Abstract:Polymer-supported chiral ligands 9 and 17 were prepared based on Noyori's (1S, 2S)- or ( 1R, 2R)-N-(p-tolylsulfonyl)1,2- diphenylethylenediamine. The combination with [RuCl2(p-cymene)](2) has been shown to exhibit high activities and enantioselectivities for heterogeneous asymmetric transfer hydrogenation of aromatic ketones (19a - c) with formic acid - triethylamine azeotrope as the hydrogen donor, whereby affording the respective optically active alcohols 20a - c, the key precursors of chiral fluoxetine. As exemplified by ligand 17 for substrate 19c, the catalysts can be recovered and reused in three consecutive runs with no significant decline in enantioselectivity. The procedure avoids the plausible contamination of fluoxetine by the toxic transition metal species.

Abstract:We present the first examples of a group of effective ruthenium catalysts for polycyclotrimerization. The polymerization reactions of an arylene bipropiolate monomer initiated by the Ru(II-IV) complexes of CpRu(PPh3)(2)Cl, Cp*Ru(PPh3)(2)Cl, [Cp*RuCl2](n), and [Ru(eta(3):eta(3)-C10H16)(mu-Cl)Cl](2) (Cp = cyclopentadienyl, Cp* = pentamethyl-Cp, and C10H16 = 2,7-dimethylocta-2,6-dicne-1,8-diyl) produce ester-functionalized poly(aroxycarbonylphenylene) (PACP) with high molecular weights (M-w up to 200 x 10(3)) in good yields (up to 90%). Topological structure of the polymer product can be hyperbranched or crosslinked, tunable by the extent of triple-bond conversion. The degree of branching (DB) of the PACP is similar to 0.76, much higher than those of the "conventional" hyperbranched polymers (DB similar to 0.5). The PACP exhibits high transparency in the visible spectral region. It works as a Molecular container, effectively trapping small molecules in its cavities with the aid of hydrogen-bond interaction. The polymer undergoes a bursting hydrolysis reaction, and the collapse of its hyperbranclied skeleton rapidly releases the loaded molecules, showing promise as a drug carrier for missile therapy.

Abstract:The preparation of a novel chiral 2,2 '-bis(MeO-PEG-supported)- 6,6 '-bis(diphenylphosphanyl)biphenyl (MeO-PEG-Bi-phep) ligand is described. The derived ruthenium complex catalyzes the hydro-enation of beta-ketoesters in up to 99% yield and 99% ee under atmospheric pressure. The accelerating effects exerted by the PEG linkage are dramatic when compared to the unsupported analogue, MeO-Biphep-RuBr2. Furthermore, the catalyst can be recovered easily and the recycled catalysts were shown to maintain their efficiency in two consecutive runs, albeit with declining activity. One of the products, (S)-ethyl-3-hydroxy-3-phenylpropanoate, is useful in the preparation of (S)-fluoxetine.

Abstract:Homogeneous chiral catalysts usually show higher catalytic activities than corresponding heterogeneous chiral catalysts, because of their easy interaction between catalytically active sites with reactant molecules. We demonstrate here superhydrophobic, chiral, and mesoporous catalysts (TsDPEN-Ru) synthesized from copolymerization of N-p-styrenesulfonyl-1,2-diphenylethylenediamine (V-TsDPEN) with divinylbenzene and loading of Ru species exhibiting much higher activities in asymmetric transfer hydrogenation (ATH) of ketones in aqueous solution than corresponding homogeneous chiral catalyst. This phenomenon is strongly related to the unique features of high enrichment for the reactants in superhydrophobic TsDPEN-Ru catalysts due to their good wettability, as well as easy transfer of product from the catalyst into water phase. Abstract:We successfully extended our Rh/bisphosphine-thiourea (ZhaoPhos) catalytic system to asymmetric hydrogenation of alpha,beta-unsaturated N-acylpyrazoles affording products with high yields and excellent enantioselectivities (up to 97% yield, 99% ee). The pyrazole moiety played an important role in providing H-bond acceptor sites, which is critical for achieving high reactivities and enantioselectivities.

Abstract:A novel enantioselective Tsuji-Trost-type cross coupling reaction between gem-difluorinated cyclopropanes and N-unprotected amino acid esters enabled by synergistic Pd/Ni/chiral aldehyde catalysis is presented herein. This transformation streamlined the diversity-oriented synthesis (DOS) of optically active α-quaternary α-amino acid esters bearing a linear 2-fluoroallylic motif, which served as an appealing platform for the construction of other valuable enantioenriched compounds. The key intermediates were confirmed by HRMS detection, while DFT calculations revealed that the excellent enantioselectivity was attributed to the stabilizing non-covalent interactions between the Pd(II)-π-fluoroallyl species and the Ni(II)-Schiff base complex.

