Abstract:There has been unprecedented progress in the development of biomedical nanotechnology and nanomaterials over the past few decades, and nanoparticle-based drug delivery systems (DDSs) have great potential for clinical applications. Among these, magnetic drug delivery systems (MDDSs) based on magnetic nanoparticles (MNPs) are attracting increasing attention owing to their favorable biocompatibility and excellent multifunctional loading capability. MDDSs primarily have a solid core of superparamagnetic maghemite (γ-Fe 2 O 3 ) or magnetite (Fe 3 O 4 ) nanoparticles ranging in size from 10 to 100 nm. Their surface can be functionalized by organic and/or inorganic modification. Further conjugation with targeting ligands, drug loading, and MNP assembly can provide complex magnetic delivery systems with improved targeting efficacy and reduced toxicity. Owing to their sensitive response to external magnetic fields, MNPs and their assemblies have been developed as novel smart delivery systems. In this review, we first summarize the basic physicochemical and magnetic properties of desirable MDDSs that fulfill the requirements for specific clinical applications. Secondly, we discuss the surface modifications and functionalization issues that arise when designing elaborate MDDSs for future clinical uses. Finally, we highlight recent progress in the design and fabrication of MNPs, magnetic assemblies, and magnetic microbubbles and liposomes as MDDSs for cancer diagnosis and therapy. Recently, researchers have focused on enhanced targeting efficacy and theranostics by applying step-by-step sequential treatment, and by magnetically modulating dosing regimens, which are the current challenges for clinical applications.

Abstract:Coupled magnetic nanoparticles in the microcapsule structure, such as magnetic microcapsules, can be delivered in specific organism or tissues under magnetic field exposure. Thus, the microcapsules can achieve active targeting functions by manipulat-ing the magnetic field. Based on the magnetic microcapsules, the antitumor drugs can also be loaded to realize magnetic response, which gives microcapsules sustained and controlled release advantages. To date, the drug microcapsules carrying magnetic nanoparti-cles have become promising novel delivery carriers for the treatment of tumor diseases. This paper mainly reviews the method of prepa-ration of the magnetic nanoparticle-coupled microcapsules, including liposomes, polyelectrolyte microcapsules, and polymer micro-spheres. The basic research progress of these microcapsules as anticancer drug carriers for the tumor therapy was also reviewed.

Abstract:This review summarizes the types of magnetic nanoparticles, microfluidic synthesis, and drug delivery, cell enrichment, bioimaging, and biosensing applications of microfluidic‐derived magnetic nanoparticles. In addition, the challenges and prospects in harnessing the full potential of these magnetic nanoparticles are also discussed to highlight the importance of ongoing research in this dynamic field. Magnetic nanoparticles have attracted great attention and become promising candidates in the biomedicine field due to their special physicochemical properties. They are generally divided into metallic and non‐metallic magnetic nanoparticles, according to their compositions. Both of the two types have shown practical values in biomedicine applications, such as drug delivery, biosensing, bioimaging, and so on. Research efforts are devoted to the improvement of synthesis strategies to achieve magnetic nanoparticles with controllable morphology, diverse composition, active surface, or multiple functions. Taking high repeatability, programmable operation, precise fluid control, and simple device into account, the microfluidics system can expand the production scale and develop magnetic nanoparticles with desired features. This review will first describe different classifications of promising magnetic nanoparticles, followed by the advancements in microfluidic synthesis and the latest biomedical applications of these magnetic nanoparticles. In addition, the challenges and prospects of magnetic nanoparticles in the biomedical field are also discussed.

Abstract:Magnetic nanoparticles (MNPs) are a special kind of nanomaterials and widely used in biomedical technology applications. Currently they are popularly customized for disease detection and treatment, particularly as drug carriers in drug targeted delivery systems, as a therapeutic in hyperthermia (treating tumors with heat), and as contrast agents in magnetic resonance imaging (MRI). Due to their biocompatibility and superparamagnetic properties, MNPs as next generation drug carriers have great attraction. Although the potential benefits of MNPs are considerable, any potential toxicity associated with these MNPs should be identified distinctly. The drug loading capability and the biomedical properties of MNPs generated by different surface coatings are the most sensitive parameters in toxicity. A lot of organic and inorganic materials are utilized as coating materials for surface functionalization and reducing toxicity of MNPs. pH or temperature sensitivity materials are widely used to manage drug loading and targeted release. In addition, MNPs can be controlled and directed to the desired pathological region by using external magnetic files (EMF). The realization of targeted drug delivery has decreased the dosage and improved the efficiency of drugs, which results in reduced side effects to normal tissues. This review discussed the possible organ toxicities of MNPs and their current advances as a drug delivery vehicle.

