Abstract:We aimed to evaluate the association of rs964184 of BUD13-ZNF259 gene with the risk of hemorrhagic stroke (HS). A total of 138 HS cases and 587 controls were recruited for the association of rs964184 of BUD13-ZNF259 gene with the risk of HS. Tm shift PCR was used for genotyping. We were unable to find the association of rs964184 of BUD13-ZNF259 gene with the risk of HS (P>0.05). Significant difference was found in the TG level among the three genotypes (CC: 1.51±1.02; CG: 1.68±1.10; GG: 1.90±1.11, P=0.036). The TG level showed strong correlation with rs964184 genotypes (P=0.010, correlation=0.101). Significantly higher TC, HDL-C, and LDL-C levels were observed in the case group. And no difference was found in the TG, ApoA-I, ApoB. Our case-control study supported the significant association between rs964184 genotype and the blood TG concentration, although we were unable to find association between BUD13-ZNF259 rs964184 and the risk of HS in Han Chinese.

Abstract:Objective: We performed this study to investigate whether the EDNRA gene rs1878406 C > T polymorphism is associated with risk of large artery atherosclerosis (LAA) stroke in the Chinese Han population.Methods: Genotyping of rs1878406 was performed in 1,112 LAA stroke patients and 1,192 healthy controls. Multivariate logistic regression analyses were applied to assess the effect of the rs1878406 C > T polymorphism on susceptibility to LAA stroke.Results: A significant increase of LAA stroke risk was found in the recessive model (TT vs. CC/TC, OR = 1.74, 95% CI = 1.23-2.48, p = 0.002) and co-dominant model (TC vs. CC, OR = 1.06, 95% CI = 0.89-1.27, TT vs. CC, OR = 1.79, 95% CI = 1.25-2.55, p = 0.006). However, the interaction between age and genotypes of rs1878406 was not statistically significant, and no significant interactive effect was observed between the rs1878406 C > T polymorphism and sex (p > 0.05).Conclusion: The rs1878406 C > T polymorphism is associated with increased risk of LAA stroke in the Chinese Han population.

Abstract:Ischemic stroke is caused by an interruption in the flow of blood to the brain and a risk factor for death and disability. Recent genome-wide association studies have identified more than 40 common sequence variants associated with ischemic stroke. However, the results are not always the same in populations with different genetic backgrounds. In the present study, we evaluated a hypothesis that a North Asian population living in a geographic area with unusually harsh environmental conditions would develop unique genetic risks. We investigated the candidate genes for ischemic stroke and risk factors in a Chinese Mongolian population which has not been explored previously. A total of 167 stroke cases and 176 controls were included in the study. Genotyping was performed by amplicon sequencing. The association was detected with single nucleotide polymorphisms (SNPs) located within genes NINJ2 (rs12425791) and ALDH2 (rs2238151) as well as intergenic rs9536591 were significantly associated with ischemic stroke, of which SNP rs12425791 of the NINJ2 gene was the strongest association. ALDH2 gene encodes mitochondrial aldehyde dehydrogenase, involved in the oxidative pathway of alcohol metabolism. Sex, age, body mass index and high blood pressure might be the risk factors. The current work also demonstrated genetic heterogeneity exists between Chinese and other populations. Our study provided the new insights into the genetic basis and environmental factors of ischemic stroke in Mongolian population.

Abstract:Background: Serum C-reactive protein (CRP) has been reported to be associated with risk of ischemic vascular disease including ischemic stroke. Genome-wide association studies have revealed several gene variants related to CRP concentration. Methods: We investigated genetic variants in CRP-related genes associated with ischemic stroke in a nested case-control study with 138 ischemic stroke cases and 276 controls. We sequenced the whole coding region of six CPR-related genes and selected eligible SNPs. Three genetic models (additive, dominant and recessive) were calculated by a multivariable conditional logistic regression to estimate the association between SNPs and risk of ischemic stroke. We also calculated gene?environment interactions by using a crossover analysis. Results: Three out of 10 eligible SNPs were shown to be associated with risk of ischemic stroke. rs1800947 in CRP gene (additive model: OR = 2.08, 95% CI: 1.00?4.23) and rs1169288 in HNF1A gene (additive model: OR = 1.45, 95% CI: 1.03?2.06) were associated with an increased risk of ischemic stroke. rs440446 in APOE gene (additive model: OR = 0.63, 95%CI: 0.44?0.88) was associated with a decreased risk of ischemic stroke. Genetic risk scores models including SC-GRS and OR-GRS both showed a significant association with risk of ischemic stroke. These three SNPs interacted with smoking and red meat intake. Conclusions: Our study showed genetic variants of CRP-related genes were associated with risk of ischemic stroke. Our findings could provide useful data for the etiology of ischemic stroke.

