Abstract:The synthesis of N-heterocycles is one of the most active areas due to their structures with the extensive applications in the fields of organic, pharmaceutical and material chemistry. It is an increasing challenging task to develop greener, milder and cheaper N-heterocycles synthetic strategies through C–N/C–C bond formation owing to requiring multistep reactions /or harsh reaction conditions in many cases. Herein, this review focuses on the synthesis of N-containing heterocycles such as pyrimidines, imidazoles, 1,2,4-triazoles, 1,3,5-triazines, benzimidazoles, isoquinolines, quinazolines, quinazolinones, 1,2,4-oxadiazoles, thiazoles, and thiadiazoles using amidines as readily available starting materials, with an emphasis on the novel green synthetic methodologies via C–N/C–C bond formation, as well as the mechanisms of these reactions. We hope this review will help to give references for researchers who are fascinated with the increasing applications in this prolific area.

Abstract:Transition metal catalyzed C-H bond activation have become one of the important tools for constructing new chemical bonds. Introducing directing group to the substrates are the key to successful reaction, these directing groups can also be further transformed in the reaction. As amidines with unique structure and reactivity, makes them ideal substrates for transition metal-catalyzed C-H transformations. This review describes the major progress and mechanistic investigations on C-H activation/annulation tandem reaction of amidines until early 2024, focusing on metal-catalyzed C-H activation of amidines with unsaturated compounds, such as alkynes, olefins vinylene carbonate, cyclopropanols and their derivatives. Meanwhile this manuscript also with a view to the reaction of amidines with diffierent carbene precursor, for example diazo compounds, azide, triazoles, pyriodotriazoles, sulfoxonium ylides. And its own C-H bond activation/cyclization reaction. A bright outlook was provided at the end of the manuscript.

Abstract:Nitro-compounds are one of the cheapest and most readily available materials in the chemical industry and are commonly utilized as versatile building blocks. Previously, the synthesis of N-heterocycles was largely based on anilines. The utilization of nitroarenes and nitroalkenes for the synthesis of N-heterocyclic compounds can save at least one step, however, as compared to anilines. Thus, considerable attention has been paid to nitroarenes and nitroalkenes as new potential amino sources. Significant progress has been made in the reductive cyclization of nitroarenes or nitroalkenes to access various N-heterocycles in recent years. Herein, we comprehensively summarize the recent progress in the construction of N-heterocycles using nitroarenes and nitroalkenes as potential amino sources. The compatibility of the reaction substrate, its mechanism, applications, advantages, and limitations in this field are also discussed in detail.

Abstract:Metal-catalyzed cyclization reactions have become a powerful and efficient approach for the stereoselective construction of both carbocyclic and heterocyclic ring systems. Transition metal complexes, with their ability to activate and selectively functionalize organic substrates, have revolutionized various areas of synthetic chemistry. This review highlights recent advancements in metal-catalyzed cyclization reactions, especially in the synthesis of nitrogen-containing heterocycles like imidazoles, pyridines, pyrimidines, and indoles. These advancements have significantly impacted fields such as natural product synthesis, pharmaceuticals, functional materials, and organic electronics. Novel catalytic systems, ligand designs, and reaction conditions continue to expand the capabilities of these reactions, driving further the progress made in synthetic organic chemistry. This review provides a comprehensive overview of recent research.

Abstract:Multisubstituted guanidines have received much attention because of important applications in many fields, such as pharmaceutics, organometallic and coordination chemistry, and organic synthesis. Although classical methods are available for the preparation of guanidines, synthetic preparation approaches to guanidines are still in great demand. In this review, we summarize recent developments on synthetic methods via the C-N bond formation. Three aspects are included: transition-metal-catalyzed guanidine synthesis based on classical methods; catalytic guanylation reaction of amines with carbodiimides; and tandem catalytic guanylation/cyclization reactions.

Abstract:A cobalt(III)-catalyzed C-H/N-H bond functionalization for the synthesis of 1-aminoisoquinolines from aryl amidines and diazo compounds has been developed. The reaction proceeds under mild reaction conditions, obviates the need for oxidants, produces only N2 and H2O as the byproducts, and features a broad substrate scope.

Abstract:Nitrogen‐containing heterocycles have attracted considerable attention due to their extensive applications in the fields of organic synthesis, pharmaceuticals, agrochemicals, and materials science. This review is focused on the recent advances in the synthesis of nitrogen‐containing heterocycles via the cyclization of alkynyl imines. By using this protocol, a variety of nitrogen‐containing heterocycles, mainly including four‐, five‐, six‐, seven‐membered nitrogen‐containing heterocycles, aza‐spirocycles, fused aza‐heterocycles, can be obtained efficiently under mild conditions. Additionally, mechanistic features of these transformations are also discussed.

Abstract:The lack of effective strategies for direct construction of nitrogen-nitrogen bonds has hampered developments in the synthesis and application of molecules containing hydrazine or azo motifs. Attracted by their properties both in material science and in medicinal chemistry, more and more attention has been drawn to this area, resulting in fast growth in the design and synthesis of azaheterocycles and substituted hydrazines. This review focuses on efficient catalytic approaches toward the formation of N-N and N & boxH;N bonds through different strategies, including oxidative dehydrogenation, nitrene-transfer reaction, reductive coupling, and some other recently developed methods.

