Protein Expression and Purification

Abstract:Lumbrokinase (LK) is an important fibrinolytic enzyme derived from earthworms. It has been found that LK is composed of a group of isoenzymes. To construct and express the mature peptide of LK PI239 in Escherichia coli, we amplified and optimized the gene of LK which was then cloned into the prokaryotic expression vector pET-22b(−). The recombinant LK (rLK) protein was expressed as inclusion bodies and we have developed a purification process of rLK from these inclusion bodies. A step-down urea concentration strategy was applied to the rLK renaturation process. The purified and renatured rLK apparently ameliorated the conditions of the model thrombosis rats used, and may be developed into a therapeutic agent for thrombotic-associated diseases.

Abstract:Enterokinase (EC 3.4.21.9) is a serine proteinase of the intestinal brush border that exhibits specificity for the sequence (ASP)4-Lys and converts trypsinogen into its active form, trypsin. A codon optimized sequence coding light chain (catalytic subunit) of bovine enterokinase gene (sBEKLC) was synthesized, and it was fused with DsbA to construct the expression vector (pET39-sBEKLC). Then, the plasmid was transformed into E. coli BL21 (DE3) for expression. Under optimal conditions, the volumetric productivity of fusion protein reached 151.2mg L-1, i.e., 80.6 mg sBEKLC L-1. The cold osmotic shock technique was successfully used to extract sBEKLC from periplasmic space, and nickel affinity chromatography was employed to obtain mature sBEKLC. Finally, about 6.8 mg of bioactive sBEKLC was purified from 1 liter fermentation broth and could be used to cleave one tested fusion protein with an inter-domain enteropeptidase recognition site. This work will be helpful for large-scale production of this increasingly demanded enterokinase.

Abstract:AIM: To construct engineered E.coli strains which can express nattokinase with fibrinolysis activity using gene engineering technology. METHODS: The pro-nattokinase (pro-NK) gene was amplified by PCR and inserted into expression vector pET3c. The recombined plasmid pENK which expressed the fusion protein of pro-NK and 22 amino acid peptide was then transferred into lysogenic host strains BL21(DE3)pLysS - and BL21(DE3)pLysS +. Both SDS-PAGE and the fibrin plate assay were used to examine the expression and the activity of the target protein. RESULTS: SDS-PAGE assay showed the fused gene encoding 42 kD fusion protein was expressed in both expression strains pENK-(DE3)pLysS - and pENK-(DE3)pLysS +, and the fibrin plate assay indicated that the expression product had fibrinolysis activity. pENK-(DE3)pLysS - exhibited the basal expression of the target gene, while fusion protein was only induced by IPTG in pENK-(DE3)pLysS +. Basal expression of the fused toxic gene in pENK-(DE3)pLysS - led to bacteriolysis and hollow lawns. CONCLUSION: A pro-NK fusion protein with fibrinolysis activity is successfully expressed in E.coli , which lay a foundation for the exploitation of nattokinase.
Bovine Enterokinase Catalytic Subunit cDNA Cloning and Construction of Fusion Expression Plasmid

Abstract:Enterokianse (EK) is one of the most useful tools as a fusion protein cleavage regent for putification of recombinant protein production in biopharmaceutical industry. This study was to obtain gene of bovine enterokinase catalytic subunit, which would be used in cleavage and purification of fusion proteins. The fragment of bovine Enterolinase catalytic subunit cDNA was obtained by RT-PCR from a sold bovine's duodenal mucosa, and then cloned into pUCm-T cloning vector and sequenced. Compared with deposited sequence in GenBank, the sequence of gene cloned in this study is accordant. The intersted gene fragment was inserted into pET32a expression plasmid. Finally it was confirmed that the nucleotide sequence was accurate on conjunction site between recombinant DNA 5' terminal multi-cloning site and recombinant fragment after the analysis of nucleotide sequence. SDS-PAGE analysis indicated that target product was about 50 kD which occupied 38% of the total protein. This investigation would be able to lay a foundation for gene expression and activity research of expression products.

