Abstract:Fatty acid metabolism is responsible not only for oilseed metabolism but also for plant responses to abiotic stresses. In this study, three novel genes related to fatty acid degradation designated GhACX, Gh4CL, and GhMFP, respectively, were isolated from Gossypium hirsutum acc. TM-1. The phylogenetic analysis revealed that amino acid sequences of GhACX and GhMFP have the highest homology with those from Vitis vinifera, and Gh4CL has a closer genetic relationship with that from Camellia sinensis. Tissue- and organ-specific analysis showed that the three genes expressed widely in all the tested tissues, including ovules and fiber at different developing stages, with expressed preferentially in some organs. Among them, GhACX showed the most abundant transcripts in seeds at 25 d post anthesis (DPA), however, GhMFP and Gh4CL have the strongest expression level in ovules on the day of anthesis. Based on real-time quantitative RT-PCR, the three genes were differentially regulated when induced under wounding, methyl jasmonate (MeJA), cold, and abscisic acid (ABA) treatments. The characterization and expression pattern of three novel fatty acid degradation related genes will aid both to understand the roles of fatty acid degradation related genes as precursor in stress stimuli and to elucidate the physiological function in cotton oilseed metabolism.

Abstract:A pair of PCR primers was designed based on the conserved sequences of WD40 family gene of different varieties. The normolization library was screened by PCR methods, the cDNA insert size of positive clones were analyzed by PCR method. A full-length cDNA of WD40 (GenBank accession number: EU219610) in cotton was obtained from sequencing. This gene is 1 796 bp in length, containing an open reading frame encoding 274 amino acids and a stop codon from 35th to 860th position. The bioinformatics characterization indicates that the protein is encoded by WD40 domain. pI and molecular weight of the protein encoded were predicted to be 4.24 and 29 kDa, respectively. The yielded gene was accondingly named as GDRP1. RT-PCR analysis showed that the expression of GDRP1 varied during the gland formation process. These results will be helpful for our further study on the gland formation of cotton.

Abstract:bHLH is a superfamily in plant kingdom,whose members participate in the processes of plant growth,development,and stresses resistance.But up to now,the functions of bHLHs in cotton are less studied and reported.This paper took G ossypium hirsutum as material,seperates and clones gene-GhbHLH4,and carried out initial analysis on its function.The results showed that there are abundant stress-response relevant cis-elements in the promoter region of gene GhbHLH4.The leaves and roots of transgenic GhbHLH4 Arabidopsis plants were wider and longer than WT.Transgenic GhbHLH4 plants grew normally under long-time treatment of 150 mmol/L NaCl.The results of this study has laid a foundation for thorough understanding the molecular mechanism of salt tolerance in GhbHLH4 gene.

Abstract:Cotton fibers are single-celled seed trichomes of major economic importance.Many important genes are expressed during cotton fiber development.DISTORTED2 encodes the Arabidopsis homolog of the ARPC2 subunit of the ARP2/3 complex,which binds to the sides of existing actin filaments and efficiently nucleates new filaments.The cytoskeleton arrangement processes in the cotton fiber initial and elongation.In this study,the Gossypium homology of the ARPC2 subunit named as GhDIS2 was isolated from the TM-1 ovule on the day of flowering.GhDIS2 contained 975 bp open reading frame,encoding a polypeptide containing 324 amino acids.GhDIS2 was expressed constitutively in every tissue with different expression levels,with the peak in 12 days post anthesis (DPA) fiber cells and low expression levels in 18,24 DPA fiber cells,roots and stems.Southern blotting showed that GhDIS2 had two copies in allotetraploid cotton genome.A shuttle vector contained GhDIS2 cDNA sequence was transformed into fission yeast.GhDIS2 led to a significant increase in cell length and width.These results indicated that GhDIS2 had the same function to DIS in other plants.