Abstract:Objective To investigate the association of rs2383206 single nucleotide polymorphism(SNP) on chromosome 9p21 and cerebral infarction in northern Chinese Han population. Methods Three hundred and fifty-five cases of patients with early cerebral infarction(C1 group) and 430 cases of surgical patients hospitalized over the same period as controls.PCR-SNP Stream technology was used to genotype rs2383206 polymorphism,and genotyping results were statistically analyzed. Results The frequency of genotype AG of rs2383206 was increased in C1 group,compared with control group,P=0.007).G allele frequency of rs2383206 was also higher in C1 group than control group.After adjusting for confounding factors,we found that,compared with AA homozygotes of the rs2383206,GG/GA genotype has significant effect to increase the risk of cerebral infarction(adjusted OR 1.23,95%CI 1.11-2.05,P=0.009).rs2383206 G-allele increased the risk of large artery atherosclerotic stroke by 2.09 times. Conclusion Rs2383206 is the susceptible locus to CI in Chinese Hans.

Abstract:Genome-wide association studies (GWAS) of ischemic stroke (IS) have been performed on several cohorts of Caucasian or African population and Japanese, resulting in somewhat inconsistent conclusion. We aimed to identify susceptibility loci for IS by exome sequencing in a Chinese Han population. Exome sequencing was used to screen susceptibility loci among 100 cases and 100 matched controls. Significant SNPs from the first stage were verified in up to 3,554 participants from three hospital-based case–control studies. In the initial exome sequencing analysis, rs10489177 in c1orf156 gene located on chromosome 1q24 (p < 1 × 10−8) and rs17118 in XYLB gene located on chromosome 3p21 (p < 1 × 10−6) were found to be significantly associated with IS. In the following validation stage, significantly increased odds ratios were observed in individuals with rs10489177 GG (OR = 2.02, 95 % CI = 1.35–3.03) or rs17118 AA genotype (OR = 1.50, 95 % CI = 1.17–1.91). The rs10489177 GG genotype was associated with significantly increased risk for IS in individuals without hypertension (OR = 2.78, 95 % CI = 1.59–4.86) and in individuals without diabetes (OR = 1.93, 95 % CI = 1.27–2.94). In contrast, the rs17118 AA genotype may significantly increase the risk for IS, particularly for individuals with hypertension (OR = 1.73, 95 % CI = 1.08–2.78) and for individuals without diabetes (OR = 1.52, 95 % CI = 1.17–1.98) or non-smoker (OR = 1.59, 95 % CI = 1.16–2.19). Collectively, our study identified two novel loci (rs17118 and rs10489177) which were associated with an increased risk for IS in Chinese Han populations. Further studies are needed to confirm these associations in other populations and elucidate the biological mechanisms underlying the observed associations.