Abstract:Objective To obtain a high-purity and high-activity recombinant bovine enterokinase catalytic subunit protein by inducing the recombinant GS115 yeast containing the bovine enterikinase catalytic subunit gene. Methods The DNA sequence encoding the light chain catalytic subunit of bovine enterokinase GenBank Accession No. NM174439 was synthesized from the fresh bovine duodenal mucosa by reverse trscriptional polymerase chain reaction and cloned into plasmid pPIC9K downstream to the gene of fusion partner His-tag. The recombinant pPIC9K-bovine enterokinase light chain vector(pPIC9K-EKL) was expressed in Pichia pastoris GS115 strain in secreted form. The active recombinant bovine enterokinase light chain rEKL was purified on nickel chelating chromatogram using His Binding Resin and Q-Sepharose FF column chromatography. The purified intact rEKL was measured by cleavage fusion protein for its biological activity. Results 3.0 mg/L of recombinant bovine enterokinase catalytic subunit protein was obtained from flask culture and the percentage of cleaved IL-11/MBP fusion protein with enterokinase recognition site was more than 90%. Conclusion The high-purity bovine recombinant enterokinase catalytic subunit protein was obtained which would be able to lay foundation for its further application of products on a large scale.

Abstract:<p>Keratinases are unique among proteolytic enzymes for their ability to degrade recalcitrant insoluble proteins, and they are of critical importance in keratin waste management. Over the past few decades, researchers have focused on discovering keratinase producers, as well as producing and characterizing keratinases. The application potential of keratinases has been investigated in the feed, fertilizer, leathering, detergent, cosmetic, and medical industries. However, the commercial availability of keratinases is still limited due to poor productivity and properties, such as thermostability, storage stability and resistance to organic reagents. Advances in molecular biotechnology have provided powerful tools for enhancing the production and functional properties of keratinase. This critical review systematically summarizes the application potential of keratinase, and in particular certain newly discovered catalytic capabilities. Furthermore, we provide comprehensive insight into mechanistic and molecular aspects of keratinases including analysis of gene sequences and protein structures. In addition, development and current advances in protein engineering of keratinases are summarized and discussed, revealing that the engineering of protein domains such as signal peptides and pro-peptides has become an important strategy to increase production of keratinases. Finally, prospects for further development are also proposed, indicating that advanced protein engineering technologies will lead to improved and additional commercial keratinases for various industrial applications.</p>

Abstract:Enterokinase is a tool protease widely utilized in the cleavage of recombinant fusion proteins. cDNA encoding the catalytic subunit of Chinese bovine enterokinase (EKL) was amplified by PCR and then fused to the 3' end of prepro secretion signal peptide gene of alpha-mating factor from Saccharomyces cerevisiae to get the alpha-MF signal-EKL-His6 encoding gene by PCR. Then the whole coding sequence was cloned into the integrative plasmid pAO815 under the control of a methanol-inducible promoter and transformed GS115 methylotrophic strain of Pichia pastoris. Secreted expression of recombinant EKL-His6 was attained by methanol induction and its molecular weight is 43 kD. Because of the existence of His6-tag, EKL-His6 was easily purified from P. pastoris fermentation supernatant by using Ni2+ affinity chromatography and the yield is 5.4 mg per liter of fermentation culture. This purified EKL-His6 demonstrates excellent cleavage activity towards fusion protein containing EK cleavage site.

Abstract:The optional high cell-density fermentation conditions,purification methods and biological activity of the recombinant enterokinase in E.coli were studied.In E.coli expressing system,the optimized conditions were temperature 37℃ and pH 7.4.When OD600 is 8.0,with 0.3 mmol/L IPTG induced at temperature 30℃,and continued 3h,the recombinant enterokinase was up to 25% of the total amount of the proteins in E.coli.The purified enterokinase reached a purity of above 80% by SDS-PAGE analysis and showed high activity at fusion protein.

