Abstract:The immobilization of tris (2,2'-bipyridyl) ruthenium (II), Ru(bpy)(3)(2+), at a glassy carbon electrode was achieved by entrapping the Ru(bpy)(3)(2+) in a vapor deposited titania sol-gel membrane. The electrogenerated chemiluminescence (ECL) of the immobilized Ru(bpy)(3)(2+) was studied. The Ru(bpy)(3)(2+) modified electrode showed a fast ECL response to both oxalate and proline. The ECL intensity was linearly related to concentrations of oxalate and proline over the ranges from 20 to 700 mumol L-1 and 20 to 600 mumol L-1, respectively. The detection limits for oxalate and proline at 3sigma were 5.0 mumol L-1 and 4.0 mumol L-1, respectively. This electrode possessed good precision and stability for oxalate and proline determinations. The electrogenerated chemiluminescence mechanism of proline system was discussed. This work provided a new way for the immobilization of Ru(bpy)(3)(2+) and the application of titania sol-gel membrane in electrogenerated chemiluminescence.

Abstract:Electrogenerated chemiluminescence(ECL) ,which is a chemiluminescence produced directly or indirectly as a result of electrochemical reactions,has proved to be a sensitive and selective detection method with a wide linear range partly due to the electrochemical excitation during ECL reaction and no external excitation light source,which gives an extremely low background signal without expensive instruments. Tris(2,2’-bipyridyl) ruthenium(Ⅱ) Ru(bpy) 32+ and its derivatives-basedECL systems have shown great importance for the analysis of various compounds such as oxalate,amino acids,aliphatic amines,cyclic amines,aromatic amines,pharmaceuticals or proteins due to its ability to undergo ECL in aqueous solutions and its good chemical,electrochemical,and photochemical stability. The ability of ruthenium complexes to produce ECL with a large number of analytes often requires separation steps prior to detection. The ECL of Ru(bpy) 32+ is often used as a detection method in flowing systems such as FIA,HPLC,CE and MCE etc. Since Ru(bpy) 32+ can be electrochemically regenerated,there is currently a strong interest in immobilizing this species onto the electrode surface to form ECL detectors. There is no need to continually deliver this reagent to the electrode surface,thus simplifying instrumentation,removing the need for an extra-pump,avoiding the problems of both sample dilution and band broadening and largely reducing the consumption of the expensive reagents. We have fabricated several novel solid-state ECL sensors with high sensitivity and good stability. More importantly,several devices such as FIA,CE and MCE in combination with the solid-state ECL detectors have been constructed in our laboratory. Based on our research works,a review related some new progresses in ECL techniques with Ru(bpy) 32+ has been presented.

Abstract:In this correspondence, we report on the first preparation of novel, robust Ru(bpy)32+-containing supramolecular microstructures via a solution-based self-assembly strategy, carried out by directly mixing H2PtCl6 and Ru(bpy)3Cl2 aqueous solutions at room temperature. It reveals that both the molar ratio and concentration of reactants have a heavy influence on the morphologies of such microstructures. The electrochemical behavior of the Ru(bpy)32+ components contained in the solid film of the microstructures formed on the electrode surface is also studied and found to exhibit a diffusion-controlled voltammetric feature. Most importantly, such microstructures exhibit excellent electrochemiluminescence (ECL) behaviors and therefore hold great promise as new luminescent materials for solid-state ECL detection in capillary electrophoresis (CE) or CE microchip.

Abstract:Electrochemiluminescence (ECL) of tris(2,2'-bipyridyl) ruthenium [Ru(bpy)(3)(2+)] has received considerable attention. By immobilizing Ru(bpy)(3)(2+) on an e electrode surface, solid-state ECL provides several advantages over solution-phase ECL, such as reducing consumption of expensive reagent, simplifying experimental design and enhancing the ECL signal.This review presents the state of the art in solid-state ECL of Ru(bpy)(3)(2+). It briefly covers the mechanism of ECL of Ru(bpy)(3)(2+), some recent developments in fabrication of solid-state ECL sensors, analytical applications of ECL and alignment of ECL with capillary electrophoresis (CE), microchip CE, flow-injection analysis, high-performance liquid chromatography. it also indicates the outlook in this field. Abstract:Over the past decades electrochemiluminescence (ECL) of tris(2,2'-bipyridyl)ruthenium [Ru(bpy)(3)(2+)] has received considerable attention from many researchers and been widely used to detect a variety of analytes that range from metal ions and small molecules to DNA, peptides, and proteins. Among all the areas of Ru(bpy)(3)(2+) ECL, the Ru(bpy)(3)(2+) based ECL sensors are of great active. In this chapter we will review the state of the art of Ru(bpy)(3)(2+) ECL sensors. After a brief introduction of Ru(bpy)(3)(2+) ECL and its mechanisms, the fabrications and applications of the Ru(bpy)(3)(2+) ECL sensors are discussed in details. It also indicates the future outlook in this field.