Abstract:Brassinosteroids （BRs） are an important class of plant steroidal hormones that are essential in a wide variety of physiological processes. To determine the effects of BRs on the development of cotton fibers,through screening cotton fiber EST database and contigging the candidate ESTs,a key gene （GhDWF1） involved in the upstream biosynthetic pathway of BRs was cloned from developing fibers of upland cotton （Gossypium hirsutum L.） cv. Xuzhou 142. The full length of the cloned cDNA is 1 849 bp,including a 37 bp 5′-untranslated region,an ORF of 1 692 bp,and a 120 bp 3′-untranslated region. The cDNA encodes a polypeptide of 563 amino acid residues with a predicted molecular mass of 65 kD. The deduced amino acid sequence has high homology with the BR biosynthetic enzyme,DWARF1/DIMINUTO,from rice,maize,pea,tomato,and Arabidopsis. Furthermore,the typical conserved structures,such as the transmembrane domain,the FAD-dependent oxidase domain,and the FAD-binding site,are present in the GhDWF1 protein. The Southern blot indicated that the GhDWF1 gene is a single copy in upland cotton genome. RT-PCR analysis revealed that the highest level of GhDWF1 expression was detected in 0 DPA （day post anthesis） ovule （with fibers） while the lowest level was observed in cotyledon. The GhDWF1 gene presents high expression levels in root,young stem,and fiber,especially,at the fiber developmental stage of secondary cell wall accumulation. Moreover,the expression level was higher in ovules （with fibers） of wildtype （Xuzhou 142） than in ovules of fuzzless-lintless mutant at the same developmental stages （0 and 4 DPA）. The results suggest that the GhDWF1 gene plays a crucial role in fiber development.

Abstract:The quality of cotton fiber is determined by the fiber development. Many factors affect fiber development of which hormones play an important role. Gibberellic acid (GA) is necessary for fiber elongation in cultured fertilized ovules of cotton (Gossypium barbadense), but the functional mechanism of GA on the promotion of fiber elongation is not completely clear. Our study indicated that the initiation and elongation of fiber are mediated by the application of GA to cultured ovules. Plant responses to GA are mediated by DELLA proteins. We isolated one DELLA-like protein gene, designated GbGI from G. barbadense. GbGI has all the typical motifs of DELLA proteins. Over-expression of GbGI cDNA in Arabidopsis plants resulted in the dwarf phenotype, indicating that GbGI functions as a GA suppressor. Real time RT-PCR analysis showed that GbGI was predominantly expressed in cotton fibers at late developing stage, indicating an important role that GbGI plays in cotton fiber elongation.

Abstract:[Objective] The WRKY transcription factors modulate various biological processes,including plant growth,development,and responses to various environmental stresses.This study aimed to clarify the role of WRKY transcription factors related to cotton fiber development.[Method] The GbWRKY40 gene,which encodes a WRKY transcription factor,was isolated from cotton (Gossypium barbadense L.).The obtained sequence was analyzed for homology with genes from other cotton species using the DNAMAN7 program.The sequences of the homologous genes were aligned with Clustalx1.83.Additionally,the GbWRKY40 expression pattern was analyzed by a quantitative real-time polymerase chain reaction.Furthermore,a yeast expression vector was constructed and inserted into yeast strain AH109 cells to investigate the transcriptional activity of GbWRKY40.[Result] Full-length GbWRKY40 cDNA contains 1713 nucleotides,including a 942 bp open reading frame,a 261 bp 5'-untranslated region,and a 510 bp 3'-untranslated region.The open reading frame was predicted to encode a 313 amino acid protein with a relative molecular mass of 34.138 × 103 and an isoelectric point of 8.46.The genomic GbWRKY40 sequence was observed to comprise five exons and four introns.The predicted GbWRKY40 protein includes a WRKY domain consisting of approximately 60 amino acids,including the conserved WRKYGQK sequence and a zinc-finger motif (C-X5-C-X23-H-X1-H).These characteristics indicated that GbWRKY40 belongs to group Ⅱa of the WRKY family.Moreover,GbWRKY40 contains three putative nuclear localization signals.GbWRKY40 is highly homologous to GhWRKY40.Expression analyses involving a quantitative real-time polymerase chain reaction revealed that GbWRKY40 is highly expressed in cotton roots and fibers at 25 days post anthesis.Transcriptional activation assay results suggested that the function of the GbWRKY40 transcription factor is unrelated to the activation of transcription.[Conclusion] These results imply that GbWRKY40 may be involved in regulating secondary cell wall development in cotton fibers.