Abstract:Enterokinase (EC 3.4.21.9) is a serine proteinase in the duodenum that exhibits specificity for the sequence (Asp)(4)-Lys. It converts trypsinogen to trypsin. Its high specificity for the recognition site makes enterokinase (EK) a useful tool for in vitro cleavage of fusion proteins. cDNA encoding the catalytic chain of Chinese bovine enterokinase was cloned and its encoding amino acid sequence is identical to the previously reported sequence although there are two one-base mutations which do not change the encoded amino acid. The EK catalytic subunit cDNA was cloned into plasmid pET32a, and fused downstream to the fusion partner thioredoxin (Trx) and the following DDDDK enterokinase recognition sequence. The recombinant bovine enterokinase catalytic subunit was expressed in Escherichia coli BL21(DE3), and most products existed in soluble form. After an in vivo autocatalytic cleavage of the recombinant Trx-EK catalytic domain fusion protein, intact, biologically active EK catalytic subunit was released from the fusion protein. The recombinant intact EK catalytic subunit was purified to homogeneity with a specific activity of 720 AUs/mg protein through ammonium sulfate precipitation, DEAE chromatography, and gel filtration. The purified intact EK catalytic subunit has a K(m) of 0.17 mM, and K(cat) is 20.8s(-1). From 100 ml flask culture, 4.3 mg pure active EK catalytic subunits were obtained.

Abstract:The invention discloses a cuttlefish arginine kinase gene and a prokaryotic expression vector pET 30a-arginine kinase (AK) thereof, and particularly relates to an important kinase--arginine kinase (AK) and a prokaryotic expression vector thereof. The arginine kinase (AK) is directly related to intracellular energy operation, muscle contraction and adenosine triphosphate (ATP) regeneration. Total-length complementary deoxyribonucleic acid (cDNA) of the AK gene is cloned from a liver of a cuttlefish by means of RACRPER technology, then a protein coding sequence of the AK gene is cloned to the prokaryotic expression vector pET 30a-AK, and expression and purification are carried out in an escherichia coli expression strain BL21. Recombinant proteins are obtained through separation, not only can be used as antigens for preparation of AK antibodies, but also have great value in research of an invertebrate intracorporal energy metabolism regulating law.

Abstract:Enterokinase is a serine protease that recognizes Asp-Asp-Asp-Asp-Lys sequence and cleaves the C-terminal peptide bond of the lysine residue.Design eighteen primers according to the sequence of NCBI,slight chain of bovine enterokinase gene was synthesized by SOE-PCR.Construct the expression vector(pET32a-EK).Then the plasmid was transformed into E.coli BL21(DE3)for expression.IPTG was added to express the fusion protein,use lon exchange column to purificy the enterokinase.The purified enterokinase was obtained which would be able to lay foudation for ite further application of products on a large scale.

Abstract:The objective of the study was to obtain the gene of bovine enterokinase light chain, which would be used in the cleavage and purification of fusion proteins. The fragment of bovine enterokinase light chain cDNA was obtained by RT-PCR from a sold bovine's duodenal mucosa, then cloned into the pUCmT cloning vector and sequenced. Compared with the sequence deposited in GenBank,the cloned gene sequence is correct. Then the interested gene fragment was inserted into the pET39b expression plasmid. The recombinant vector pET39b-EKL was transformed into E.coli BL21(DE3) and induced by IPTG. It was confirmed that the nucleotide sequence was correct on the conjunction site between the recombinant DNA 5'terminal multi-cloning site and recombinant fragment after the analysis of the nucleotide sequence. SDS-PAGE analysis indicated that target product was about 65 kDa which occupied 28% of the total protein. A pure fusion protein was obtained by nickel chelating chromatogram using His*Binding Resin. After desalting and changing buffer, the crude kinase was incubated at 21 degrees overnight and demonstrated a high autocatalytic cleavage activity. This investigation would be able to lay a foundation for enterokinase activity research and farther application of expression products on a large scale.

