Abstract:In this paper, we report a facile synthesis of 2-carbamoyl-2-cyanocyclopropanecarboxylates through a tandem iodosobenzene/tetrabutylammonium iodide-induced oxidative cyclization and a subsequent neighboring group-assisted decarboxylation of the Michael adducts of 2-cyanoacetamides with α,β-unsaturated malonates. This method affords the desired highly functionalized cyclopropanes in moderate to good yields and with excellent diastereoselectivities. In addition, the reaction proceeds smoothly under mild conditions and with good functional group tolerance.

Abstract:A selective synthesis of either 2H-pyran-2-ones (4) or chiral dihydropyranones (6) from the same substrates of 3-bromoenals and 1,3-dicarbonyl compounds upon oxidative N-heterocyclic carbene catalysis is presented. It is shown that the oxidative transformation of 3-bromoenals under NHC catalyst can be well controlled to proceed through two pathways, i.e., elimination of reducible beta-bromide or by an external oxidant 3, allowing the selective generation two sorts of unsaturated acyl azoliums, respectively.

Abstract:Catalytic ring-opening/functionalization of unactive cyclopropanes has proven to be a significant but challenging task in organic synthesis. Herein, we disclose the photoredox and cobalt cocatalyzed ring-opening/acceptorless dehydrogenative functionalization of monodonor cyclopropanes, which provides a promising platform to achieve a sustainable and atom-economic approach to assemble allylic N -acyl-acetal derivatives. The reaction features mild conditions, broad substrate scopes, and excellent functional group compatibilities. The optimized conditions accommodate various cycloalkylamides and primary, secondary, and tertiary alcohols, with applications in late-stage functionalization of pharmaceutically relevant compounds, stimulating the further utility in medicinal chemistry. Abstract:The union of two powerful transformations, directed C-H activation and decarboxylative cross-coupling, for the enantioselective synthesis of vicinally functionalized alkyl, carbocyclic, and heterocyclic compounds is described. Starting from simple carboxylic acid building blocks, this modular sequence exploits the residual directing group to access more than 50 scaffolds that would be otherwise extremely difficult to prepare. The tactical use of these two transformations accomplishes a formal vicinal difunctionalization of carbon centers in a way that is modular and thus, amenable to rapid diversity incorporation. A simplification of routes to known preclinical drug candidates is presented along with the rapid diversification of an antimalarial compound series.

Abstract:1,4-cis-disubstituted cyclic compounds play a pivotal role in pharmaceutical development, offering enhanced potency and bioavailability. However, their stereoselective and modular synthesis remains a long-standing challenge. Here, we report an innovative strategy for accessing these structures via mild conditions employing cyclic 1,3-dienes/alkyl(aryl)halides and amines. This procedure exhibits a wide substrate scope that tolerates various functional groups. The utility of this method is demonstrated in the efficient synthesis of a TRPV6 inhibitor, CFTR modulator and other bioactive molecules. Combined experimental and computational studies suggest that the hybrid palladium-catalyzed radical-polar crossover mechanism is crucial for achieving the exceptional 1,4-syn-addition selectivity (dr > 20:1).

Abstract:The first highly enantioselective catalytic protocol for selective manipulation of tetrol benzylidene acetals through chiral phosphoric acid mediated oxidative desymmetrization is reported. This efficient approach provides a general access to vicinal all-carbon quaternary stereogenic center structural motifs, as well as structures containing vicinal tertiary and quaternary stereocenters enantioselectively. Abstract:Ir-catalyzed asymmetric tandem allylation/iso-Pictet-Spengler cyclization of arylidenaminomalonates with indolyl allylic methyl carbonates was successfully developed, which provided a direct and practical approach to access synthetically useful and biologically active tetrahydro-γ-carboline derivatives bearing multiple functional groups and stereogenic centers in good to high yields and excellent stereoselective control (44%-96% yields, >20:1 dr, 94% → 99% ee). A wide range of substrate generality, easily available substrates, and simple chiral catalytic system displayed great potential practicality of this efficient protocol.

Abstract:Natural products are a vital source of lead compounds in drug discovery. Development of efficient tandem reactions to build useful compounds and apply them to the synthesis of natural products is not only a significant challenge but also an important goal for chemists. Here we describe a tandem C-H oxidation/cyclization/rearrangement of isochroman-derived allylic silylethers, promoted by DDQ and InCl3. This method allows the efficient construction of tricyclic benzoxa[3.2.1] octanes with a wide substrate scope. We employ this tandem reaction to achieve the asymmetric total syntheses of (-)-brussonol and (-)-przewalskine E.

Abstract:A tandem Prins/Friedel-Crafts cascade strategy has been developed for the synthesis of hexahydro-2H-benzo[g]chromene derivatives. Under action of equimolecular amount of BF3 etherate, delta,epsilon-unsaturated alcohols and arylacetaldehydes are coupled to form oxocarbenium ion intermediates, which are captured by the alkene moiety through a favored six-membered transition state. The Prins-cyclization resultant tertiary carbenium ions undergo consecutive intramolecular Friedel-Crafts cyclization to create the tricyclic products. This process enables a tandem formation of three new chemical bonds in one single operation with high yields and stereoselectivity in most cases. When cyclic substituents are involved, a variety of spiro derivatives could also be synthesized successfully, which further proved the robustness of this methodology.