Abstract:A new full-length cDNA clone was isolated from the fiber cDNA library of Gossypium hirsutum L. The cDNA,designated GhbZIP, encoded a polypeptide of 645 amino acids. GhbZIP protein had the structure characteristics of plant bZIP proteins,including two conserved domains, DUF630 and DUF632 with unknown functions, a proline-rich domain and a phenylalanine-rich domain. Meanwhile, the protein contained a leucine zipper-like motif in DUF632. The hydropathy analysis showed that the GhbZIP was a membrane protein. GhbZIP gene was preferentially expressed in ovule and fiber cells since three days post-anthesis, as indicated it might be involved in the transcription regulation of genes during cotton fiber elonga-tion.

Abstract:In this research, one 1156 bp cDNA containing full open reading frame and encoding a novel 24-kDa protein with four tandem WD repeat motifs was cloned from cotton, therefore was named GhWDR and the GenBank accession number is AY870657. By search of GhWDR cDNA and amino acid sequences in the database, we found that GhWDR and OSJNBa0003G23.2 from Oryza sativa show 90% sequence identity and 84% identity to WD-repeat protein from Arabidopsis thaliana, and also has high sequence identity to other WD repeat proteins, most of which are similar to Pop3 from fission yeast (accession number T39922) and Lst8p from Saccharomyces cerevisiae (accession number NP014392). Therefore, we proposed that GhWDR could act in some cellular processes as pop3 or LST8 does. In addition, the expression of GhWDR in various tissues was studied by RT-PCR, and it is expressed in all of the studied tissues, but the level of expression is low in the leaves when compared to that of other tissues.

Abstract:A new member of the WD repeat protein family, named GhWD40, was cloned from a near-isogenic line for glands in cotton. It has 2629 bp cDNA and a complete opening reading frame (ORF) of 1239 bp, containing the initial code (ATG) and terminal code (TAG); there is a 1061 bp non-coding sequence at the 5′-end, and a 329 bp non-coding sequence at the 3′-end, including the poly(A) sequence (accession number: JN714279). The predicted protein of the complete ORF comprised 412 amino acids with a calculated molecular mass of 47.1 kDa and an isoelectric point of 8.88. Protein domain scanning showed that the novel protein has five wd40 motifs and belongs to the WD40 family. From a search for GhWD40 cDNA and amino acid sequences in the database, it has 77% sequence identity and was 90% sequence positive with the WD-40 repeat protein from Trifolium pratense (accession number BAE71307.1), and 80% sequence identity and 89% sequence positivity with the ribosome biogenesis protein bop1 from Ricinus communis (accession number XP 002529002.1). We propose that GhWD40 may play the same role as bop1. In addition, expression of GhWD40 in near-isogenic lines 11 and 3 (with and without glands, respectively) was studied by quantitative RT-polymerase chain reaction, and the level in near-isogenic line 11 was higher than that in near-isogenic line 3, suggesting that GhWD40 may be related to gland formation.

Abstract:Transport inhibitor response 1 ( TIR1 ), as an F-box family protein, is a nucleus receptor of auxin in plants. It is essential in the auxin signal transduction pathway and the expression of auxin-responsive genes. As the most important hormone in plants, auxin has a great influence on the ovule and fiber development of cotton ( Gossypium hirsutum L.). Studies on the effect of auxin signal transduction pathway on cotton fiber development can fundamentally solve the problem of fiber yield and quality. The cDNA sequence of G. hirsutum TIR1 was obtained through homologous cloning and rapid-amplification of cDNA ends (RACE). The homology between GhTIR1 and that in Arabidopsis thaliana was 64.8%. To study the functions, subcellular localization, and the specific localization of GhTIR1 sequence, we constructed the fusion expression vector of GhTIR1 and its N-terminal or C-terminal deletion mutant genes with GFP . By transiently transforming Arabidopsis protoplasts and stably transforming Nicotiana tabacum BY-2 cell lines, GhTIR1 was localized in the nucleus through the N-terminal 1–197 amino acid sequence. The expression patterns of GhTIR1 in different organs and ovule and cotton fiber cells at different developmental stages showed that GhTIR1 was highly expressed in the initial and elongation stages of cotton fiber development. These results indicate that GhTIR1 plays an important role in the initiation and elongation of cotton fibers, especially the formation of primary fibers of cotton fiber cells.

