Abstract:Surfactants have long been underestimated in synthetic organic chemistry despite their great potential in promoting solubility, reactivity and selectivity in metal-mediated cross-couplings. In this study the strategy of surfactant promotion was exemplified in copper-catalyzed N-arylation of indoles in water. The different effects of surfactants during the coupling reaction were first explored. The superior promoting effect of the natural zwitterionic surfactant, betaine, was found and attributed to the capability of the formed complex to stabilize the copper intermediate in the catalytic transformation and facilitate transportation of the reactants. The concept of surfactant promotion through complexation is valuable in metal-mediated cross-couplings. This method also exhibited good recycling ability. This study will greatly facilitate exploration of surfactant promotion in metal-mediated cross-couplings.

Abstract:The copper-catalyzed sulfenylation of indoles with aryl iodide and sulfur powder has been investigated both experimentally and theoretically. This protocol provides a direct and facile approach to prepare 3-sulfenylindoles with moderate to excellent yields and good functional-group tolerance. The in-situ IR analysis provided evidence for that NaOAc could promote the synthesis of diphenyl disulfide by the coupling of aryl iodide and sulfur powder. According to DFT calculations, the coupling pathway involving the intermediate N-methyl-3-iodoindole is more favored than the direct coupling by the CH activation of indole. The N-methyl-3-iodoindole was identified as a crucial intermediate in the catalytic cycle.

Abstract:A highly regioselective iodosulfonylation of allenes in the presence of CuI and 1,10-phenanthroline has been developed for the synthesis of various useful ( E)-α-iodomethyl vinylsulfones in moderate to excellent yields. This practical reaction is fast, operationally simple, and in particular, proceeds under very mild conditions to afford the target products with high regio- and stereoselectivity. The selectivity was illustrated by a conceptual DFT analysis.

Abstract:The copper-catalyzed azide-alkyne cycloaddition (CuAAC) reaction is regarded as a prime example of "click chemistry", but the asymmetric click cycloaddition of internal alkynes still remains challenging. A new asymmetric Rh-catalyzed click cycloaddition of N-alkynylindoles with azides was developed, providing atroposelective access to C-N axially chiral triazolyl indoles, a new type of heterobiaryl, with excellent yields and enantioselectivity. This asymmetric approach is efficient, mild, robust and atom-economic, and features very broad substrate scope with easily available Tol-BINAP ligands.

Abstract:A practical copper-catalysed oxidative radical alkylarylation of activated alkenes using AIBN (azobisisobutyronitrile) and related reagents has been developed. This has allowed the general incorporation of nitrile moieties into oxindoles by a cascade addition/C(sp2)–H cyclization process. This protocol demonstrates for the first time that DIAD (diisopropyl azodicarboxylate) can act as a new activator for the C(sp3)–H functionalization of acetonitrile instead of the usual Ag salts. The use of an inexpensive copper salt as the catalyst, the general access to primary and tertiary nitriles, as well as the air- and moisture-tolerant reaction conditions make this protocol a highly attractive approach to cyano-containing oxindoles.

Abstract:A palladium-/copper-catalyzed intermolecular C-H amination reaction of indoles has been developed. This reaction proceeds in good to excellent yields to produce a variety of 2-amino-substituted indoles and exhibits excellent regioselectivity at room temperature. Furthermore, chloroamination of indoles provides a simple method for the construction of C-N and C-Cl bonds in one step.

Abstract:A novel and facile approach to 3-allyl-2-aminoindoles via the copper-catalyzed reaction of 3-diazoindolin-2-imines with allyltrimethylsilane is described. The domino reaction proceeds efficiently in one-pot and shows broad substrate scope with respect to the diazo substrates. Furthermore, the synthesized 3-allyl-2-aminoindoles can be conveniently transformed into 3-allyl-3-hydroxylindolin-2-imines and 3-allyl-3-fluoroindolin-2-imines.

Abstract:We have developed a highly efficient base- and additive-free chemoselective CuO-catalyzed strategy for the O-arylation of 2-oxindoles to synthesize 2-phenoxy-3H-indole and 2-phenoxy-1H-indole derivatives in the presence of diaryl iodonium salts. This method offers a variety of O-arylated oxindoles in good to excellent yields under relatively milder reaction conditions. Furthermore, this methodology was extended for the O-arylation of 2-pyridinone and isoindoline-1-one derivatives as well.

Abstract:Transition metal-catalyzed asymmetric hydrogenation (AH) of unprotected indoles has mainly been applied to alkyl substituted unprotected indoles. However, the challenging aryl substituted unprotected indoles with poor reactivity and enantioselectivity (<= 42% ee) could not be hydrogenated well. In this work, a highly efficient Ir/bisphosphine-thiourea ligand ZhaoPhos catalytic system for the AH of challenging aryl substituted unprotected indoles has been successfully developed for the first time with high reactivity and excellent stereoselective control. Moreover, a series of 2-alkyl-substituted and 2,3-disubstituted unprotected indoles were also well tolerated in this catalytic system. A wide variety of chiral indoline derivatives were obtained in good to high yields with excellent stereoselectivities (75-99% yields, >20:1 dr, and 86-99% ee). The anion-binding activation strategy played an important role in accessing both high reactivity and excellent stereoselectivity, which was formed between the catalyst and unprotected indoles in situ-generating iminium ion with the assistance of Bronsted acid. A possible catalytic mechanism was proposed for this Ir-catalyzed AH according to density functional theory calculations and control experiment results. Readily available substrates, a broad range of substrate tolerance, an efficient chiral catalytic system, and a gram-scale protocol further demonstrated the potential practicality of this methodology. Abstract:2,3'‐Bisindoles with C–C linkages have attracted interest in medicinal chemistry, yet their synthesis is intricate with many steps. Notably, direct C–H/C–H cross‐coupling of non‐directed heteroaromatics remains challenging, often requiring precious metals and oxidants to enhance coupling efficiency. Herein, we present a copper‐catalyzed C–H/C–H cross‐coupling method for N‐substituted indoles without directing groups, facilitated by molecular oxygen under gentle conditions. It showed reasonable functional group compatibility and provided one‐pot access to a variety of 2,3'‐bisindoles derivatives in moderate to good yields.