Abstract:Enterokinase is a protease of the intestinal brush border that specifically cleaves the acidic propeptide from trypsinogen to yield active trypsin. This cleavage initiates a cascade of proteolytic reactions leading to the activation of many pancreatic zymogens. The full-length cDNA sequence for bovine enterokinase and partial cDNA sequence for human enterokinase were determined. The deduced amino acid sequences indicate that active two-chain enterokinase is derived from a single-chain precursor. Membrane association may be mediated by a potential signal-anchor sequence near the amino terminus. The amino terminus of bovine enterokinase also meets the known sequence requirements for protein N-myristoylation. The amino-terminal heavy chain contains domains that are homologous to segments of the low density lipoprotein receptor, complement components C1r and C1s, the macrophage scavenger receptor, and a recently described moth shared by the metalloprotease meprin and the Xenopus A5 neuronal recognition protein. The carboxyl-terminal light chain is homologous to the trypsin-like serine proteases. Thus, enterokinase is a mosaic protein with a complex evolutionary history. The amino acid sequence surrounding the amino terminus of the enterokinase light chain is ITPK-IVGG (human) or VSPK-IVGG (bovine), suggesting that single chain enterokinase is activated by an unidentified trypsin-like protease that cleaves the indicated Lys-Ile bond. Therefore, enterokinase may not be the ''first'' enzyme of the intestinal digestive hydrolase cascade. The specificity of enterokinase for the DDDDK-I sequence of trypsinogen may be explained by complementary basic-amino acid residues clustered in potential S2-S5 subsites.

Abstract:Purpose The sequence of bovine EKL gene with Asp-Asp-Asp-Asp-Lys sequence in the N-terminus was obtained and analyzed,and the fusion protein of the EKL-GST was produced in E.coli.Methods The fragment of bovine enterokinase light chain cDNA was obtained by RT-PCR from bovine′s duodenal mucosa,then cloned into the pMD18-T cloning vector and sequenced.Compared with the sequence deposited in GenBank,the cloned gene sequence is correct.Then the interested gene fragment was inserted into the pGEX-2T expression plasmid.The recombinant vector pGEX-rEKL was transformed into E.coli BL21 and induced by IPTG.Results SDS-PAGE analysis indicated that the target product was about 61 kDa.The research successfully clones and expresses EKL-GST fusion protein in E.coli.Selt-activation by microamount cutting the fusion protein,and purified the EKL by minobenzene-Sepharose.Conclusion This investigation would be able to lay a foundation for enterokinase activity research and further application of expression products on a large scale.

Abstract:Nattokinase is a kind of alkaline serine proteinase mainly from Bacillus subtilis and is well known for its strong thrombolytic property.Compared with most of other thrombolytic agents,it has good safety,low cost,easy absorption,low side effects and other advantages,which could be developed as a promising agent for thrombosis therapy.In this paper,the thrombolytic efficacy,physicochemical properties,fermentation process,activity determination,purification,modification and products of nattokinase at home and abroad were reviewed.The key issues in the current research of nattokinase were analyzed,and the future research direction of nattokinase was also prospected.

Abstract:Nattokinase gene was amplified using overlapping polymerase chain reaction (PCR), and cloned into pTWIN1, PET 32a and PML-c2x vectors. The expression plasmids of NK/pTWIN1, NK/PET32a and NK/PML-c2x were constructed and used to transform E. coli. The results showed that the level of the nattokinase protein was a little higher in E. coli transformed by NK/pTWIN1 and NK/PET32a. Nattokinase protein was purified by a method including protein denaturation, protein renaturation and SP sepharose from the E. coli ER2566 transformed by NK/pTWIN1. SDS-PAGE showed that the expression protein was about 36% of total cell protein. The experiment of aga-fibrous protein plate showed that the recombinant nattokinase had potent fibrinolytic activity equivalent to 600 urokinase units per milligram. The expression and purification of such a fibrinolytic enzyme would be helpful in producing nattokinase by gene engineering strain. Fig 5, Tab 1, Ref 11
