Abstract:The pyrrolo[2,3-b] pyridine core structure, a bioisostere of quinolones, is found in several molecules that possess important biological activity. We describe here a new, concise, three-step synthesis of pyrrolo[2,3-b]pyridines starting from L-alanine. A series of 4,7-dihydro-4-oxo-1H-pyrrolo[2,3-b]pyridine-5-carboxylic acid derivatives, which have not been previously reported, were synthesized using this approach.

Abstract:The preparation of a series of 2-pyridinamines and their alkyl derivatives is described.The target compounds are synthesized starting from corresponding 2-cyanopyridines, via incomplete hydrolysis in the presence of hydrogen peroxide in the dilute alkaline mixture of acetone-2% sodium hydroxide solution, and Hoffmann degradation reaction with freshly made NaBrO.This route is of industrial value because of cheap and readily available materials, moderate reaction conditions and convenient operations.

Abstract:A concise and efficient synthetic approach was developed for the synthesis of functionalized chromeno[4,3-b]pyrrol-4(1H)-ones via a one-pot, three-component reaction of 4-aminocoumarin, arylglyoxal monohydrates, and aromatic amines promoted by KHSO4 in toluene under reflux condition. The attractive features of this protocol are operational simplicity, short reaction time, easy purification of products and good to excellent yields.

Abstract:A one-pot two-step reaction of 4-aminocoumarin with arylglyoxal monohydrates and p-toluenesulfonates has been developed, which offers 3-alkoxy-substituted chromeno[4,3-b]pyrrol-4(1H)-ones in moderate to good yields. At first, arylglyoxal monohydrates and 4-aminocoumarin were converted into 3-hydroxychromeno[4,3-b]pyrrol-4(1H)-one intermediates in AcOH under reflux. Then the title compounds in an in situ reaction of the intermediates with p-toluenesulfonates catalyzed by DBU in refluxing toluene were obtained.

Abstract:A novel tandem Blaise reaction for the one-pot synthesis of substituted NH-pyrroles was described. The Blaise reaction intermediate, generated in situ from Reformatsky reagent and nitrile, reacted with arylglyoxals chemoselectively to afford a wide variety of substituted NH-pyrroles in good yields.

Abstract:An efficient and practical method was developed for the synthesis of tetrahydro-1H-pyrrolo[1,2-a]imidazol-2-ones based on the decarboxylative cyclization reaction of alpha-ketoamides and proline. Abstract:The potentially bioactive 2-aryl-thiazolo[4,5-b]pyridines were synthesized via palladium-catalyzed desulfitative cross-coupling reaction between multisubstituted thiazolo[4,5-b]pyridine thioethers and boronic acids. Abstract:Pyrroles are a fundamental class of heterocyclic compounds that play a crucial role in many biologically active natural products and synthetic pharmaceuticals. Their unique electronic properties and structural versatility make them indispensable for the development of molecules with significant therapeutic potential. In this work, we introduce a novel approach to the formation of the pyrrole core, catalysed by L-proline, employing Paal-Knorr synthesis as the key transformation. This innovative method facilitates the efficient construction of the core structure while allowing for the introduction of diverse substituents, leading to the synthesis of over 20 highly functionalized pyrrole derivatives. Building upon this pyrrole core synthesis, we have extended our strategy to the construction of the pyrrolo[2,1-a]isoquinoline scaffold, an essential structural motif present in several alkaloids and pharmacologically active compounds. This scaffold is notable for its biological relevance, particularly in anti-cancer and anti-inflammatory applications. Our method showcases the versatility of the Paal-Knorr synthesis, enabling precise construction of the pyrrolo[2,1-a]isoquinoline core, starting from substituted chalcones. The synthetic route offers a reliable pathway for constructing complex heterocyclic structures, advancing our understanding of pyrrole and isoquinoline chemistry and providing valuable insights for the development of new therapeutic agents.

Abstract:An efficient and concise method is hereby reported for the synthesis of pyrido[3,4- c ]pyridazin-8(7H)-ones from the commercially available material, methyl 4,6-dichloropyridazine-3-carboxylate. The four-step reaction process involves Suzuki coupling, Buchwald coupling, ammoniation, and intramolecular cyclization. The advantage of this route represents the development of a novel C5 site-selective Suzuki monocoupling of 3,5-dichloropyridazine, with an overall yield of over 73 %.

Abstract:Development of facile synthetic methods for various disubstituted pyrroles from pyrrolidine and 2-substituted 1pyrrolines is reviewed based on our recent findings. Various 1,3-disubstituted pyrroles were synthesized by reaction of pyrrolidine and aldehyde heating without any catalyst. On the other hand, 2,3-disubstituted pyrroles were synthesized from 2-substituted 1-pyrrolines by a sequece involving its aldol-type condensation with aldehyde and subsequent base-catalyzed double-bond isomerization. Application of the former method to one-pot synthesis of a pyrrolocyclophane and preparation of N-substituted indoles from indoline and aldehydes is disclosed. Some pyrrole products by the latter method were transformed into dipyrrin derivatives via their pyrrole-2-carbaldehydes, and chemical and structural properties of the dipyrrin derivatives are described.

Abstract:AbstractA general and efficient protocol was developed for the synthesis of polysubstituted pyridines from propargyl amines and unsaturated carbonyl compounds through a tandem condensation/alkyne isomerization/6π 3‐azatriene electrocyclization sequence. This process was found to be applicable to a wide range of readily available substrates (30 examples, up to 95 % yield) and could be readily performed on a preparative (20 g) scale. By taking advantage of this method for late‐stage pyridine incorporation, we successfully completed the collective total synthesis of suveoline, norsuveoline, and macrophylline.
