Abstract:A useful transformation of nitriles to N-phenyl amides has been achieved through a novel intermolecular phenyl transfer reaction from phenyl hydrazides and N-addition to nitriles in the presence of PIFA under mild and solvent-free conditions. This cross coupling reaction includes the oxidative cleavage of sp(2) C-N bonds of phenyl hydrazides to form a phenyl radical and the subsequent N-addition to cyanos to form new sp(2) C-N bonds and provides efficient access to various N-phenyl amides in moderate to good yields under mild reaction conditions.

Abstract:A novel zinc-catalyzed hypervalent iodine mediated cascade transformation of anilines and aldehydes to benzimidazoles was developed. A series of 1,2-disubstituted benzimidazoles were achieved with aromatic, heteroaromatic, and aliphatic aldehydes. In this procedure, initial condensation of anilines with aldehydes afforded N-arylimine intermediates which underwent C-H/C-H amination/annulation to generate the desired products in moderate to excellent yields. The reaction features an extremely simple reaction system, and good functional group toleration with water as the only byproduct.

Abstract:A metal‐free, 4,5‐difluoro‐2‐iodobenzoic acid‐catalyzed C–H functionalization strategy is developed for the synthesis of 2‐(hetero)aryl benzothiazoles from aryl isothiocyanates and unfunctionalized arenes/thiophenes/furans at room temperature. The procedure entails a one‐pot, cascade carbon–carbon and carbon–sulfur bond formation facilitated by triflic acid, which serves as both a Bronsted acid and a ligand source at the in situ generated I(III) centre. This process is atom‐economical and offers benefits over existing protocols for synthesizing benzothiazoles in terms of ease of use, eco‐friendliness, and not requiring the typical use of aryl aldehyde/carboxylic acid precursors. Mechanistic studies were carried out, and the synthetic utility of the protocol was demonstrated for the synthesis of the benzothiazole‐based antitumor drug GW‐610.

Abstract:Imidazoles represent one of the most important heterocycles which are known to exhibit a wide range of biological and medical activity.Existing methodologies for the synthesis of imidazoles are limited to the generality and accessibility of substrates,and the harsh reaction conditions.The development of milder and more practical protocols should be still desirable and necessary.Here we report a new method for the synthesis of polysubstituted imidazoles.A series of imidazoles was obtained directly from 2-phenylacetaldehyde or acetophenone derivatives and benzylamine derivatives via a metal-free-catalyzed tandem oxidative cyclization.The reaction was performed under mild conditions with iodine as a catalyst and TBHP as an oxidant.The effect of catalyst loading,oxidants,solvents and temperature on this transformation was investigated.The optimal reaction conditions were as follows: 0.3 equiv.of iodine as the catalyst,TBHP as the oxidant,acetonitrile as the solvent and the reaction being carried out at 70 ℃.Electron-withdrawing group of acetophenone derivatives and benzylamine derivatives benefit the oxidative reaction.Compared with traditional methods,this reaction was conducted under milder conditions with facile starting materials.

Abstract:A facile method for the synthesis of 3-iodoimidazo[1,2- a]pyridines has been successfully developed involving an I2O5-mediated iodocyclization cascade of N-(1-arylallyl)pyridin-2-amines with concomitant C═C bond cleavage. Preliminary mechanistic studies reveal that this protocol might undergo an oxidative cyclization/decarboxylation/iodination sequence in which I2O5 is used as both an oxidant and an iodine source. The present protocol has advantages of wide substrate scope, simple operation, and metal-free conditions.

Abstract:This manuscript reports a novel method for the direct iodination of the C‐4 pyrazole ring generated in situ by the reaction of 1,1,3,3‐tetramethoxypropane and various hydrazines. In this approach, potassium iodate (KIO3) and (PhSe)2 are used as catalysts under acidic conditions. This protocol provides a convenient method for the efficient synthesis of 4‐iodo‐1‐aryl‐1H‐pyrazoles, valuable intermediates for several different coupling reactions.

Abstract:A novel and convenient one-step method for the synthesis of 1,3,4,-Triaryl-substituted-3,4,5,6-tetrahydropyrimidin-2(1H)-ones (3) was developed by cyclization of aromatic isocyanates (1) with 3-arylamino-1-phenylpropan-1-ones (2) under refluxing of toluene. Effects of reaction conditions on the converting ratio and yield were investigated. The method offered several advantages including simple experimental procedure, easily available starting materials and excellent yields (79%similar to 96%).

Abstract:We report herein a general and effective system achieving cyclization of β-trifluoromethyl enones with amidines in the presence of 1,3-diiodo-5,5-dimethylhydantoin (DIH), which affords a range of trifluoromethylated 2-imidazolines in synthetically useful yields with good diastereoselectivities (up to 95% yield, up to 98:2 dr) and good functional group tolerance. Furthermore, the one-pot synthesis of trifluoromethylated imidazoles via sequential cyclization and oxidation is demonstrated. More significantly, the reaction mechanism was verified by ESI-MS studies of possible intermediates, and a reasonable reaction mechanism was proposed.

Abstract:<p>In this paper, two novel and efficient methods for the synthesis of trisubstituted imidazoles have been developed through iodine-catalyzed [3+2] cycloaddition. By controlling one of the reaction materials, we selectively synthesized 1,2,4-trisubstituted imidazoles and 1,2,5-trisubstituted imidazoles. The desired products were obtained in moderate to good yields with wide functional groups compatibility under standard conditions.</p>

Abstract:A novel six-membered heterocyclic skeleton of imidazochromone was prepared via an efficient one-pot reaction including a key step of copper-catalyzed aerobic C-H intramolecular cycloetherification. Notably, this process does not require the presence of strong para electron-withdrawing groups on the phenol component. Also, the results of this effort show that acyl phenols containing electron-rich heterocycles participate in an efficient C-H activation/C-O formation process.

Abstract:In this paper, novel sulfur-containing 1,6-dihydrofuro[3,2-b]pyrazolo[3,4-e][1,4]thiazine skeletons were constructed from the simple and readily available materials enaminone, 5-aminopyrazole, and 1,4-dithiane-2,5-diol. Furthermore, a novel 1,4-dithiane-2,5-diol reaction mode has been developed through a double-dipole-reversal process induced by iodine that results in the formation of six new bonds and two new rings in a one-pot reaction. This method shows good substrate compatibility, and the products can be further modified with a variety of pharmaceuticals. Additionally, this novel skeleton exhibits good fluorescence properties in solution, enabling bright and stable green fluorescence imaging in HeLa cells.

Abstract:Reactions of easily accessible chloropyrimidinyl hydrazones with bromine were investigated. Oxidative cyclization followed by concomitant bromination led to the formation of [1,2,4]triazolo[4,3-c]pyrimidines, which then underwent the Dimroth rearrangement to afford highly functionalized 2-substituted [1,2,4]triazolo[1,5-c]pyrimidines.
