Evaluation of Nitrogen Source, Concentration and Feeding Mode for Co-Production of Fucoxanthin and Fatty Acids in Phaeodactylum Tricornutum

Abstract:Microalgae-based wastewater treatment exhibits a competitive alternative to traditional biological techniques due to its ability to recover nutrients and produce high value-added bioproducts such as fucoxanthin and fatty acids. As an essential nutrient element, nitrogen plays a critical role in microalgal metabolism and bio-compositions synthesis including fucoxanthin and fatty acids, whose preference for nitrogen varied. However, it lacks information on whether it is feasible to achieve a suitable nitrogen-related culture condition that is preferred by the productions of both fucoxanthin and fatty acids, and simultaneously enhances microalgal growth performance and nutrient removal efficiency. This study investigated the effects of nitrogen sources, nitrogen levels and nitrogen feeding modes on microalgal biomass, nitrogen removal, and the microalgal intracellular response as represented by the co-production of fucoxanthin and fatty acids in Phaeodactylum tricornutum. With the same overall nitrogen concentration, the urea supplementation achieved over 90% nitrogen removal efficiency and compared with the other nitrogen sources including mixed ones, it also showed enhanced microalgal growth and fatty acid accumulation with the production of fucoxanthin 2.35-3.36 times higher than the other groups. Subsequently, a significantly higher accumulation of fucoxanthin (1.47 mg/g) and fatty acids (152.65 mg/g) were observed when 12.35 mg-N/L urea was added, which was the same nitrogen level included in the f/2 medium. Accordingly, with the 12.35 mg-N/L urea in total, one-time feeding on D0 and two-time feedings (fed equally on D0 and D5) were superior for the co-production of fucoxanthin and fatty acids, with the latter one also exhibiting a slightly enhanced nitrogen removal efficiency. This study systematically explored the effects of nitrogen on the co-production of fucoxanthin and fatty acid and provides a novel comparative idea for optimizing the nitrogen-related parameters for microalgal cultivation.
