Hydrogel Encapsulated Ph-Responsive Berberine@NMOF Nanoparticles for Accelerating Diabetic Wound Healing Via Multimodal Therapy

Abstract:Treatment options for chronic non-self-healing diabetic wounds are limited. The strategies of eliminating excess reactive oxygen species (ROS) or suppressing inflammatory responses on the wound bed is proved to be effective for diabetic wound healing. In this work, a zinc-based nano metal-organic framework (NMOF) was acted as a carrier for loading a natural product of berberine (BR) to form BR@Zn-BTB nanoparticles, and a kind hydrogel with ROS scavenging ability was used to encapsulate the nanoparticles to prepare an innovative hydrogel composite system BR@Zn-BTB/Gel (denoted as BZ-Gel). The results showed that BZ-Gel can be controlled to release Zn 2+ and BR in simulated physiological mediums, and efficiently eliminate ROS, inhibit inflammation and possess an excellent antibacterial effect. The fabricated hydrogels could not only reduce the skin contact toxicity of BR, but also promote the proliferation and migration of skin tissue cells. In vivo experiments further proved that BZ-Gel significantly inhibit the inflammatory response and enhance collagen deposition, as well as re-epithelialization of the skin wound, enhanced collagen and ultimately promoted wound healing in diabetic mice. These results indicate that the ROS-responsive hydrogel with BR@Zn-BTB incorporated synergistically promote diabetic wound healing, providing a new strategy for the development of the chronic wound dressings.