Abstract:Background and purpose: The first genomewide association study of ischemic stroke in whites has identified multiple susceptibility loci. We confirmed this study by examining associations with ischemic stroke in a Chinese Han population. Methods: Twenty-five common variants were genotyped in a relatively large sample size including 1123 subjects with ischemic stroke cases (thrombosis stroke=716, lacunar infarction=407) and 557 normal control subjects. The association analyses were performed at both single nucleotide polymorphism and haplotype levels. False discovery rate q value method was applied for multiple testing corrections. Results: rs11052413, a intergenic single nucleotide polymorphism, was most significantly associated with ischemic stroke independent of traditional cardiovascular risk factors in additive (OR=1.51, 95% CI=1.19 to 1.92, P=7.4 x 10(-4), q=0.018) and dominant models (OR=1.59, 95% CI=1.20 to 2.08, P=9.2 x 10(-4), q=0.023). In addition, both ZNF650 rs10204475 and intergenic single nucleotide polymorphism rs10486776 were associated with ischemic stroke as well as independent of traditional cardiovascular risk factors in dominant models (OR=1.47, 95% CI=1.12 to 1.96, P=0.005, q=0.040 and OR=1.53, 95% CI=1.15 to 2.02, P=0.003, q=0.036, respectively). No significant results were found in stroke subtype analysis after multiple corrections. Conclusions: Our study confirmed previously reported associations between ischemic stroke and rs11052413, rs10486776, and ZNF 650 rs10204475 in a Chinese Han population. The mechanism whereby the genetic variants exert their effects on ischemic stroke remains to be further elucidated.
Correlation between single nucleotide polymorphism rs6797312 of neuroserpin and progressive ischemic stroke

Abstract:Objective To analyze the association of single nucleotide polymorphism (SNP) rs6797312 within neuroserpin (NSP) with progressive ischemic stroke (PIS) in the Han population of Chongming area of Shanghai. Methods A total of 106 Han patients with cerebral arterial thrombosis were selected as the progress group, and 102 health subjects for physical examination were enrolled as the control group (with hospitalized time limited from April 2013 to January 2015). On admission, data were collected on gender, age, smoking history, history of diseases (hyperten-sion, diabetes, angina pectoris). The polymorphism of SNP locus rs6797312 of NSP gene was detected by Taqman-MGB probe technique, and DNA extraction, SNP genotyping were performed for all the people. Results There was no signifi-cant difference in age [(58 ± 8.8) years vs (57 ± 8.5) years] and sex ratio (65:41 vs 56:46) between the progress group and the control group (P>0.05), while there were significant differences between the two groups in history of smoking [37.74%(40/106) in the progress group vs 20.59%(21/102) in the control group], history of hypertension [48.11%(51/106) vs 28.43%(29/102)], history of diabetes [21.70%(23/106) vs 4.90%(5/102)], history of angina pectoris [14.15%(15/106) vs 43.92%(4/102)], P<0.05. The progress group and control group appeared AA, TT, AT genotypes [6 cases (5.7%), 42 cas-es (39.6%), 58 cases (54.7%) in the progress group;4 cases (3.9%), 37 cases (36.3%), 61 cases (59.8%) in the control group] and A, T two alleles [54 cases (25.5%) and 160 cases (74.5%) in the progress group;46 cases (22.5%) and 158 cases (77.5%) in the control group], with no statistically significant differences (P>0.05). Conclusion SNP site rs6797312 of NSP gene were not associated with the progressive ischemic stroke in the Han population of Chongming ar-ea of Shanghai.

Abstract:Genome-wide association studies have identified 2 single-nucleotide polymorphisms (SNPs) on chromosome arm 9p21, rs10757278, and rs2383207 that confer susceptibility to myocardial infarction. However, these data are mostly from Italian, American Caucasian, South Korean, and Japanese cohorts. This study is the first to investigate whether 6 SNPs (rs10757277, rs10757278, rs10757279, rs1333049, rs1333047, and rs10811656) are associated with acute coronary syndrome (ACS) in a Chinese Han population.We performed a case-control analysis in 359 patients with ACS diagnosed by coronary angiography and 398 non-ACS controls of Han background between April 2007 and January 2008 to determine whether these 6 SNPs were associated with ACS. Exon fragments were genotyped by polymerase chain reaction-restriction fragment length polymorphism.After we adjusted for clinical parameters, we found the rs10757278 GG genotype to be associated with a significantly elevated risk of ACS (odds ratio [OR], 1.91; 95% confidence interval [CI], 1.35-2.68; P = 0.00035), the rs10811656 T allele to be associated with a higher risk of ACS (OR, 1.67; 95% CI, 1.26-2.23; P = 0.0016) than the CC genotype, and the rs1333047 TT genotype also to be associated with a higher risk of ACS (OR, 1.57; 95% CI, 1.15-2.06; P = 0.0052) than the CC and CT genotypes. After 14.2 ± 4.5 months of follow-up, the end-point data were obtained: death (cardiac and noncardiac), nonfatal myocardial infarction, and recurrent angina leading to repeated coronary angiography. We found that the rs10757278 GG genotype was significantly associated with recurrent angina compared with the AA and AG genotypes (P = 0.013).Polymorphisms on 9p21 were associated with ACS in a Chinese Han population. The rs10757278 GG genotype was further associated with adverse cardiac outcomes after ACS.
Association between 12p13 SNPs rs11833579/rs12425791 near NINJ2 gene and ischemic stroke in East Asian population: evidence from a meta-analysis