Abstract:A novel method based on click chemistry for the immobilization of Ru(bpy)32+ derivative on electrode to fabricate a solid state electrochemiluminescence (ECL) sensor was reported. Pentenyl functionalized Ru(bpy′)3Cl2 was synthesized and immobilized on a (3-mercaptopropyl) trimethoxysilane (MPTMS) pretreated indium tin oxide (ITO) substrate via thiol-ene click chemistry reaction between its alkenyl group and the sulfhydryl group of MPTMS. The immobilization method is simple, fast, and efficient. More importantly, the resulting Ru(bpy′)32+ monolayer film exhibited excellent stability in organic solvent, which is superior to many previously reported methods. Using tri-n-propylamine (TPrA) as a model analyte, the ECL sensor gave a wide linear range over 5×10−6 to 5×10−3M, with a satisfactory detection limit of 1×10−6M (S/N=3).

Abstract:Spherical Ru(bpy)2+3-doped silica (RuSi) nanoparticles were prepared via a water-in-oil microemulsion approach. The electrochemical and electrochemiluminescent properties of the RuSi nanoparticles immobilized on an indium tin oxide (ITO) electrode were investigated. Further, electrochemiluminescence (ECL) of the RuSi nanoparticles with covalently coated biomacromolecules was studied. By covalent cross-linking with glutaraldehyde, γ-(aminopropyl) triethoxysilane (APTES)-pretreated RuSi nanoparticles were coupled with different concentrations of bovine serum albumin (BSA), hemoglobin, and myoglobin, respectively. ECL from these biomacromolecule-coated RuSi nanoparticles decreased with the increase of the loading of biomacromolecules. Moreover, the ECL of coreactants with different sizes was studied. The ECL decrease could be assigned to the steric hindrance and limited diffusion of coreactant molecules into the RuSi nanoparticles after biomacromolecule conjugation. Since tens of thousands of Ru(bpy)2+3 molecules are contained in the silica particles and the RuSi nanoparticle surface modification could improve their biocompatibility, the biomacromolecule-coated RuSi nanoparticles could be readily used as efficient and stable ECL tag materials in the future.

Abstract:An electrochemically induced sol-gel process has been used to efficiently immobilize Ru(bpy)(3)(2+) in a 3D porous silica film matrix deposited on a glassy carbon electrode (GCE), forming a solid state electro-chemiluminescence (ECL) sensor. In this approach, electrolysis of the GCE at cathodic voltages from a solution of ammonium fluorosilicate containing Ru(bpy)(3)(2+) results in the reduction of water to hydroxyl ions and hydrogen bubbles. The former product catalyzes the hydrolysis of ammonium fluorosilicate to form a silica film; while the hydrogen bubbles act as a dynamic template in forming a porous silica matrix. Therefore, a large quantity of Ru(bpy)(3)(2+) ions can be efficiently encapsulated in the porous silica matrix, and the formed porous structure offers a good mass transport path. The fabricated [ Ru(bpy)(3)](2+) solid-state ECL sensor shows high sensitivity and stability towards the determination of tripropylamine (TPA). The electrochemically generated luminescence signal shows a good linear relationship to TPA concentration ranging from 3.46 x 10(-10) to 3.70 x 10(-6) M and 3.70 x 10(-6) to 3.60 x 10(-4) M, with an extremely low detection limit of 17 pM (S/N = 3). The present approach is effective for encapsulation of various molecules and could find wide application in the construction of various sensors.

