A New Insight into Revealing Food Sources Contribution of Seagrass Meadow Food Web by Using a Combination of Multiple Stable Isotopes and Fatty Acid Signatures

Abstract:Identifying the trophic role of each primary producer is the basis of assessing seagrass meadow functions but remains difficult due to the underdetermined analysis method. Carbon and nitrogen stable analysis are one of the most commonly used approaches to examine food source contribution, while the results of this approach are highly uncertain since the isotopes of food sources may be similar. The use of sulfur stable isotope and fatty acids may offer extra help that can resolve the uncertainty. Here, we analyzed the variations of multiple stable isotopes (δ 13 C, δ 15 N and δ 34 S) and fatty acids of food sources and macrobenthos in a tropical seagrass meadow in summer and winter, and tried a new method with the combination of multiple stable isotopes and fatty acids to resolve the limitation in the study of food source contribution in seagrass-based food webs. We found that the δ 13 C and δ 15 N values of epiphytes were similar to that of seagrass and macroalgae, while the δ 34 S values of epiphytes and macroalgae were significantly different, and the dominant poly-unsaturated fatty acids of seagrass (18:2n6c and 18:3n3) and epiphytes (16:1n7) were obviously different. Therefore, the combination of multiple stable isotopes and fatty acids can effectively distinguish the complex food sources in seagrass meadows. In addition, we also found that this combination is essential in identifying the diets of different macrobenthos sine there were some mislead when only isotopes or fatty acids were used (e.g., the wide range of isotopes for omnivores result in ambiguous food sources identification; fatty acid 18:2n6c may be converted into 20:5n3 and 22:6n3 during snails and polychaetes assimilation).
