Abstract:In recent years, the synthesis of axially chiral compounds has received considerable attention due to their extensive application as biologically active compounds in medicinal chemistry and as chiral ligands in asymmetric catalysis. Chiral phosphoric acids are recognized as efficient organocatalysts for a variety of enantioselective transformations. In this review, we summarize the recent development of chiral phosphoric acid-catalyzed synthesis of a wide range of axially chiral biaryls, heterobiaryls, vinylarenes, N-arylamines, spiranes, and allenes with high efficiency and excellent stereoselectivity.

Abstract:This review discusses the progress achieved in chiral phosphoric acid (CPA)-catalyzed organic reactions. CPA has demonstrated great versatility as a catalyst, effectively contributing to various reactions like cycloadditions, Friedel-Crafts reactions, dearomatizations, conjugate additions, Pictet-Spengler reactions, tandem reactions, and other asymmetric transformations. Acting as both a chiral Brønsted acid and a Lewis base, CPA facilitates the synthesis of enantioselective organic products that hold importance in biological and pharmaceutical chemistry. This advancement in CPA-catalyzed reactions has sparked further research interest. Although several chemists have published review articles on the advancements in chiral phosphoric acid catalysis until 2019, the recent surge of research necessitates an updated summary of the latest breakthroughs. This review is organized based on the specific types of reactions involved, aiming to provide valuable insights to the scientific community and highlight significant accomplishments in chiral phosphoric acid catalysis.

Abstract:Chiral spirocyclic phosphoric acids (SPAs) are introduced in 2010 and have been versatile catalysts capable of promoting a wide range of asymmetric organocatalytic reactions, such as multi-component reactions, Friedel-Crafts reactions, Pictet-Spengler reactions, Fischer indolizations, cycloaddition reactions, desymmetrization reactions, dearomatization reactions, conjugate addition reactions and rearrangement reactions, etc. Moreover, a diverse range of applications in metal-organic cooperative catalysis, organic photoredox catalysis, total synthesis, materials science and molecular recognition are beautifully illustrated. Abstract:Kinetic resolution, dynamic kinetic resolution and desymmetrization are practical methods to produce enantioenriched products from relatively simple racemic or achiral starting materials, which are also sometimes difficult to be accessed by other asymmetric methods. Chiral phosphoric acids, which have been recognized as one of the most powerful chiral organocatalysts to date, have also been extensively applied in the above‐mentioned research fields. In this minireview, we summarized the tremendous development of kinetic resolution, dynamic kinetic resolution and desymmetrization reactions enabled by chiral phosphoric acids since the year of 2016, which were not included in the two previous outstanding reviews by Petersen et al and Dixon et al. A number of examples involving the use of CPA as cooperative catalyst are also included in this review.

Abstract:Asymmetric cycloaddition/annulation reactions have contributed greatly to the synthesis of a variety of novel cyclic compounds used in organic synthesis, natural product synthesis, material science, and chemical biology. Thus, chiral phosphoric acid-catalyzed asymmetric cycloaddition reactions have received the attention of the synthetic community to design new vital asymmetric reactions and synthesize complex molecules for the past two decades. Although several review articles on organocatalyzed asymmetric cycloaddition reactions can be found in the literature, there has been no specific review on CPA-catalyzed asymmetric cycloaddition/annulation reactions. Herein, the most recent research progress on chiral phosphoric acid catalyzed asymmetric [3+2], [4+2], [2+2], [3+3], [4+3], [4+1] annulation, and 1,3-dipolar cycloadditions have been compiled and analyzed to demonstrate the importance of chiral phosphoric acid catalysis in asymmetric cycloaddition or annulation reactions.

Abstract:Chiral allenes and styrenes are essential components in fields like medicinal chemistry, materials science, and organic synthesis. They serve a crucial role as chiral ligands and catalysts in asymmetric synthesis. Over the past decade, there has been a significant advancement in the development of practical methods utilizing organocatalytic strategies for the synthesis of chiral allenes and styrenes. It is noteworthy that despite extensive studies on the formation of allenes and styrenes, existing reviews on their construction confined to a specific organocatalysis, called chiral phosphoric acid catalysis, are less documented. This review aims to explore different conceptual approaches based on the electrophilic species involved in the reaction to produce stereoselective chiral allenes and styrenes, providing insights into recent advancements in the field. Emphasis is placed on works published since 2017, with detailed discussions on reaction mechanisms and examples from recent literature.

Abstract:Chiral benzothiazepines constitute the core structures of many foremost pharmaceuticals with diverse biological activities endowed by their unique scaffolds, which poses a great challenge to organic chemists and pharmaceutical researchers. This review provides a concise overview for the asymmetric synthesis of chiral benzothiazepine derivatives, focusing on advances in asymmetric catalysis, including metal catalysis, small-molecule organocatalysis and enzymatic catalysis. The catalytic asymmetric reactions, involving asymmetric epoxidation, reduction, dihydroxylation, hydrogenation, aldol reaction and other sulfa-Michael addition, have emerged as powerful strategies for the rapid construction of chiral benzothiazepine through single or multistep reactions. The booming asymmetric synthetic methodology affords us instructive clues for the highly efficient preparation of chiral benzothiazepines, facilitating their large-scale preparation and diversity-oriented synthesis.

Abstract:A new type of robust planar chiral phosphoric acid bearing a [2.2]paracyclophane scaffold was synthesized and shown to be an optimal catalyst in asymmetric aza-Friedel-Crafts reactions for the first synthesis of enantioenriched styryl indolylmethanamine derivatives in good yields with excellent enantioselectivities (93->99% ee) under 0.5-1 mol % catalyst loading.