Abstract:INTRODUCTION: In the field of drug delivery, controlling the release of therapeutic substances at localized targets has become a primary focus of medical research, especially in the field of cancer treatment. Magnetic nanoparticles are one of the most promising drug carriers thanks to their biocompatibility and (super)paramagnetic properties. These properties allow for the combination between imaging modalities and specific release of drugs at target sites using either local stimulus (i.e. pH, conjugation of biomarkers, …) or external stimulus (i.e. external magnetic field).AREAS COVERED: This review provides an update on recent advances with the development of targeted drug delivery systems based on magnetic nanoparticles (MNPs). This overview focuses on active targeting strategies and systems combining both imaging and therapeutic modalities (i.e. theranostics). If most of the examples concern the particular case of cancer therapy, the possibility of using MNPs for other medical applications is also discussed.EXPERT OPINION: The development of clinically relevant drug delivery systems based on magnetic nanoparticles is driven by advantages stemming from their remarkable properties (i.e. easy preparation, facile chemical functionalization, biocompatibility, low toxicity, and superior magnetic responsiveness). This literature review shows that drug carriers based on magnetic nanoparticles can be efficiently used for the controlled release of drug at targeted locations mediated by various stimuli. Advances in the field should lead to the implementation of such systems into clinical trials, especially systems enabling drug tracking in the body.

Abstract:Magnetic drug targeting is a method by which magnetic drug carriers in the body are manipulated by external magnetic fields to reach the target area. This method is potentially promising in applications for treatment of diseases like cancers, nervous system diseases, sudden sensorineural hearing loss, and so on, due to the advantages in that it can improve efficacy, reduce drug dosage and side effects. Therefore, it has received extensive attention in recent years. Successful magnetic drug targeting requires a good magnet system to guide the drug carriers to the target site. Up to date there have been many efforts to design the magnet systems for targeted drug delivery. However, there are few comprehensive reviews on these systems. Here we review the progresses made in this field. We summarized the systems already developed or proposed, and categorized them into two groups: static field magnet systems and varying field magnet systems. Based on the requirements for more powerful targeting performance, the prospects and the future research directions in this field are anticipated.

Abstract:Magnetic nanoparticles (MNPs) have evolved tremendously during recent years, in part due to the rapid expansion of nanotechnology and to their active magnetic core with a high surface-to-volume ratio, while their surface functionalization opened the door to a plethora of drug, gene and bioactive molecule immobilization. Taming the high reactivity of the magnetic core was achieved by various functionalization techniques, producing MNPs tailored for the diagnosis and treatment of cardiovascular or neurological disease, tumors and cancer. Superparamagnetic iron oxide nanoparticles (SPIONs) are established at the core of drug-delivery systems and could act as efficient agents for MFH (magnetic fluid hyperthermia). Depending on the functionalization molecule and intrinsic morphological features, MNPs now cover a broad scope which the current review aims to overview. Considering the exponential expansion of the field, the current review will be limited to roughly the past three years.

Abstract:Nanomaterials, in addition to their small size, possess unique physicochemical properties that differ from the bulk materials, making them ideal for a host of novel applications. Magnetic nanoparticle (MNP) is one important class of nanomaterials that have been widely studied for their potential applications in nanomedicine. Due to the fact that MNPs can be detected and manipulated by remote magnetic fields, it opens a wide opportunity for them to be used in vivo. Nowadays, MNPs have been used for diverse applications including magnetic biosensing (diagnostics), magnetic imaging, magnetic separation, drug and gene delivery, and hyperthermia therapy, etc. This review aims to provide a comprehensive assessment of the state-of-the-art biological and biomedical applications of MNPs. In addition, the development of high magnetic moment MNPs with proper surface functionalization has progressed exponentially over the past decade. Herein, we also reviewed the recent advances in the synthesis and surface coating strategies of MNPs. This review is not only to provide in-depth insights into the different synthesis, biofunctionalization, biosensing, imaging, and therapy methods but also to give an overview of limitations and possibilities of each technology.