Abstract:Electrochemical behavior and electrogenerated chemiluminescence (ECL) of tris(2,2'-bipyridyl) ruthenium(II) (Ru(bpy)(3)(2+)) immobilized in poly(3,4-ethylenedioxythiophene)/poly(styrenesulfonate)poly(vinyl alcohol) (PEDOT/PSS-PVA) composite films via ion-exchange have been investigated with tripropylamine (TPA) as the co-reactant at a glassy carbon electrode. The immobilized Ru(bpy)(3)(2+) performed a surface-controlled electrode reaction. The Ru(bpy)(3)(2+) modified electrode showed a fast ECL response to TPA, and was used for the ECL detection of TPA with high sensitivity. The ECL intensity was linearly related to concentrations of TPA over the range from 0.50 mu mol L-1 to 0.80mmol L-1, and the detection limit was 0.10 mu mol L-1 (S/N=3). Highly efficient electrochemiluminescence of functionalized tris(2,2′-bipyridyl)ruthenium(II) and selective concentration enrichment of its co-reactants

Abstract:An enhanced electrochemiluminescence (ECL) efficiency is obtained from the ruthenium complex tris(2,2'-bipyridyl)ruthenium(II) (Ru(bpy)(3)(2+)) by introduction of an ionic liquid (IL) 1-butyl-3-methylimidazolium tetrafluoroborate (BMImBF(4)). Upon addition of 1% (v/v) BMImBF(4) to 0.1 mm Ru(bpy)(3)(2+) solution, a maximum increase in ECL intensity is obtained both at an indium tin oxide (ITO) electrode (15-fold) and at a glassy carbon (GC) electrode (5- to 64old). Furthermore, upon addition of 1% (v/v) BMImBF4 to 5 pm Ru(bpy)(3)(2+)/100 mm co-reactant systems at a GC electrode, IL adsorption occurs at the electrode surface, which results in a change of the polarity of the electrode surface. Such functionalization greatly improves the functions of both Ru(bpy)(3)(2+) and ionic liquids, as is demonstrated in the sensitive and selective concentration enrichment of the Ru(bpy)(3)(2+) co-reactants.
Chemiluminescent Investigation of Tris(2,2'-Bipyridyl)ruthenium(ii) Immobilized on a Cationic Ion-Exchange Resin and Its Application to Analysis

Abstract:Tris-(2, 2'-bipyridyl)ruthenium(II) complex, Ru(bpy)(3)(2+), was immobilized on the Dowex-50 W cationic ion-exchange resin. The chemiluminescent characteristics of Ru(bpy)(3)(2+) in solution and in resin form were compared by using batch and flow injection methods. A strong chemiluminescence was observed during the reaction of Ru(bpy)(3)(2+) both in solution and in resin with KMnO(4) or Ce(SO(4))(2) under acidic or basic conditions. The Ru(bpy)(3)(2+) immobilized resin is stable, which can be used at least for 6 months when it reacts with the dilute KMnO(4) solution. Based on this property, Ru(bpy)(3)(2+) immobilized in the resin phase was developed as a flow-through chemiluminescent sensor that could be used to determine oxalate, sulfite and ethanol chemically or electronically with Ru(bpy)(3)(3+) generation on the surface of resin. The limits of detection were 1 x 10(-6) M for oxalate, 0.5% (v/v) for ethanol and 1 x 10(-7) M for sulfite. The method has been successfully applied to determine sulfite in sugar.

Abstract:Ru(bpy)3Cl2 was used to modify the glass carbon electrodes (GCE) by oxidation and co-deposition on the electrode surface. The modified electrodes were characterized by atomic force microscopy (AFM) and X-ray photoelectron spectroscopy (XPS). About 2.2 × 10−9 mol Ru(bpy)3 2+ was immobilized on the GCE surface (ϕ = 4 mm). The modified GC electrodes showed stable electrochemiluminescence with tripropylamine (TPrA) as the co-reactant with a linear range from 10 to 500 μM (R 2 = 0.999). Among the 10 amino acids tested, the modified electrode system showed selective response to arginine and lysine, indicating that the molecular structure played an important role as co-reactant. This simple and sensitive electrode modifying method when combined with flow-injection or liquid chromatography systems has the potential for amino acid analyses.
Particle Size-Controlled Oxygen Reduction and Evolution Reaction Nanocatalysts Regulate Ru(bpy)32+'s Dual-potential Electrochemiluminescence for Sandwich Immunoassay