Abstract:Reported here is the development of a class of chiral spirosilabiindane scaffolds by Rh-catalyzed asymmetric double hydrosilation, for the first time. Enantiopure SPSiOL (spirosilabiindane diol), a new type of chiral building block for the preparation of various chiral ligands and catalysts, was readily prepared on greater than 10 gram scale using this protocol. The potential of this new spirosilabiindane scaffold in asymmetric catalysis was preliminarily demonstrated by development of the corresponding monodentate phosphoramidite ligands (SPSiPhos), which were used in both a Rh-catalyzed hydrogenation and a Pd-catalyzed intramolecular carboamination.

Abstract:In the last few years, the research area of chiral β-aminophosphines capable of promoting a wide range of diverse organic transformations has attracted more attention. Their derivatives constitute attractive reagents towards this end, due to their stability, low toxicity and ease of handling. These novel β-aminophosphine derivatives are potentially useful as organocatalysts and ligands in metal-catalysed reactions. In particular, chiral β-aminophosphines have recently emerged as powerful phosphine catalysts in asymmetric synthesis and catalysis. The asymmetric transformations to which metal complexes of these ligands have been applied include palladium-catalysed allylic substitutions and copper-catalysed 1,4-additions to enones among others. This review summarizes the most significant developments in the area of synthesis and application of chiral β-aminophosphine derivatives, providing a detailed overview of the current state of the art and including future aspects.

Abstract:An enantioselective synthesis of unnatural pyrazole-based alpha-chiral amino acid derivatives from the asymmetric reaction of N-aryl-5-aminopyrazoles with beta,gamma-alkynyl-alpha-imino esters using a chiral spirocyclic phosphoric acid catalyst was developed. Using the established methodology, various pyrazole-based alpha-amino acid derivatives with tetrasubstituted carbon stereocenters were obtained in 67-98% yields and with 73-99% enantioselectivities. The NH2 functionality in the corresponding products enables further transformations to a chiral thiourea and a lactam.

Abstract:Chiral amino acids featuring a phosphonate pendant arm are an important type of biologically active scaffolds. Here in this review, we comprehensively summarize the modern synthetic methods towards asymmetric construction of chiral amino carboxylicphosphonic acid derivatives. The main streams of such interesting compounds include phosphono‐containing α‐, β‐, and γ‐amino acid derivatives, amino acid fluorophosphonate derivatives, amino acid cyclopropanylphosphonate derivatives, and bisphosphono‐amino acid derivatives. Chiral resolution protocols, chiral auxiliary‐directed syntheses, and valorization of the pool of abundant chiral amino acid resources remain contemporary concerns, meanwhile catalytic enantioselective synthesis has also emerged as a potent strategy as demonstrated by the latest advances.

Abstract:A series of chiral spiroketal bisphosphine ligands containing 1,1'-spirobi(3 H,3' H)isobenzofuran backbones was accessed through asymmetric synthesis and subsequently tested in enantioselective Rh-catalyzed hydrogenation of α-dehydroamino acid esters. The ligand providing the highest enantioselectivity (up to 99.5%) was obtained in seven steps in an overall 38% yield. The synthesis could be performed on a gram scale, and no kinetic resolution of enantiomers is required. Overall, the developed ligand provides an easily accessible alternative to SDP ligands as well as other chiral bisphosphine ligands.

Abstract:Catalytic enantioselective synthesis of chiral helicenes and helicenoids has emerged as an important area of research. In the efforts toward efficient and enantioselective synthesis of chiral helicenes and helicenoids, chemists have developed several catalytic asymmetric reactions, primarily transition metal-catalyzed asymmetric intra- or intermolecular [2 + 2 + 2] cycloaddition, hydroarylation, olefin metathesis, and oxidative coupling reactions. In addition, asymmetric organocatalysis has shown potential for use in the enantioselective synthesis of chiral helicenes and helicenoids, including chiral phosphoric acid- and chiral amide-catalyzed enantioselective [3 + 2] and [4 + 2] cyclizations. In this review, advancements in the catalytic enantioselective synthesis of chiral helicenes and helicenoids from 1999 to the beginning of 2022 have been classified based on asymmetric transition metal catalysis and asymmetric organocatalysis. In addition, this review highlights the remaining challenges in this research field and sheds light on the future of this field.

Abstract:As an excellent representative of organic small molecular catalysts, chiral phosphoric acid played an important role in asymmetric catalytic hydrogenation. According to the skeleton of chiral phosphoric acid, the development and applications of chiral phosphoric acid were summarized. The applications of chiral phosphoric acid in asymmetric catalytic hydrogenation were introduced according to three kinds of hydrogen source systems used in chiral phosphoric acid catalytic reaction. It was found that chiral phosphoric acid has high activity and excellent selectivity. Chiral products with excellent yields and enantioselectivities were obtained in most reactions.

Abstract:Although there have been various reviews on the asymmetric construction of C3-spirooxindoles, there is a scarcity of reviews focusing on the asymmetric organocatalytic synthesis of C3-spiro-cyclopentaneoxindole derivatives. This particular scaffold has garnered significant attention from synthetic chemists due to its relevance in medicinal chemistry. In this review, we provide an overview of recent advancements in the asymmetric organocatalytic synthesis of various C3-spiro-cyclopentaneoxindoles using organic catalysts. The work is divided into sections according to the type of catalysis, including covalent catalysis (amine catalysis and N-Heterocyclic carbenes catalysis) and non-covalent catalysis (chiral phosphoric acid catalysis, hydrogen bond catalysis, and phase transfer catalysis). Furthermore, we discuss existing challenges and future directions within this field. It is our belief that this review will serve as an informative resource for researchers engaged in synthesizing C3-spiro-cyclopentaneoxindoles and inspire further advancements in this area.
