Abstract:Two to three-wwek-old mature seed-derived rice callus (Oryza sativa L., japonica cv. Eyi 105) was cotransformed via particle bombardment with two plasmids, pJIMB15 and pBISAPl, containing the selectable marker hygromycin phosphotransferase gene (hpt), the reporter beta-glucuronidase gene (gusA) and the ap1 gene. The ap1 gene encodes an amphipathic protein previously shown to delay the hypersensitive response induced in non-host plants by the pathogen Pseudomonas syringae pv. syringae (Pss). After two rounds of selection on hygromycin (30mg/L)-containing medium, resistant callus was transferred to hygromycin (30 mg/L)-containing regeneration medium for plant regenaration. Thirty-two independent transgenic plants were regenerated from 186 bombarded callused, with a transformation frequency of 17.2%. Eighty-four percent of the transgenic plants carried all three transgenes as shown by PCR/Southern blot analysis.
