ee). This organocatalysis protocol is able to afford the products with complementarily diastereoselectivity versus the previous work with transition metal catalysis.

Abstract:Chemodivergent reactions of oximes and diazo esters involving Rh-catalyzed [3+2] annulation and photodriven O-H insertion have been developed to generate oxazoles and oxime ethers. A range of aldehyde and ketone oximes reacted with α-diazocarbonyl compounds in a controllable manner in which functional groups, including ketone, ester, amide, ether, thiol ether, silane, alkene, allene, and alkyne groups, were well tolerated.

Abstract:Dual photoredox–transition-metal catalysis has emerged as a powerful tool for the development of chemical transformations. However, its application in the selective difunctionalization of 1,3-dienes remains essentially unexploited. By the synergistic action of photoredox and copper catalysts, we report herein an enantioselective 1,2-amidocyanation of 1,3-dienes. Visible-light irradiation of a chloroform solution of conjugated dienes, N-Boc-amidopyridinium salts, and TMSCN in the presence of a catalytic amount of fac-Ir(ppy)3, Cu(OTf)2·xH2O, and a chiral Box ligand affords three-component adducts in good to high yields with high regio- and enantioselectivities. A L*CuCN complex, fully characterized spectroscopically and confirmed by X-ray crystallographic analysis, is able to catalyze the transformation, therefore supporting an inner-sphere cyanide transfer pathway.
