Abstract:The acid-catalyzed nucleophilic addition reaction of spiroketal enol ether-containing tonghaosu analogs 2 was explored. Soft nucleophiles, such as mercaptans, alcohols and heteroaromatic compounds, gave rise exclusively to 1,6-adducts, while harder nucleophiles, such as Grignard reagents, afforded mixtures of 1,2- and 1,6-adducts. The reaction with cysteine and glutathione, which might be related to the mode of action of insect antifeeding, also took place smoothly. Friedel-Crafts dimerization reactions of 10 and 11 gave the dimers 12 and 13 respectively. By comparison of H-1 NMR spectroscopic data, we also suggest that the structure of compound 9 reported in the literature for a natural product is revised to that of compound 13. Abstract:Seventeen novel dihydrothiazole derivatives were synthesized from compounds A and A'in one step. The structures of the compounds were characterized by IR, 1H-NMR, 13C-NMR,and MS analysis.The bioas-say showed that some of the synthesized dihydrothiazole derivatives exhibited excellent antibacterial activities. Especially,compounds 5,5'and 6 exhibited much higher activities against bacillus subtilis and staphylococcus aureus than penicillin.

Abstract:With the increasing incidence of antibiotic resistance, there is an urgent need to develop new antibiotics with excellent activity against drug-resistant bacteria. Three novel series of tylosin semisynthetic derivatives were designed, synthesized and evaluated for their antibacterial activities against various Gram-positive and Gram-negative bacteria. Among these derivatives, compound C-2 demonstrated potent antibacterial activity against both gram-positive and gram negative bacteria, and non mutagenic. More importantly, compound C-2 displayed high antimicrobial potency against Gram-positive bacteria in a murine model, and was found to be more efficient than tildipirosin. Thus, compound C-2 had great potential as a promising lead compound for the treatment of bacterial infection.

Abstract:In order to discover more promising antifungal and antibacterial agents, a series of new derivatives were designed and synthesized by structure modification based on the naturally occurring antimicrobial compound lophanic acid. The structures of all the target compounds were well characterized by spectroscopic data. The stereochemistry of these compounds was further determined through the X-ray diffraction analysis of 6a. The synthetic compounds were evaluated for their antimicrobial activities against filamentous fungi (T. rubrum, T. mentagrophytes), yeasts (C. neoformans, C. albicans) and Gram-positive and Gram-negative bacteria (MRSA, S. mutans, S. sobrinus, and E. coli). Among them, 3d and 3i are found as the most promising leads that showed potent inhibitory effects against all the tested fungal and bacterial strains except for E. coli. The presence of the C-20 carboxylic ester groups and the free hydroxy group at C-13 was found to be essential for the antifungal and antibacterial activities of the lophanic acid derivatives.

Abstract:Two novel sulfur-containing dibenzofurans, sorbusins A (1) and B (2), two unprecedented biphenyl glycosides, 2'-hydroxyaucuparin 2'-O-ɑ-L-rhamnoside (3) and noraucuparin 5-O-ɑ-L-rhamnoside (4), and four known analogues (5-8), were isolated from Sorbus pohuashanensis suspension cell induced by yeast extract. Their structures were elucidated based on spectroscopic analyses and quantum calculation of 13C NMR data. Structurally, compound 1 possessed a rare naturally occurring benzothiazole moiety and represents the first example of thiazole fused dibenzofuran. A plausible biosynthetic pathway for the sulfur-containing dibenzofurans is proposed. These dibenzofuran and biphenyl phytoalexins were evaluated for their antimicrobial activities against pathogenic fungi and drug-resistant bacteria. Compound 7 exhibited significant antibacterial activity against methicinllin-resistant Staphylococcus aureus with an MIC value of 3.13 μg/mL.

Abstract:Three isocoumarins, ascoisocoumarin A ( 1 ), embeurekol ( 2 ), and sclerotinin A ( 3 ), and five biosynthetically related derivatives, ascospinols A–C ( 4 , 6 , and 7 ), and talaflavuols C and B ( 5 and 8 ), together with twelve polyketides or terpenes ( 9 – 20 ) were isolated from the fungus Aspergillus sp. LY-1-2 inhabited in a sample of Cordyceps sp. Most of them belong to the family of oxygen-containing aromatic compounds and compounds 1 , 4 , 6 , and 7 are previously undescribed compounds. Their planar structures were established by a combined spectroscopic analysis of HRESIMS and NMR, and their stereochemistry was determined by 13 C NMR calculations with sorted training set (STS) protocol analysis, and ECD calculations. New compounds 1 and 6 displayed potential anti-inflammatory effects in lipopolysaccharide (LPS)-induced BV2 microglia cells.