Abstract:Background: Meta-analysis in European population found no association between rs12425791/rs11833579 and ischemic stroke. Several studies focused on East Asians have evaluated the association between this two SNPs and risk of ischemic stroke, but the results have been inconsistent. The aim of this study was to perform a meta-analysis to investigate a more authentic association between rs12425791 and rs11833579 G>A mutation and ischemic stroke in East Asian population, as well as in Chinese Han population. Methods: Systematic searches of electronic databases Embase, PubMed, Web of Science, and CBM as well as hand searching of the references of identified articles and the meeting abstracts were performed. Study selection, data abstraction and study quality evaluation were independently conducted in duplicate. Statistical analyses were performed using software Stata 11.0. The pooled odds ratios (ORs) with 95% confidence intervals (95%CIs) were performed. Different effects models were used according to the difference in heterogeneity. Publication bias was tested by Begg's funnel plot and Egger's regression test. Results: A total of 4 publications including 7 studies were involved. For rs12425791, significant association was found in allelic model (OR=1.06, 95%CI=1.00-1.11) and dominant model (OR=1.10, 95%CI=1.03-1.18), whereas no evidence of association was found for additive model (OR=1.04, 95%CI=0.93-1.17) and for recessive model (OR=0.99, 95%CI=0.88-1.10). For rs11833579, no evidence of association was found for all genetic models. In the analysis of Chinese Han population, there is lack of evidence for association of ischemic stroke for both SNPs. Conclusions: In summary, our meta-analysis suggests that rs12425791 is significantly associated with ischemic stroke in East Asian population but not Chinese Han population, of which A alleles increase the risk of ischemic stroke, whereas no evidence of association was found for rs11833579 in East Asian population as well as Chinese Han population.
Association Study Between WNK1 Gene Polymorphisms and Ischemic Stroke in Uygur

Abstract:Objective To explore the association between single nucleotide polymorphisms (SNPs) in WNK1 gene and ischemic stroke in Uygur population. Methods Ten tagging single nucleotide polymorphisms (tSNPs) of the WNK1 gene in 295 ischemic stroke patients and 318 control subjects were genotyped,and the association between these tSNPs and ischemic stroke were conducted.The 10 tSNPs were rs3858703, rs11611246, rs7305065, rs1990021, rs34408667, rs12309274, rs1012729, rs956868,rs12828016 and rs953361. They were determined by the Multiplex SNaPshot platform. All data were analyzed using t-test,x2-test and Logistic regression. Linkage disequilibrium and Haplotype were analyzed by Haploview software.Results There were no significant differences between cases (25.6％) and controls(30.0％ ) of the 10 tSNPs in WNK1 gene.When the samples were further stratified according to gender,rs11611246 T allele was found to be associated with a reduced risk of ischemic stroke,with a per-allele OR of 0.448(95％ CI 0.269-0.746,P =0.002) in female cases than in female controls. The significance remained after adjustment for the covariates of age,and for the covariates of age,BMI,cigarette smoking,alcohol drinking,hypertension,diabetes and hyperlipidemia.In addition,no association between the other 9 tSNPs and ischemic stroke were found in Uygur subjects.Conclusion The study reports a new genetic variant,rs11611246,located in the fourth intron of the WNK1 gene,decreasing the risk of ischemic stroke in Uygur population.The T allele might be the protecting factor of ischemic stroke in female Uygur.
Relationship between chromosome 3q22.3 gene polymorphisms and ischemic stroke in Chinese