Abstract:In planta, a vital regulatory complex, MYB-basic helix-loop-helix (bHLH)-WD40 (MBW), is involved in trichome development and synthesis of anthocyanin and proanthocyanin in Arabidopsis. Usually, WD40 proteins provide a scaffold for protein-protein interaction between MYB and bHLH proteins. Members of subgroup 9 of the R2R3 MYB transcription factors, which includes MYBMIXTA-Like (MML) genes important for plant cell differentiation, are unable to interact with bHLH. In this study, we report that a cotton (Gossypium hirsutum) seed trichome or lint fiber-related GhMML factor, GhMML4_D12, interacts with a diverged WD40 protein (GhWDR) in a process similar to but different from that of the MBW ternary complex involved in Arabidopsis trichome development. Amino acids 250-267 of GhMML4_D12 and the first and third WD40 repeat domains of GhWDR determine their interaction. GhWDR could rescue Arabidopsis ttg1 to its wild type, confirming its orthologous function in trichome development. Our findings shed more light towards understanding the key role of the MML and WD40 families in plants and in the improvement of cotton fiber production.

Abstract:Abstract Auxin response factors (ARFs) are a class of transcription factors that regulate the expression of auxin-responsive genes and play important functions in plant growth and development. To understand the biological functions of the auxin response factor GhARF2 gene in upland cotton, the coding sequence (CDS) of GhARF2 gene was cloned, and its protein sequence, evolutionary relationship, subcellular localization and expression pattern were analysed. The results showed that the full-length CDS of GhARF2 gene was 2,583 bp, encoded 860 amino acids, and had a molecular weight and an isoelectric point of 95.46 KDa and 6.02, respectively. The GhARF2 protein had multiple phosphorylation sites, no transmembrane domain, and secondary structures dominated by random coils and α-helices. The GhARF2 protein has 3 conserved typical domains of ARF gene family members, including the B3 DNA binding domain, the Auxin_resp domain, and the Aux/IAA domain. Phylogenetic analysis revealed that ARF2 proteins in different species were clustered in the Group A subgroup, in which GhARF2 was closely related to TcGhARF2 of Theobroma cacao L. (Malvaceae). The subcellular localization results showed that the GhARF2 protein was localized in the nucleus. Analysis of tissue expression pattern showed that the GhARF2 gene was expressed in all tissues tested, with the highest expression level in sepal, followed by leaf, and the lowest expression level in fiber. Further stress expression analysis showed that the GhARF2 gene was induced by drought, high-temperature, low-temperature and salt stress, and had different expression patterns under different stress conditions. These results established a foundation for understanding the functions of GhARF2 and breeding varieties with high-stress resistance in cotton.

Abstract:This study is to isolate full-length cDNA of a key enzyme gene GbHCT related to lignin metabolism in cotton(Gossypium barbadense L.),and analyze the expression of GbHCT in the different developing stages of cotton’s fiber. The primers were designed according to the HCT sequence in transcriptome data of sea island cotton,then the gene was cloned using RT-PCR,and the cDNA sequence and the amino acid sequence of GbHCT were analyzed by bioinformatics methods. The expressions of gene GbHCT in developing fibers of cotton were analyzed from the transcriptome data of sea island cotton. A gene encoding HCT,designated as GbHCT,was isolated from G. barbadense. The full-length of the GbHCT consisted of a 1,311 bp open reading frame(ORF)encoding 436 amino acids. The molecular weight of protein was 48.58 kD,with an isoelectric point(pI)of 6.03. The deduced amino acid sequence of the GbHCT had two conservative motifs HTLGD and DFGWG. The deduced amino acid sequence had a high homology with HCT from Gossypium hirsutum,Gossypium arboretum and Gossypium raimondii,however,it showed 55.7%-60.7% identities with HCTs of other plants. Analyzing the transcriptome data of sea island cotton revealed that gene GbHCT was expressed in all different development stages of fiber,and the peak of expression reached in 5 days post anthers (DPA)fiber cells,suggesting that this gene may be involved in fiber development.