Abstract:Multiple signal strategies remarkably improve the accuracy and efficiency of electrochemiluminescence (ECL) immunoassays, but the lack of potential-resolved luminophore pairs and chemical cross talk hinders their development. In this study, we synthesized a series of gold nanoparticles (AuNPs)/reduced graphene oxide (Au/rGO) composites as adjustable oxygen reduction reaction and oxygen evolution reaction catalysts to promote and modulate tris(2,2'-bipyridine) ruthenium(II) (Ru(bpy)32+)'s multisignal luminescence. With the increase in the diameter of AuNPs (3 to 30 nm), their ability to promote Ru(bpy)32+'s anodic ECL was first impaired and then strengthened, and cathodic ECL was first enhanced and then weakened. Au/rGOs with medium-small and medium-large AuNP diameters remarkably increased Ru(bpy)32+'s cathodic and anodic luminescence, respectively. Notably, the stimulation effects of Au/rGOs were superior to those of most existing Ru(bpy)32+ co-reactants. Moreover, we proposed a novel ratiometric immunosensor construction strategy using Ru(bpy)32+'s luminescence promoter rather than luminophores as tags of antibodies to achieve signal resolution. This method avoids signal cross talk between luminophores and their respective co-reactants, which achieved a good linear range of 10-7 to 10-1 ng/ml and a limit of detection of 0.33 fg/ml for detecting carcinoembryonic antigen. This study addresses the previous scarcity of the macromolecular co-reactants of Ru(bpy)32+, broadening its application in biomaterial detection. Furthermore, the systematic clarification of the detailed mechanisms for converting the potential-resolved luminescence of Ru(bpy)32+ could facilitate an in-depth understanding of the ECL process and should inspire new designs of Ru(bpy)32+ luminescence enhancers or applications of Au/rGOs to other luminophores. This work removes some impediments to the development of multisignal ECL biodetection systems and provides vitality into their widespread applications.

Abstract:In this paper, we report significantly enhanced electrochemiluminescence (ECL) of tris(2,2'-bipyridyl)ruthenium(II) (Ru(bpy)₃²⁺) at indium tin oxide (ITO) electrodes modified with a thin film consisting of vertically aligned silica mesochannels (SMCs). The perpendicular SMCs allow for the effective mass transport of Ru(bpy)₃²⁺ and its co-reactants from the solution to the underlying ITO electrode surface. Moreover, due to the ultrasmall diameter (namely 2-3 nm) and the negatively charged surface, SMCs exerted a strong electrostatic attraction to the positively charged Ru(bpy)₃²⁺ to accelerate its mass transport, resulting in a remarkably increased ECL signal. As a model system, when using tri-n-propylamine (TPrA) as the co-reactant, the intensity of ECL generated at the SMCs-modified ITO electrode was stronger than that at a bare ITO electrode by more than two orders of magnitude (i.e., ∼107-fold increase). In this case, the amount of Ru(bpy)₃²⁺ required for ECL-based analysis can be considerably reduced. A very low concentration of Ru(bpy)₃²⁺, namely 9 μM, was demonstrated to be enough for achieving the sensitive detection of TPrA, nicotine and atropine. Both photomultiplier (PMT) and charge coupled device (CCD) were used to measure the luminescence from the ECL reaction. The obtained detection limits were 0.17 nM for TPrA on the basis of the ECL intensity measurement, 72 nM for nicotine and 38 nM for atropine based on the gray value analysis of captured ECL images.

Abstract:Tris(2,2'-bipyridine)ruthenium(II) ([Ru(bpy)3]2+) is one of the most extensively studied and used electrochemiluminescent (ECL) compounds owing to its superior properties, which include high sensitivity and stability under moderate conditions in aqueous solution. In this paper we present a simple method for the preparation of [Ru(bpy)3]2+-containing microstructures based on electrostatic assembly. The formation of such microstructures occurs in a single process by direct mixing of aqueous solutions of [Ru(bpy)3]Cl2 and K3[Fe(CN)6] at room temperature. The electrostatic interactions between [Ru(bpy)3]2+ cations and [Fe(CN)6]3- anions cause them to assemble into the resulting microstructures. Both the molar ratio and concentration of reactants were found to have strong influences on the formation of these microstructures. Most importantly, the resulting [Ru(bpy)3]2+-containing microstructures exhibit excellent ECL behavior and, therefore, hold great promise for solid-state ECL detection in capillary electrophoresis (CE) or CE microchips.