Abstract:Objective: To investigate the relationship between chromosome 3q22.3 gene polymorphisms and ischemic stroke with its subtypes. Methods: A case-control study was conducted in the study. The case group was 377 consecutive patients with ischemic stroke registered in the Nanjing Stroke Registry Program from June 2012 to December 2012. The patients were classified according to TOAST classification. The control group was 246 age- and sex-matched healthy subjects. The genotypes were determined with the improved multiplex ligase chain reaction. Single nucleotide of polymorphism (SNP) was analyzed, including rs2293252, rs12631438, and rs10935282. The Haploview 4.2 software was used to conduct linkage disequilibrium. Phase software was used to conduct haplotype analysis. Results: Circled digit oneThe rs2293252 and rs10935282 loci genotypes were GG, GA, and AA. The rs12631438 locus genotypes were CC, CT, and TT. Circled digit twoCompared to the control group, there were no significant differences in genotype frequencies and allele frequencies of three SNP loci in the case group (P > 0.05). The frequency of CC genotype on rs12631438 locus (17.7% [25/141 ] vs. 9. 3% [23/246]) and C allele frequency (41.1% [116/282] vs. 32.5% [160/492]) in patients with large artery atherosclerotic (LAA) stroke were higher than those in the control group. There were no significant differences for the comparement of the other stroke subtypes. Multivariate Logistic regression analysis showed that the CC genetype was associated with LAA stroke (OR = 2.03, 95% Cl 1.08-3.91, P = 0.040). Circled digit threeThe haplotype analysis of these three loci found that the frequencies of A-C haplotype on rs2293252-rs12631438 loci in patients with LAA were higher than those in the control group (16.7% vs. 10.4%, OR = 1.81, 95% Cl 1.16-2.80, P = 0.008). The frequencies of A-C haplotype on rs10935282-rs12631438 loci were also higher than those in the control group (9.9% vs. 4.5%; OR = 2.56, 95% Cl 1.41-4.64, P = 0.001). Conclusions: Gene polymorphisms of rs12631438 located in chromosome 3q22.3 region in Chinese Han population may be associated with LAA stroke. The A-C haplotype on rs2293252-rs12631438 and rs10935282-rs12631438 loci may be a risk factor for LAA stroke.

Zinc finger protein 208 (ZNF208) was found to be involved in coronary heart disease, although little information is available about its association with ischemic stroke. We performed the present case-control study to clarify the association between single-nucleotide polymorphisms (SNPs) within ZNF208 and the risk of ischemic stroke in a southern Chinese Han population.Methods A total of 799 subjects (400 cases and 399 healthy controls) were enrolled in the present study. Five SNPs within ZNF208 gene were selected and genotyped using Sequenom MassARRY technology (Sequenom, Inc., San Diego, CA, USA). Data management and statistical analyses were conducted using Sequenom Typer, version 4.0, and a chi-squared test, as well as unconditional logistic regression.Results Statistical results showed that three variants were associated with the risk of ischemic stroke under allele models (rs2188971, rs2188972, rs8103163 and rs7248488). The variant rs2188972 was also associated with the risk of ischemic stroke in a recessive model after adjustment for age and sex. Haplotype analysis suggested that a significant difference existed between the Ars2188972 Trs2188971 Ars8103163 Ars7248488 haplotype and the risk of ischemic stroke, although this disappeared after adjustment for sex and age.Conclusions The results obtained in the present study indicate a potential association between ZNF208 variants and the risk of ischemic risk in a southern Chinese Han population.
The Rs42524 COL1A2 Polymorphism is Associated with Primary Intracerebral Hemorrhage in a Chinese Population

