Multiple-Fingerprint and Spectrum-Effect Relationship of Bioactivity from Codonopsis Pilosula Polysaccharide

Abstract:CPP has been widely concerned for its immunocompetence. In this study, CPPs were prepared from 40 batches of Codonopsis pilosula from 3 varieties in 4 provinces by water extraction and alcohol precipitation method. HPGPC, GC and FT-IR fingerprints were constructed. After the transmittance of FT-IR fingerprint was converted into absorbance, the fingerprints of FT-IR, HPGPC and GC were fused at a low level to form a CPP multiple-fingerprint. Multiple-fingerprint combined with chemometrics methods showed that the polysaccharides of C.pilosula, C.pilosula var. modesta and C.tangshen were clustered respectively, with distinct boundaries. Furthermore, the macrophage proliferation rates were in the order of C.tangshen polysaccharide > C.pilosula var. modesta polysaccharide > C.pilosula polysaccharide, while the polysaccharide yield and content of C.tangshen and C.pilosula var. modesta were significantly higher than that of C.pilosula. The spectrum-effect relationship analysis results showed that the main macrophage proliferative active ingredients of CPP were the components with molecular weight of 9.74×104-1.08×105, rhamnose and the characteristic peaks of 2942.84cm-1, 1027.87cm-1 and 3370.96cm-1. This study systematically analyzed the CPP information, constructed the multiple-fingerprint of CPP, and further clarified the relationship between the multiple fingerprint characteristics of CPP and its bioactivity, which could provide a basis for quality evaluation of CPP.