Abstract:Toona sinensis is one of the traditional Chinese medicinal materials with rich physiological activities. This study aimed to investigate the chemical composition differences of the essential oils from the tender leaves and shoots of T. sinensis , collected from four main producing areas in China, and to verify their anti-listeria and antioxidant properties. Gas chromatography-mass spectrometry (GC-MS) analysis was conducted on the essential oils obtained by hydro-distillation. The results indicated that compounds such as β-elemene (3.5∼8.1%), α-caryophyllene (3.7∼7.2%), and trans -caryophyllene (4.0∼9.7%) were the main common components of T. sinensis essential oils (TSEO) from four producing areas. The results of DPPH and ABTS showed that the essential oil from Dazhou, Sichuan, China had the best antioxidant effect. At a concentration of 1.25 mg/mL, the antioxidant activity of TSEO-DZ from Sichuan was comparable to 0.08 mg/mL of BHT and was only slightly lower than 0.06 mg/mL of VC. The broth dilution method showed that the essential oil from Tai 'an, Shandong, China, showed better antibacterial effect, with the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MLC) values of 1.25 μL/mL and 2.54 μL/mL, respectively. This study demonstrates that TSEO possesses promising application potential in antioxidant and anti-listeria activities, and it confirms that the growth environment of T. sinensis significantly influences the quality, content, and biological activity of the essential oil.

Abstract:Streptomide (1), a new amide analogue, streptomynone (2), a new quinolinone, and ten known compounds including three aliphatic acids (3‐5), two amides (6‐7), four cyclic dipeptides (8‐11), and an adenosine (12) were also obtained from the fermentation broth of a Streptomyces isolate collected from a sediment sample of soil sample collected in Bendong Village, Huadong Town, Chuxiong, China. Their structures were determined by analysis of the 1D/2D‐NMR and HR‐ESI‐MS spectra. Compound 12 presented weak antimicrobial activities against Candida albicans and Aligenes faecalis (MIC = 64 μg/mL). Compounds 7 and 12 showed weak cytotoxic activity against MHCC97H.

Abstract:The collective total synthesis of (+)-sinensilactam A, (+)-lingzhilactone B, (+)-lingzhilactone C and (-)-lingzhiol has been accomplished from a common epoxide intermediate 9. Chemoselective epoxy opening with either an aryl or alkene moiety of styrene led to different carbon skeletons, which can be advanced to a divergent and concise total synthesis of four meroterpenoids.

Abstract:Structural modification of natural products by biotransformation with fungi is an attractive tool to obtain novel bioactive derivatives. In the present study, cryptotanshinone (1), a quinoid abietane diterpene from traditional Chinese medicine Salvia miltiorrhiza (Danshen), was transformed by two marine-derived fungi. By using Cochliobolus lunatus TA26-46, one new oxygenated and rearranged product (2), containing a 5,6-dihydropyrano[4,3-b]chromene moiety, together with one known metabolite (10), were obtained from the converted broth of cryptotanshinone (1) with the isolated yields of 1.0% and 2.1%, respectively. While, under the action of Aspergillus terreus RA2905, seven new transformation products (3–9) as well as 10 with the fragments of 2-methylpropan-1-ol and oxygenated p-benzoquinone were produced and obtained with the isolated yields of 0.1%–1.3%. The structures of the new compounds were elucidated by comprehensive spectroscopic analysis including High Resolution Electrospray Ionization Mass Spectroscopy (HRESIMS), Nuclear Magnetic Resonance (NMR) and Electronic Circular Dichroism (ECD). The metabolic pathways of cryptotanshinone by these two fungi were presumed to be the opening and rearrangement of furan ring, and/or oxygenation of cyclohexane ring. Cryptotanshinone (1) and its metabolites displayed anti-inflammatory activities against NO production in LPS-stimulated BV-2 cells and antibacterial activities towards methicillin-resistant Staphylococcus aureus. These findings revealed the potential of marine fungi to transform the structures of natural products by biotransformation.

Extensive expansion of the chemical diversity of fusidane-type antibiotics using a stochastic combinational strategy

Abstract:Fusidane-type antibiotics, represented by helvolic acid, fusidic acid and cephalosporin P1, are fungi-derived antimicrobials with little cross-resistance to commonly used antibiotics. Generation of new fusidane-type derivatives is therefore of great value, but this is hindered by available approaches. Here, we developed a stochastic combinational strategy by random assembly of all the post-tailoring genes derived from helvolic acid, fusidic acid, and cephalosporin P1 biosynthetic pathways in a strain that produces their common intermediate. Among a total of 27 gene combinations, 24 combinations produce expected products and afford 58 fusidane-type analogues, of which 54 are new compounds. Moreover, random gene combination can induce unexpected activity of some post-tailoring enzymes, leading to a further increase in chemical diversity. These newly generated derivatives provide new insights into the structure‒activity relationship of fusidane-type antibiotics. The stochastic combinational strategy established in this study proves to be a powerful approach for expanding structural diversity of natural products.

Abstract:A series of new 2-(1-(2-(substituted-phenyl)-5-methyloxazol-4-yl)-3-(2-substitued-phenyl)-4,5-dihydro-1H-pyrazol-5-yl)-7-substitued-1,2,3,4-tetrahydroisoquinoline derivatives were synthesized. The results showed that compounds 9q and 10q can strongly inhibit Staphylococcus aureus DNA gyrase and Bacillus subtilis DNA gyrase.

Abstract:In this paper, we report twelve new (4S, 5R)-Lactivicin derivatives. They have been first synthesized by using L-threonine as starting materials through two different routes. The in vitro antibacterial test showed that five of them have antibacterial activities and three of them have higher antibacterial activities than the Lactivicin. Their chemical structures have been identified with IR, PMR, MS and elementary analysis.