Abstract:Hypertensive arteriosclerosis and amyloid angiopathy account for 78-88% of primary cerebral hemorrhages, and collagen plays a critical role in the pathology of these hemorrhages. The COL1A2 gene on chromosome 7q22.1 has been shown to be associated with intracranial aneurysms. In our study, 393 Han Chinese patients with primary intracerebral hemorrhage (mean age 58.49 ± 9.49 years) and 486 controls (mean age 59.22 ± 8.79 years) underwent genotyping for the rs42524 polymorphism of COL1A2 using polymerase chain reaction-based restriction analysis. Differences in genotypic and allelic frequencies between the cases and controls were evaluated using the χ(2) test. The association between the rs42524 polymorphism and primary intracerebral hemorrhage was also tested using logistic regression analysis with adjustment for risk factors. We found that there was a significant difference between patients with primary intracerebral hemorrhage and control subjects with respect to genotype distribution (χ(2)=16.33, P<0.001) and allelic frequencies (χ(2)=15.01; P<0.001; odds ratio 2.261; 95% confidence interval 1.482-3.451) for the rs42524 polymorphism of COL1A2. Adjustment for risk factors did not change our conclusions. We conclude that the COL1A2 rs42524 polymorphism could be a genetic risk factor for primary intracerebral hemorrhage among Han Chinese.
Association of CDKN2B-AS1 rs1333049 with Brain Diseases: A Case-control Study and a Meta-analysis.

Abstract:Objective: CDKN2B-AS1 polymorphisms were shown to associate with the risk of stroke in European. The goal of this study was to evaluate the contribution of CDKN2B-AS1 rs1333049 to the risk of hemorrhagic stroke (HS) and brain tumor (BT) in Han Chinese. Methods: A total of 142 HSs, 115 BTs, and 494 controls were included in the current association study. The genotyping test was performed using the melting temperature shift method, Results: We failed to validate the association of CDKN2B-AS1 rs1333049 with the risk of brain disease. Significantly higher levels of low density lipoprotein cholesterol (LDL-C) (p=0.027), high density lipoprotein cholesterol (HDL-C) (p<0.001) and total cholesterol (TC) (p<0.001) were found in HSs in the genotype GG/GC carriers, but not the genotype CC carriers (p>0.05). The meta analysis of 10 studies among 133,993 individuals concluded that rs1333049 of CDKN2B-AS1 gene was likely to increase a 16% incidence rate of cerebrovascular disease (CD) among various populations (odds ratio 1.16, 95% confidence interval 1.08-1.25; p<0.0001, random effect method). Conclusion: Our case control study identified rs1333049 genotypes showed different association with the concentration of the LDL-C, HDL-C and TC in the HS patients, Meta analysis supported the association between rs1333049 and CD risk in various populations, although we were unable to observe association between rs1333049 and the risk of HSs in Han Chinese.
Association of Endoglin Gene Polymorphisms with Cerebral Stroke: a Multiple Center Study of 1414 Cases

Abstract:OBJECTIVE:To investigate the association of 5'TCCCCC3' insertion polymorphism in intron7 and the C1096G allele gene with the risk of cerebral stroke in Chinese population.METHODS:Peripheral blood samples were collected from 1414 Chinese patients with cerebral hemorrhage (n = 376), infarction (n = 629) or lacunar infarction (n = 409) selected from 7 clinical centers in China and 1376 controls Single nucleotide polymorphism (SNP) at the position 1096 was genotyped with polymerase chain reaction (PCR) and restriction fragment length polymorphism assay. PCR and polyacrylamide gel electrophoresis were used to examine the insertion/deletion polymorphism in 187 cases of cerebral stroke and 190 controls.RESULTS:The frequency of allele G at nucleotide 1096 of the cerebral stroke patients was 13.5%, not significantly different from that of the controls (13.3%, chi(2) = 0.827, P = 0.352). There were not significant differences in the distribution of GG and GC genotypes among the patients with cerebral hemorrhage, infarction, and lacunar infarction respectively and the controls. In the 187 cases and 190 controls, only 2 cases of heterozygote of the insertion sequence were found with a frequency of 0.003.CONCLUSION:The SNP at the position 1096 of the Endoglin gene and insertion/deletion polymorphism in 5'TCCCCC3' of intron 7 are not the risk factors of cerebral stroke in Chinese population.
CD36 Genotype Associated with Ischemic Stroke in Chinese Han.

