A Ratiometric Fluorescent Probe Activated by Neutrophil Elastase As a Novel Strategy for Early Tumor Diagnosis

Abstract:The expression level of neutrophil elastase (NE) in vivo is closely related to the occurrence of inflammation. This enzyme has been identified as a research target for the development of anti-inflammatory drugs and for the early diagnosis and treatment of many diseases. This work developed the first near-infrared ratiometric fluorescence sensor ( TMNF ) for monitoring and imaging NE i n vivo and in vitro . When TMNF interacts with NE, the fluorescence changes from yellow to bright red with a Stokes shift of 115 nm. The response between TMNF and NE was fast (18 min) and sensitive (DL 32 ng/mL). In addition, histopathological experiments also proved that the probe has good biocompatibility and low cytotoxicity. Further cell imaging results showed that NE was highly expressed in cells treated with pro-inflammatory factors, whereas NE activity was significantly reduced after pretreatment with the enzyme inhibitor. It is worth mentioning that, for the first time, real-time dynamic monitoring and imaging of NE activity in inflammatory mice and tumor mice was performed. Relevant experiments have shown that inflammation can lead to increased expression of NE in vivo, and the activity of this enzyme in tumor mice is much higher than that in normal mice. The imaging of the probe in inflammation and tumor mouse models shows excellent practicality, making the probe an effective tool for the early diagnosis and treatment of diseases and tumors related to inflammatory lesions.