Abstract:Cytokinins are plant hormones that play crucial roles in plant growth and development. Cytokinin dehydrogenase (CKX), regarded as a main negative regulator in cytokinin metabolism in plants, irreversibly degrades cytokinins into adenine/adenosine moiety. A CKX homologous gene, designated GhCKX , was cloned from upland cotton ( Gossypium hirsutum L.). Transgenic tobacco plants over-expressing GhCKX showed a typical cytokinin-deficient phenotype, while CKX-silenced tobacco plants exhibited cytokinin over-producing phenotype. Tissue specifically enhancing the expression of GhCKX in the ovule epidermis of transgenic cotton led to a significant decrease of trans-zeatin and trans-zeatin riboside contents in the ovule. The decline of cytokinins resulted in a significant decrease in fiber initials on a single ovule. Our results indicate that GhCKX encodes a functional CKX, and cytokinins may be required for the initiation of cotton fiber cells.

Abstract:Background: Heat shock transcriptional factors (Hsfs) play important roles in the processes of biotic and abiotic stresses as well as in plant development. Cotton (Gossypium hirsutum, 2n = 4x = (AD)(2) = 52) is an important crop for natural fiber production. Due to continuous high temperature and intermittent drought, heat stress is becoming a handicap to improve cotton yield and lint quality. Recently, the related wild diploid species Gossypium raimondii genome (2n = 2x = (D-5)(2) = 26) has been fully sequenced. In order to analyze the functions of different Hsfs at the genome-wide level, detailed characterization and analysis of the Hsf gene family in G. hirsutum is indispensable.Results: EST assembly and genome-wide analyses were applied to clone and identify heat shock transcription factor (Hsf) genes in Upland cotton (GhHsf). Forty GhHsf genes were cloned, identified and classified into three main classes (A, B and C) according to the characteristics of their domains. Analysis of gene duplications showed that GhHsfs have occurred more frequently than reported in plant genomes such as Arabidopsis and Populus. Quantitative real-time PCR (qRT-PCR) showed that all GhHsf transcripts are expressed in most cotton plant tissues including roots, stems, leaves and developing fibers, and abundantly in developing ovules. Three expression patterns were confirmed in GhHsfs when cotton plants were exposed to high temperature for 1 h. GhHsf39 exhibited the most immediate response to heat shock. Comparative analysis of Hsfs expression differences between the wild-type and fiberless mutant suggested that Hsfs are involved in fiber development.Conclusions: Comparative genome analysis showed that Upland cotton D-subgenome contains 40 Hsf members, and that the whole genome of Upland cotton contains more than 80 Hsf genes due to genome duplication. The expression patterns in different tissues in response to heat shock showed that GhHsfs are important for heat stress as well as fiber development. These results provide an improved understanding of the roles of the Hsf gene family during stress responses and fiber development.

Abstract:【Objective】The objective of this study was to clone new genes related with fiber development in upland cotton. 【Method】Genechips were performed with wild line li1li1 and the Ligon lintless mutant line Li1li1 segregated from the self-crossing progenies of the Ligon lintless mutant line Li1 to isolated genes preferentially expressed during elongating for providing the mechanism of cotton fiber rapid elongation. Two cDNA clones differently expressed were separated from cotton fiber library of elite quality material 7235 during fiber elongation and the second wall synthesis by genechips with probes of the 4 DPA ovule-fiber compound from the wild line li1li1 and the Ligon lintless mutant line Li1li1. 【Result】RT-PCR analysis indicated that the expression quantity of two genes which named GhSAMS (GenBank accession: EF643509) and GhNLP (GenBank accession: EF643508) were less in the wild line li1li1 than that in the Ligon lintless mutant line Li1li1 at 4 DPA. The result of Southern blotting indicated that there are a few copies of both genes in the genome of upland cotton. The authors used the BC1 mapping population derived from the hybridization between the upland cultivar TM-1 and the island cultivar Hai7124, further TM-1 as recurrent parent. GhSAMS and GhNLP were localized on the chromosome D2 and D5, respectively. 【Conclusion】 Two genes related with fiber development in upland cotton were cloned and characterizated with trancription level, genomic copies and genetic location. Future studies including analyses of these proteins function may shed light on the puzzles about the mechanism of cell elongation and excellent quality formation for cotton fibers.
