Abstract:Based on high performance liquid chromatography (HPLC), a fast, efficient and simple route for Smiles rearrangement reaction has been developed in the presence of 2-pyrimidinyloxy-N-arylbenzylamine derivatives as reactants with HC1 as acid catalyst in mixed CH3OH/H2O medium at room temperature. The advantage of the proposed method is that the products with excellent yields and high purity can be obtained employing reused solvent without further separating process. Considering the cheapness of catalyst and solvent, as well as high efficient and environment-friendly reaction condition, the developed method has the practicability and environmentally friendliness.
Kinetics of Acid-Catalyzed Smiles Rearrangement of 2, 6-Dimethoxy-2-pyrimidinyloxy-n-arylbenzylamine Derivatives

Abstract:The kinetics of the acid-catalyzed Smiles rearrangement reactions of 2,6-dimethoxy-2-pyrimidinyloxyN-arylbenzylamine derivatives was investigated. The effects of initial concentrations of hydrochloric acid, solvent, temperature, and substituent on reaction rates were examined. The results show that the rates increase with an increase in the initial concentration of hydrochloric acid. The reactivity order is CH3OH > C2H5OH > CH3SOCH3 > CH3CN in a single solvent, but rates markedly increase in mixed CH3OH/H2O (1 : 1, V/V) and the apparent reaction rate constant (k(obs)) is 5.27 times that of methanol. The rates for the derivatives are found to increase with an increase in temperature at 25-45 degrees C, and no significant differences in activation energy (73.99-76.92 kJ.mol(-1)) activation enthalpy (71.57-74.38 kJ.mol(-1)), and Gibbs free energy (81.51-85.77 kJ.mol(-1)) are observed between them, except that there is difference in activation entropy (-24.38-47.11 J.K-1.mol(-1)) There is a good linear relationship between substituents and the apparent reaction rate constants, and it is speculated that electron-withdrawing groups in the benzene ring will increase the reaction rates. A relevant reaction mechanism is suggested.

Abstract:The paper describes a convenient and facile methodology for the regioselective synthesis of fused oxazepinone scaffolds. This process is an efficient construction of the oxazepinone scaffold by a one-pot coupling/Smiles rearrangement/cyclization approach. This transition metal-free process has potential applications in the synthesis of biologically and medicinally relevant compounds.

Abstract:The one-pot sequential reaction of arylamines, methyl propiolate and 2-aryl-3-nitrochromenes without any catalyst in refluxing ethanol afforded the polysubstituted 3-(3-nitro-2-phenylchroman-4-yl)-3-arylaminoacrylates in good yields and with high diastereoselectivity. Reaction mechanism was believed involving the initial formation of β-enamino ester and sequential Michael addition.

Abstract:Asymmetric assembly of atropisomeric anilines from abundant and readily available precursors is one of the most challenging but valuable processes in organic synthesis. The use of highly efficient Smiles rearrangement to accomplish switchable enantioselective amination reactions of O-arenes provides access to nonsymmetric 2'-amino[1,1'-binaphthalen]-2-ol (i.e., NOBIN-type) and [1,1'-binaphthalene]-2,2'-diamine (i.e., BINAM-type) derivatives. This transition metal-free strategy provides a powerful way to access a wide range of advanced highly functionalized enantioenriched anilines.

Abstract:A practical synthetic method has been developed for polyfunctionalized biaryls based on a facile one-pot nucleophilic aromatic substitution (SNAr) reaction and [5,5]- or [3,3]-sigmatropic rearrangement reaction cascade. Under mild basic conditions, N-arylhydroxylamines reacted with o-activated fluoro (het)arenes to form N,O-diarylhydroxylamine intermediates which underwent spontaneously selective [5,5]-sigmatropic rearrangement reaction to produce diverse functionalized 4-amino-4′-hydroxy-1,1′-biaryls. A sequential SNAr reaction and [3,3]-sigmatropic rearrangement took place between N-arylhydroxylamines and 2-fluoropyridine derivatives or 4-fluorobenzonitrile to afford functionalized 2-amino-2′-hydroxy-1,1′-biaryls. As invaluable and unique building blocks, the resulting biaryls were applied in the straightforward synthesis of N2,O2-coronarene, carbazole, aza- and diaza carbazole derivatives.