Abstract:Cycloaddition and annulation strategies are among the most powerful methods for creating molecular complexity in organic molecules. In this manuscript, we report a highly site-selective palladium-catalyzed decarbonylative [4+2] cyclization of carboxylic acids with terminal alkynes by a sequential C–C/C–H bond activation. Most notably, this method represents the first use of carboxylic acids as the ubiquitous and underdeveloped synthons for intramolecular cycloadditions by decarbonylative C–C bond cleavage. The method provides a solution to the long-standing challenge of the regioselective synthesis of substituted naphthalenes by cycloaddition. Mechanistic studies show that this reaction occurs through a sequential process involving the formation of key palladacycle by a sequential C–C/C–H bond activation and highly regioselective alkyne insertion enabled by cluster catalysis. Wide substrate scope for both carboxylic acids and terminal alkynes is demonstrated with high functional group tolerance. Moreover, this reaction is scalable and applicable to the synthesis of functionalized molecules featuring bioactive fragments. This reaction advances the toolbox of redox-neutral carboxylic acid interconversion to cycloaddition processes. We anticipate that this approach will find broad application in organic synthesis, drug discovery and functionalized material research fields.

Abstract:An one-pot stereoselective synthesis of 2-aminobenzylidene derivatives from readily available 5-nitro/cyano-activated 2-halobenzaldehydes (2-chloro-5-nitrobenzaldehyde, 2-bromo-5-nitrobenzaldehyde, 2-fluoro-5-nitrobenzaldehyde, 5-cyano-2-fluorobenzaldehyde), the active methylidene compounds (cyanoacetamide and ethyl cyanoacetate), and secondary cycloamines via a novel parallel convergent Knoevenagel-nucleophilic aromatic substitution and nucleophilic aromatic substitution-Knoevenagel condensation cascade approach under mild conditions has been developed with high stereoselectivity and in 52-88% yields.

Abstract:An enantioselective nickel-catalyzed intramolecular reductive cross-coupling of C(sp2) electrophiles and cyano groups is reported. Enantioenriched CN-containing all-carbon quaternary stereocenters are assembled by desymmetrizing cyclization of aryl/alkenyl halide-tethered malononitriles. The use of an organic reductant, (EtO)2MeSiH, is crucial to the enantioselectivity and reactivity. Applications of the method are demonstrated through the synthesis of bioactive molecules and their cyanated analogues and the total synthesis of the natural product diomuscinone. Abstract:An efficient approach to prepare trifluoromethyl-α-carbolines and ester group-substituted α-carbolines via the tandem cyclization reaction of 2-(2-aminophenyl)acetonitriles and trifluoromethyl 1,3-diones or β,γ-unsaturated α-ketoesters was reported. The transformation proceeded smoothly in the presence of catalytic environmental-benign iron salts, which are used to prepare the desired products in moderate to good yields.

Abstract:Sulfocoumarins are key structural motifs in several bioactive molecules. Herein, we describe a simple, one-pot procedure for the synthesis of structurally diverse sulfonocoumarin-3-carboxylates by heating 2-hydroxyaryl aldehydes with an active sulfonyl chloride in the presence of pyridine. The process tolerates numerous functional groups including alkoxy, alkyl, halogen, nitro, and even nucleophilic phenolic hydroxy. Additionally, reactions of 2-hydroxyaryl ketones and 2-methylaminoaryl aldehydes give 4-substituted sulfocoumarins and 1-aza-2-sulfocoumarins, respectively. A gram-scale synthesis and further derivatizations are also reported. The ester group is easily removed via Happer’s decarboxylation.

Abstract:We report here a sequential enantioselective reduction/C-H functionalization to install contiguous stereogenic carbon centers of benzocyclobutenols and cyclobutanols. This strategy features a practical enantioselective reduction of a ketone and a diastereospecific iridium-catalyzed C-H silylation. Further transformations have been explored, including controllable regioselective ring-opening reactions. In addition, this strategy has been utilized for the synthesis of three natural products, phyllostoxin (proposed structure), grandisol and fragranol.

Abstract:Polycyclic conjugated hydrocarbons have acquired increased interests recently because of their potential applications in electronic devices. On metal surfaces, the selective synthesis of four‐ and five‐membered carbon rings remains challenging due to the presence of diverse reaction pathways. Here, utilizing the same precursor molecule, we successfully achieved substrate‐controlled highly selective cycloaddition reactions towards four‐ and five‐membered carbon rings. A 97% yield for four‐membered carbon rings on Au(111), while a 96% yield towards five‐membered carbon rings is achieved on Ag(111). The detailed topological structures of the reaction products are carefully examined by bond‐resolving scanning tunneling microscopy (BR‐STM) imaging with a CO functionalized tip. The underlying mechanism of the novel surface‐directed reaction selectivity is elucidated by extensive density functional theory (DFT) calculations. Our study paves the way for high selective synthesis of polycyclic conjugated hydrocarbons with non‐benzenoid rings.
