Abstract:A highly enantioselective oxidative cyclopropanation of 1,6-enynes catalyzed by cationic Au(I)/chiral phophoramidite complexes is presented. The new method provides convenient access to densely functionalized bicyclo[3.1.0]hexanes bearing three contiguous quaternary and tertiary stereogenic centers with high enantioselectivity (up to e.r. 98:2). Control experiments suggest that the quinoline moiety of the β-gold vinyloxyquinolinium intermediate in the reaction plays an important role in promoting good enantioselectivity through a transitional auxiliary effect in the transition state.

Abstract:Dinuclear-gold-catalyzed radical difunctionalization of alkyl bromides with 1,7-enynes has been established via dehalogenation and 1,5-HAT processes. This protocol was used to construct, in a facile and efficient manner, a wide range of cyclopenta[c]quinolines bearing two quaternary carbon centers with good yields (28 examples, up to 84% yield). The good functional group compatibility and gram-scale preparation ability of the reaction proved its synthetic robustness.

Abstract:A tandem coupling-ketooxygenation reaction of readily accessible enediyne-carboxylic compounds with inner alkynes has been developed that utilizes the PdCl2/CuBr2 catalytic system under an O2 atmosphere and assembles a class of isoindolinones and o-acylbenzoic acids. The two oxygen atoms are regioselectively incorporated into enediyne units at the 1- and 6-positions from atmospheric molecular oxygen and H2O, respectively, during the present process. This study uncovered a formal [4C + 2C] benzannulation-diketonization of enediynes and alkynes via a coupling and decoupling strategy.

Abstract:With the well‐documented chemical and biological applications, piperidine and pyridine are among the most important N‐heterocycles, and a new synthetic strategy, especially one with an alternative bond‐forming design, is of general interest. Using the gold‐catalyzed intermolecular condensation of amine and diyne‐ene, we report herein the first example of enantioselective 1,2‐dihydropyridine synthesis through a formal [3+2+1] fashion (up to 95 % yield, up to 99 % e.e.).

Abstract:An enyne cycloisomerization/[5+1] reaction sequence was developed to synthesize tetrahydroisoquinolinones from linear enyne-enes and CO. The first step is a gold(i)-catalyzed enyne cycloisomerization, generating six-membered-ring-fused vinylcyclopropanes. The second step is a rhodium(i)-catalyzed [5+1] reaction of vinylcyclopropanes with CO. This two-step reaction could also be carried out in one-pot without isolating the cycloisomerization product generated from the first step of this sequence.

Abstract:An efficient new method has been developed for the synthesis of ketone‐integrated cyclopentene‐fused indoline derivatives with a high regioselectivity through gold‐catalyzed oxidative cyclo‐coupling of 1,3‐enynes with indoles by using 2,6‐dichloropyridine N‐oxide as the terminal oxidant. A plausible mechanism has been proposed, in which, a cascade process of nucleophilic trapping of the vinyl gold carbene surrogate with indole followed by the subsequent intramolecular cyclization provides the dearomative formal [3+2] cycloaddition product. In addition, a scale‐up reaction and further derivation via cyclopentene's epoxidation & hydrazone derivative's 1,5‐ electrocyclization have been performed.

Abstract:In our previous study, a gold(I)-catalyzed stereoselective formal [4+3]-cycloaddition reaction of 1-(alk-1-ynyl)cyclopropyl ketones with nitrones was found to afford 5,7-fused bicyclic furo[3,4-d][1,2]oxazepines in good to excellent yields with high diastereoselectivity catalyzed by (triphenylphosphine)gold(I) triflate. In order to gain insight into the mechanism of this [4+3] cycloaddition process, an enantioselective version used a MeO-DTBM-BIPHEP derived gold(I) complex as the catalyst, which resulted in the kinetic resolution of the 1-(alk-1-ynyl)cyclopropyl ketones, affording the recovered ketones in high yield with excellent enantioselectivity. An interesting chirality transfer was observed in the gold(I)-catalyzed [4+3] cycloaddition of optically active 1-(alk-1-ynyl)cyclopropyl ketones with nitrones, indicating the reaction undergoes a stereospecific S(N)2-type ring-opening of the cyclopropane.

Abstract:Transition-metal-catalyzed intramolecular carboalkoxylation of alkynes has attracted much attention in the past decades due to its high bond-forming efficiency and atom economy in the rapid assembly of functionalized cyclic compounds. However, compared with the well-established alkyne 1,2-carboalkoxylation, the related 1,1-carboalkoxylation has been far less exploited. Here, we report an unprecedented gold-catalyzed 1,1-carboalkoxylation of ynamides for the generation of exocyclic gold carbenes via exo-cyclization under room temperature. Subsequent 1,2-N-migration and 1,2-H-migration into the gold carbenes lead to the divergent and atom-economical synthesis of valuable tetrahydrofuran-fused 1,4-dihydroquinolines and furoindolines. Moreover, the asymmetric synthesis of these tricyclic N-heterocycles can be achieved by a chirality-transfer strategy, and the possibility of enantioselective cyclization via chiral gold-catalyzed kinetic resolution also emerges.