Abstract:An efficient Cu0-promoted Truce-Smiles rearrangement for the aryl-difluoromethylenation of C═C bonds by the reaction of N-alkyl-N-(arylsulfonyl)methacrylamide and 2-bromodifluoromethyl-1,3-benzodiazole via a reductive radical-polar crossover process under mild reaction conditions is presented. The protocol enables practical access to a variety of single regioisomer α-aryl-β-difluoromethylene amides in good to excellent yields through consecutive difluoromethylenation, radical-polar crossover, 1,4-aryl migration, SO2 extrusion, and N-H bond formation cascade reaction.

Abstract:Based on the successful application of a nitroaryl transfer cascade reaction in the total synthesis of various monoterpene indole alkaloids proceeding via fused bicyclic intermediate, we investigated analogous cascade reactions involving spiro, bridged, and alternatively fused bicyclic intermediates. Unfortunately, none of these was found to afford the desired nitroaryl transfer product. DFT studies of the original cascade process revealed that the Truce‐Smiles rearrangement is the rate‐determining step. In addition, we found that the subsequent SO2 extrusion is accompanied by retro‐Mannich ring opening, resulting in a stabilized enolate that only recyclizes after activation of the cyclic imine moiety. Computation of the reaction profiles of the proposed alternative cascade reactions showed that the barrier for the Truce‐Smiles rearrangement is unreasonably high for the bridged and alternatively fused systems, but only moderately higher for the five‐membered spiro system. Reasoning that even more electron‐deficient arenesulfonamides should have a lower barrier for the Truce‐Smiles rearrangement, we synthesized the corresponding 2,4‐dinitrobenzenesulfonamide precursor and found that it indeed smoothly undergoes the nitroaryl transfer cascade at room temperature. In this case, however, the cascade reaction produces a cyclic imine product, as the intermediate 2,4‐dinitrophenyl‐substituted enolate is insufficiently nucleophilic to undergo the Mannich cyclization.

Abstract:An efficient pyridine-catalyzed chemoselective four-component cascade reaction of aromatic aldehydes, malononitrile/cyanoacetates, Morita-Baylis-Hillman (MBH) carbonates, and alcohols has been established. This one-pot reaction progressed in an unusual reaction with solvent participation via a Knoevenagel condensation/oxa-Michael addition/SN2' substitution sequence. This method allowed for facile access to an array of functionalized chain alkylbenzenes and dihydroquinolinones bearing one all-carbon quaternary center in moderate to excellent yields. It is worth noting that the configuration of the all-carbon quaternary center could be modulated by changing only the electron-withdrawing groups via a tandem reduction/cyclization reaction.

Abstract:We describe here an organocatalytic asymmetric cascade formal [3 + 3] cycloaddition of benzothiazoles with 2-nitroallylic acetates and nitroenynes. This dearomative methodology provided a facile and efficient strategy for the construction of a broad range of valuable benzothiazolopyridines bearing two adjacent stereogenic centers in moderate to good yields with good to excellent stereocontrol.

Abstract:A highly efficient asymmetric [1,3]- and [3,3]-O-to-C sigmatropic rearrangement of 3-allyloxy-4 H -chromenones and their analogues was developed. Chiral N , N ′-dioxide complexes of 3d late transition metal complexes enabled two mechanistically different processes, giving a series of optically active 2,2-disubstituted chromane-3,4-diones and 2-allyl-3-hydroxy-4 H -chromen-4-ones as well as their related compounds in excellent yield and enantioselectivity. Systemic mechanistic studies and DFT calculation revealed the nature of the vinyl ether unit of the substrate, which biased regioselectivity via a stepwise tight ion pair pathway and a concerted pericyclic pathway, respectively. The enantioselectivity of the two processes is also disclosed.

