Density Functional Theory Investigation on Aqueous Degradation Mechanism of Norfloxacin Initiated by Hydroxyl Radical

Abstract:Aqueous degradation mechanism of norfloxacin (NOR) initiated by hydroxyl radical (•OH) was investigated employing density functional theory (DFT) calculation. The most favorable degradation pathways were determined based on the obtained Gibbs free energy profiles, and the relative reactivity of cationic, neutral, and anionic forms of NOR (NOR(C), NOR(N), and NOR(A)) as the substrate was analyzed. The obtained results revealed that both H atom abstraction (HAA) by •OH and •OH addition (ADD) reactions play important roles in the degradation processes of NOR. Two HAA pathways involving axial C − H bonds on piperazine ring are more advantageous than other five initial HAA reactions generally, while ADD pathways involving C2, C3, C6, and C8 positions are kinetically more favorable than those related to C4, C5, and C7 atoms. The order of reactivity of the three existing forms of NOR in the reactions with •OH from low to high is as follows: NOR(C) < NOR(N) < NOR(A). The subsequent reactions of the most favorable HAA intermediates can lead to the destruction of piperazine ring, and the succeeding processes of the ADD intermediates involving C2 and C3 sites may result in the transformation of quinolone ring, decarboxylation, hydroxylation, and substitution of carboxyl by hydroxyl, while the further defluorination step of the ADD intermediate involving C6 atom occur hardly at ground state. Dissolved oxygen has an important promoting effect on aqueous NOR degradation.
