Effect of puerarin on proliferation of retinal vascular endothelial cells in vitro

Abstract:Objective To investigate the effect of puerarin on proliferation of retinal vascular endothelial cells(RVEC) in vitro.Methods The seventh generation of bovine RVEC was used for this research.RVEC were exposed to different concentrations of puerarin in experimental group and blank medium in control group.The effect of puerarin on the proliferation of RVEC was measured by MTT assay at 24 hours and 48 hours.Results Compared with the control group,the proliferation of RVEC cultured with 0.05 mg ·L-1,0.50 mg·L-1,5.00 mg·L-1,50.00 mg·L-1 and 500.00 mg·L-1 puerarin were obviously at 24 hours and 48 hours.At 24 hours,OD value in 0.05 mg·L-1,0.50 mg· L-1,5.00 mg·L-1,50.00 mg·L-1 and 500.00 mg·L-1 puerarin group were 0.283 ± 0.060,0.337 ± 0.030,0.349 ± 0.030,0.405 ± 0.030 and 0.352 ± 0.020,which were significantly higher than that in control group(0.161 ± 0.06),there were significant differences(all P 0.01) ;At 48 hours,OD values in 0.05 mg·L-1,0.50 mg·L-1,5.00 mg·L-1,50.00 mg·L-1 and 500.00 mg·L-1 puerarin group were 0.371 ± 0.030,0.377 ± 0.040,0.410 ± 0.040,0.400 ± 0.020 and 0.310 ± 0.04,which were significantly higher than that in control group(0.279 ± 0.05),there were significant differences(all P 0.01).Conclusion Puerarin has obvious promoting proliferation effect on RVEC,and the effect is related with the dose of puerarin.