Abstract:N,O-Diarylhydroxylamines generally favor the [3,3] sigmatropic shift rearrangement. Possible N/O[1,3] sigmatropic shift rearrangements of multisubstituted N,O-diarylhydroxylamines were investigated experimentally with rationally designed substrates, which were generally in situ prepared from suitable nitroaryl halides and N-arylhydroxylamines via aromatic nucleophilic substitution. The results indicate that both N- and O-(2,4,6-trimethylphenyl)hydroxylamines still favor the [3,3] sigmatropic shift followed by tautomerization rather than N[1,3] and O[1,3] sigmatropic shifts and the rearranged products of N-(2,4,6-trimethylphenyl)hydroxylamines further undergo an intramolecular nucleophilic addition to afford dibenzo[b,d]furan-4a(9bH)-amine derivatives, while N-(4-mono- and 3,5-disubstituted phenyl)-O-(2,4,6-trinitrophenyl)hydroxylamines favorably first undergo the O[1,3] sigmatropic shift followed by tandem Smiles rearrangement and amide/ester exchange reactions, generating 2-arylaminoaryl benzoate derivatives. N-Phenyl-O-(2,4,6-trinitrophenyl)hydroxylamines undergo tandem double O[1,3] sigmatropic shift rearrangement to produce formal O[1,5] shift products. However, O-(2,6-dinitrophenyl)-N-(4-substituted phenyl)hydroxylamines undergo tandem O[1,3] and double [3,3] sigmatropic shift rearrangements to give formal 3,5-shift products. The proposed mechanism is rationalized by density functional theory (DFT) calculations. The current investigation provides not only a comprehensive understanding of the chemoselective sigmatropic shift rearrangements of N,O-diarylhydroxylamines, but also some novel synthetic strategies for dibenzo[b,d]furanamines, diarylamines, diaryl ethers, 2′-amino-[1,1′-biphenyl]-2(1H)-one, and 2′-amino-[1,1′-biaryl]-4-ol derivatives.

Abstract:A Pd-0-catalyzed four-component cascade reaction of an aryl halide, CO, an N-tosylhydrazone, and an amine affording alpha-amino ketone has been developed. This reaction involves a sequential carbonylation, metal carbene migratory insertion, and amination. Control experiments and DFT calculations further reveal the reaction sequence and chemoselectivity of individual components in this cascade process.

Abstract:A conceptually new and synthetically valuable radical Smiles rearrangement reaction is reported under undivided electrolytic conditions. This protocol employs an entirely new strategy for the electrochemical radical Smiles rearrangement. Remarkably, an amidyl radical generated from the cleavage of the N-O bond under reductive electrolytic conditions plays a crucial role in this transformation. Various hydroxylamine derivatives bearing different substituents are suitable in this electrochemical transformation, furnishing the corresponding amides in up to 86% yield.

Abstract:We describe novel amine‐mediated transformation of alkynyl ketones and amides to generate 2‐methylene‐2H‐pyrans, substituted 3‐hydroxy‐9H‐fluoren‐9‐ones, and amine‐incorporated arenes. These cascade processes are initiated by conjugate addition of secondary amine followed by hydrolysis of the enamine/vinylogous amide intermediates. The product distribution is highly sensitive to the steric and electronic effects of the substituents on both the alkyne moieties, the tether structure connecting them, and the nature of the amine. Alkynyl amide participates in the Alder‐ene reaction favorably to generate more reactive allene amide that reacts with amine to generate amine‐incorporated arene products. These metal‐free cascade reactions are a useful synthetic method that can be exploited for the construction of various hetero‐ and carbocyclic systems.

Abstract:The Brønsted base promoted (3 + 1) annulation reaction of C,N-cyclic azomethine imines and 3-chlorooxindoles was investigated, finally delivering complex hexahydroindeno[2,1-c] pyrazoles incorporating a spirocyclic oxindole motif after an unexpected rearrangement process. The asymmetric version of this new transformation could be accomplished, but slow racemization of the chiral product was observed at ambient temperature. Experiments and DFT calculations were further conducted to elucidate the reaction process and racemization mechanism.