Abstract:The transformation of C-H bonds into other chemical bonds is of great significance in synthetic chemistry. C-H bond-activation processes provide a straightforward and atom-economic strategy for the construction of complex structures; as such, they have attracted widespread interest over the past decade. As a prevalent directing group in the field of C-H activation, the amide group not only offers excellent regiodirecting ability, but is also a potential C-N bond precursor. As a consequence, a variety of nitrogen-containing heterocycles have been obtained by using these reactions. This Focus Review addresses the recent research into the amide-directed tandem C-C/C-N bond-formation process through C-H activation. The large body of research in this field over the past three years has established it as one of the most-important topics in organic chemistry.

Abstract:The sustainable preparation of N‐heterocycles is one of the most active research areas owing to their predominance as synthetics building blocks with extensive applications in organic, pharmaceutical, and material chemistry fields. Among the various catalytic protocols, the C–H bond functionalization with the concomitant C–N bond formation, so‐called C–H bond annulation, has become one of the most sustainable routes to access N‐heterocycles because it starts from low‐functionalized materials and generates a limited amount of waste, all respecting the concept of atom economy. Rhodium complexes often catalyze these reactions. This review focuses on the synthesis of 5‐ and 6‐membered ring N‐containing heterocycles such as indoles, pyrroles, indolines, (iso)quinolinones, dihydroquinolines, and pyrrolidones from readily available starting materials, with an emphasis on the novel C–H bond cascade synthetic methodologies via C–N/C–C bond formation, as well as on the mechanisms of these reactions, especially the oxidation steps. We hope this review will help researchers looking to prepare N‐heterocycles in a minimum of steps and those who want to develop new methodologies based on C–H bond activation/functionalizations.

Abstract:A convenient synthetic approach for the synthesis of alpha-oxo-acetamidines via copper-catalyzed C(sp(3))-H amidination has been developed. This approach allows the direct amidination of three C(sp(3))-H bonds without cyclization.Methyl ketones and primary and secondary amines are tolerant and afford the corresponding products in moderate to good yields. Furthermore, this protocol is also applicable to the synthesis of unsymmetrical alpha-oxo-acetamidines via one-pot, multicomponent reactions (MCRs).

Abstract:The diverse methodologies to synthesize N-heterocycles through transition-metal-catalyzed cascade addition/cyclization of a nitrile are discussed in this review. Aspects relating to three types of transition-metal-catalyzed addition of a nitrile with subsequent cyclization include (1) a transition-metal acting as a Lewis acid to accelerate the nucleophilic addition of a nitrile, (2) the late-transition-metal-catalyzed 1,2-insertion of a nitrile, and (3) an in situ generated radical by transition-metal catalysis to implement a radical addition/cyclization tandem reaction. Abstract:A general and efficient copper-catalyzed aerobic oxidative method for the synthesis of N-fused heterocycles has been developed by using readily available α-amino acids as the nitrogen source. The reactions underwent N-arylation, aerobic oxidative dehydrogenation, intramolecular cyclization and dissociation of formic acid. Abstract:Developing versatile methodologies to construct various nitrogen-containing heterocycles is a crucially significant part of contemporary organic chemistry. This review summarizes recent developments on the formation of nitrogen-containing heterocycles triggered by diaryliodonium salts. Diaryliodonium salts, as electrophilic arylating agents in the presence of catalytic copper salts, can react with nitriles to give N-arylnitrilium cations, which are highly reactive species. These species can efficiently react with nucleophiles, including C-, N- and O-nucleophiles, to give the corresponding products. Abstract:Nitrogen-containing heterocycles (N-heterocycles) are ubiquitous in both organisms and pharmaceutical products. Biocatalysts are providing green approaches for synthesizing various N-heterocycles under mild reaction conditions. This review summarizes the recent advances in the biocatalysis of N-heterocycles through the discovery and engineering of natural N-heterocycle synthetic pathway, and the design of artificial synthetic routes, with an emphasis on biocatalysts applied in retrosynthetic design for preparing complex N-heterocycles. Furthermore, this review discusses the future prospects and challenges of biocatalysts involved in the synthesis of N-heterocycles.

Abstract:Organic azides are highly reactive, which have long established as versatile building blocks in the assembly of structurally diverse N‐containing heterocycles. The conversion of organic azides into high‐value compounds is of great importance and a subject of enormous current interest. Transition metal‐catalyzed C(sp2)–H amination/annulation of organic azides provides a powerful tool for the transformation of organic azides into a wide range of biologically important heterocyclic frameworks. In this review, we aim to summarize the recent progress on organic azides‐mediated C(sp2)–H amination/annulation for N‐heterocycle synthesis enabled by transition metal catalysts. Representative strategies are discussed in detail, including catalytic systems, reaction scopes, limitations and mechanisms.