Abstract:OBJECTIVE:CD36 is involved in oxidant stress, hyperlipidemia, and thrombosis in the pathology of stroke. CD 36 single nucleotide polymorphisms (SNPs) were reported to be associated with abnormalities of serum FA, triglyceride level and to increase risk of metabolic syndrome, coronary artery disease and type 2 diabetes. Based on these finding we hypothesized that CD36 is an important candidate gene of stroke; therefore, we set out a case-control study to explore the association of CD36 SNPs with ischemic stroke.METHODS:We enrolled 374 patients with atherothrombotic stroke as cases and 1,013 people without stroke as controls. CD36 rs3211842, rs3211870, rs1761667, rs9784998, and rs10499859 loci were detected by PCR-ligase detection reaction.RESULTS:Only rs1761667 (P=0.042) and rs10499859 (P=0.038) polymorphisms were associated with cases of ischemic stroke. Under a dominant genetic model, logistic regression analysis revealed a 1.34-fold increased risk (95% CI 1.05-1.72) of ischemic stroke with rs1761667 A than non-A carriers (P=0.020); the adjusted odds ratio (AOR) was 1.38 (95% CI 1.06-1.78) after adjusting for the covariates age, gender, body mass index (BMI), cigarette smoking, hypertension, and diabetes. For rs10499859, the risk was increased 1.36-fold for G than non-G carriers (P=0.016), and the AOR was 1.39 (95% CI 1.08-1.81) (P=0.012). The 5 SNPs were in strong linkage disequilibrium. CD36 SNPs may have no association with plasma lipid levels and thromboxane B2 (TXB2) expression.CONCLUSION:CD36 rs1761667 and rs10499859 may indicate genetic susceptibility to ischemic stroke among Chinese Han.

Abstract:To investigate whether natriuretic peptide receptor-C (NPR3) gene polymorphisms were associated with ischemic stroke (IS) and hypertension (a conventional risk factor for stroke), we conducted a case-control study in Chinese Han population. We found that rs696831, located in intron 2, was associated with IS. In addition, we found that rs16890208 and rs700925, in linkage disequilibrium (LD) with each other in intron 3, were associated with hypertension. The A allele of the rs16890208, T allele of the rs700925, and the AT haplotype, derived from rs16890208 and rs700925, increased the risk of hypertension with odds ratios (ORs) of 1.74 (95% CI = 1.23-2.47), 1.72 (95% CI = 1.21-2.42), and 1.54 (95% CI = 1.14-2.08), respectively. Further, we found that rs11745562 and rs2270915, in LD with each other in intron 5 and exon 8, were associated with hypertension. The A allele of the rs11745562 and the G allele of the rs2270915 increased the risk of hypertension with ORs of 1.53 (95% CI = 1.07-2.19) and 1.55 (95% CI = 1.08-2.22), respectively. Therefore, we provided novel evidences that polymorphisms or haplotype in NPR3 gene may influence the risk of IS or hypertension independently in Chinese population.

Abstract:In a hospital based case control study, we investigated the association of cyclin-dependent kinase inhibitor 2A (CDKN2A) gene, cyclin-dependent kinase inhibitor 2B (CDKN2B) gene, and two genetic variants (rs10757274 and rs2383206) on chromosome region 9p21 with ischemic stroke in Chinese Hans. Two polymorphisms in the CDKN2A gene (rs3088440 and rs3731245) and two polymorphisms in the CDKN2B gene (rs3217992 and rs1063192) were selected by using a strategy of tagging single nucleotide polymorphisms (tSNP). We observed significant association of rs2383206 with ischemic stroke. Subjects with the GG/GA genotype of rs2383206 had a 1.51-fold (95%CI 1.11-2.05, p=0.009) increased risk of stroke, compared with those with the AA genotype. In addition, the GG/GA genotypes of rs2383206 and rs3731245 was associated with an increased risk of large vessel subtype and small vessel subtype of ischemic stroke, respectively, with ORs of 2.09 (95%CI 1.30-3.37, p=0.002) and 1.63 (95%CI 1.06-2.51, p=0.026), respectively. In gene-environmental interaction analysis, elevation of ischemic stroke risk was observed among AG+GG genotype carriers who consume alcohol, smoke cigarette, and have hypertension, with adjusted combined ORs of 2.86(1.51-5.41), 4.30(2.38-7.77), and 13.97(7.78-25.07), respectively, compared with low-risk individuals for rs2383206 (GG carriers who did not consume alcohol, smoke cigarette, and without hypertension). We provide evidence that genetic variants on chromosome region 9p21 may implicated in the prevalence of ischemic stroke in Chinese.
