Hypoxia-Induced Glioma-Derived Exosomal miRNA-199a-3p Promotes Ischemic Injury of the Peritumoral Neurons in Glioma by Inhibiting the mTOR Pathway: A Novel Ischemic Injury Mechanism in Peritumoral Neurons

Abstract:Background: It has been reported that the peritumoral hypoxic microenvironment could promote the injury of peritumoral neurons in glioma, but underlying mechanisms are unknown. We hypothesized that the exosomes, exosomal miRNAs and mTOR signaling pathway might be involved in hypoxic peritumoral neuronal injury. Methods: Patients’ glioma samples and their radiological data were retrospectively analyzed. HT22 and C6 cell lines were co-cultured in hypoxia system. Hypoxia-induced glioma derived exosomes (HIGDE) and non-HIGDE (NHIGDE) were administrated to primary neuron cells. The target miRNA was identified by performing miRNA microarray analysis. The expression of target miRNA, and mTOR activity were interfered to clarify our hypothesis. Findings : Multimodal radiological images, HE and HIF-a staining of WHO III glioma samples revealed that the peritumoral hypoxic area overlapped with the cytotoxic edema region. In either direct or indirect coculture system, hypoxia could promote HT22 neuron injury; and the growth of HT22 cells was suppressed by C6 cells under hypoxic condition. HIGDE aggravated OGD injury in normal HT22 cells, and miRNA-199a-3p was identified as the target miRNA that induced the hypoxic injury. The upregulation of miRNA-199a-3p, induced by hypoxia-related HIF-1a activation, could aggravate neuronal injury with HIGDE administration by suppressing mTOR signaling pathway. Interpretation: Hypoxia-induced glioma-derived exosomal miRNA-199a-3p can be upregulated by the activation of HIF-1a and is able to increase the ischemic injury of peritumoral neurons by inhibiting the mTOR pathway.