Abstract:Magnetic nanospheres have numerous applications in biomedicine, biotechnology and wastewater treatment, due to their high surface area, tunable sphere size and superparamagnetic properties. Magnetic nanoparticles can be designed and endowed with optical, electronic and fluorescent properties, allowing a wide range of functionality. Multifunctional magnetic particles with heterodimer structures allow various kinds of target molecules to be attached onto their specific parts via affinity or coordinate bonding, etc. The abilities of these nanodevices, including the encapsulation of target molecules in magnetic hybrid nanostructures and easy magnetic separation in the presence of external magnetic fields, show much promise for magnetic imaging, magnetic separation and drug delivery. Consequently, magnetic particles offer excellent potential future uses in disease diagnosis, hyperthermia, immunoassays, electrochemical biosensors, contaminated water treatment and optical detection. In this article, we review the preparation and application of inorganic and organic magnetic composite spheres in the fields of magnetic separation, drug delivery, hyperthermia, magnetic resonance imaging, and others. The size, specific surface area, structure, magnetic properties and surface functional groups of nanospheres have a great influence on their effectiveness in these applications. The encapsulation of target molecules in magnetic hybrid nanostructures and their easy separation using an external magnetic field show promise for the fabrication of novel nanodevices for many applications. Abstract:This review outlines diverse magnetic nanoparticle (MNP) designs, materials, morphologies, and characterization methods. It assesses various MNP synthesis techniques, their pros and cons, and explores inorganic and organic surface coatings. Biomedical MNP applications are covered, spanning hyperthermia, MRI, drug delivery, biosensors, and magnetic particle imaging. Future MNP structure trends and novel biomedical prospects are also discussed. Nowadays, magnetic nanoparticles (MNPs) are applied in numerous fields, especially in biomedical applications. Since biofluidic samples and biological tissues are nonmagnetic, negligible background signals can interfere with the magnetic signals from MNPs in magnetic biosensing and imaging applications. In addition, the MNPs can be remotely controlled by magnetic fields, which make it possible for magnetic separation and targeted drug delivery. Furthermore, due to the unique dynamic magnetizations of MNPs when subjected to alternating magnetic fields, MNPs are also proposed as a key tool in cancer treatment, an example is magnetic hyperthermia therapy. Due to their distinct surface chemistry, good biocompatibility, and inducible magnetic moments, the material and morphological structure design of MNPs has attracted enormous interest from a variety of scientific domains. Herein, a thorough review of the chemical synthesis strategies of MNPs, the methodologies to modify the MNPs surface for better biocompatibility, the physicochemical characterization techniques for MNPs, as well as some representative applications of MNPs in disease diagnosis and treatment are provided. Further portions of the review go into the diagnostic and therapeutic uses of composite MNPs with core/shell structures as well as a deeper analysis of MNP properties to learn about potential biomedical applications.

Abstract:Magnetic microsphere, a new kind of functional material, was widely used in biomedical fields. At first, some preparative methods of magnetic microsphere were summarized, including physical and chemical methods such as spray evaporation, heat treatment, emulsion polymerization, self-assembly and so on. Abstract:Magnetic nanoparticles (MNPs) bear many intriguing properties such as superparamagnetism, high specific surface area, remarkable colloidal stability and biocompatibility, which evoke great interest and desire of exploration in biomedical applications. For the use in the complicated physiological environment, MNPs are still being developed to have the enhanced performances and down-to-earth practicality. Engineering of MNPs into hierarchical structures is thus proposed to create a new family of magnetic materials, magnetic colloidal supraparticles (MCSPs), which exhibit collective properties and unique nanomaterial characters. From a biomedical point of view, applicability of MCSPs is somewhat more distinctive in contrast to their primary MNPs, because MCSPs are amenable to modulation of secondary structure, promotion of magnetic responsiveness and ease of function design. As a result, MCSPs have been subject to intense researches in recent years, with the aim to develop outstanding composite materials for biomedical applications. In this review, we embark on an overview of foundational topics that detail the design and fabrication of MCSPs by evaporation-induced emulsion and solvothermal techniques, and continue with a guideline for modification of MCSPs with inorganic oxides and organic polymers. Particular focus is then placed on the biomedical applications of modified MCSPs. Many examples illustrate the latest progress in design of MCSP-based microspheres for magnetic resonance imaging, targeted drug delivery, sensing, and harvesting of peptides/proteins. After these detailed accounts, the current challenges and future development of researches and applications are discussed as a conclusion to the review.