Abstract:Attachment of the proline-derived ligand 5 to two types of polymers, i.e. aminomethylated polystyrene resin and respectively MeO-PEG, was achieved via a succinyl linkage from the 4-hydroxy group. Both of the supported ligands (7 and 8) were proved to be highly active with good enantioselectivity (up to 90% ee) for the catalyzed asymmetric diethylzinc addition to aldehydes. The insoluble polymer-supported catalyst could be easily recovered and was reusable for several consecutive catalytic runs without significant loss in enantioselectivity.

Abstract:Despite paramount applications of chiral trifluoromethylated compounds in medicinal chemistry and materials science, limited strategies have been developed for catalytic asymmetric synthesis of such valuable fluorinated structures. Here, we report a nickel catalyzed enantioselective dicarbofunctionalization of inexpensive industrial chemical 3,3,3-trifluoropropene (TFP) with readily available tertiary alkyl and aryl iodides. The reaction overcomes the β-F elimination side reaction of TFP, and proceeds efficiently under mild reaction conditions. The protocol possesses advantages, such as synthetic convenience, high enantioselectivity, and excellent functional group tolerance, providing rapid and straightforward access to chiral trifluoromethylated compounds of medicinal interest.

Abstract:The asymmetric hydrogenation of 2,4-pentadien-1-ones has been achieved by using trans-RuCl2[(R)-XylylSunPhos][(R)-Daipen] as a catalyst under basic conditions. This hydrogenation demonstrated exclusive C1-carbonyl selectivity, and thus the conjugated 2,4-diene motifs remained untouched, which provides a synthetically useful method for various chiral 2,4-pentadien-1-ols.

Abstract:The first example of highly enantioselective fluoroarylation of gem-difluoroalkenes with aryl halides is presented by using a new chiral sulfinamide phosphine (Sadphos) type ligandTY-Phos. N-Me-TY-Phoscan be easily synthesized on a gram scale from readily available starting materials in three steps. Salient features of this work including readily available starting materials, good yields, high enantioselectivities as well as broad substrate scope make this approach very practical and attractive. Notably, the asymmetric synthesis of an analogue of a biologically active molecule is also reported.

Abstract:This work reports the mild and efficient Ru-catalyzed trifluoroisopropylation of arenes using 2-bromo-1,1,1-trifluoropropane. Various bioactive molecules, such as purine and nucleoside derivatives, were well-suited for this transformation, affording the corresponding products in moderate-to-good yields. This method provides an efficient strategy for synthesizing trifluoroisopropyl molecules for drug discovery.

Abstract:A series of fluorinated dendritic chiral ligands have been designed and synthesized. These fluorinated dendrimers are capable of forming a well-defined semi-rigid structure, which was revealed to play a vital role in the ruthenium(II) bifunctional catalyst for asymmetric transfer hydrogenation of prochiral ketone substrates. In contrast to the classical non-fluorinated dendrimer carrier, both NMR and DFT study exhibit that the introduction of fluorine atoms leads to considerable intramolecular weak interactions such as π–π stacking and hydrogen bonding interactions, which make the dendritic backbone exist with a semi-rigid structure in the catalyst. This influences the performance of the catalytic center in terms of the stability and reusability. This concept was employed as a strategy to design a new Ru(II)-complex catalyst Ru-G-2′-F, which demonstrated obviously improved recycling ability up to fifteen times. Significantly enhanced activity, high enantioselectivity and outstanding recycling ability have also been achieved even at high reaction temperature with Ru-G-2-F.

Abstract:2,3-Dihydrobenzofurans are crucial building blocks in the synthesis of natural products and pharmaceutical molecules. However, their asymmetric synthesis has been a long-standing formidable challenge so far. In this work, we developed a highly enantioselective Pd/TY-Phos-catalyzed Heck/Tsuji-Trost reaction of o-bromophenols with various 1,3-dienes, allowing expedient access to chiral substituted 2,3-dihydrobenzofurans. This reaction features excellent regio- and enantiocontrol, high functional group tolerance, and easy scalability. More importantly, the demonstration of this method as a highly valuable tool for the construction of optically pure natural products (R)-tremetone and fomannoxin is highlighted.

Abstract:The direct diastereo‐ and enantioselective Ru(II)‐ catalyzed cyclopropanation reaction of alkenes with ((2‐diazo‐1,1‐difluoroethyl)sulfinyl)benzene was described. The desired enantioenriched PhSO2CF2‐cyclopropanes were obtained in high yields with high diastereo‐ and enantioselectivities in the presence of a chiral ruthenium complex Ru(II)‐Pheox (17‐94% yields, >20:1 d.r. and 83:17‐98:2 e.r.). The robustness of the approach was showcased and This methodology gave access to chiral 1,2‐trans difluoro‐methylcyclopropanes.
