Abstract:Sialic acid is one of the key constituents in tumor-specific glycans and an important biomarker for a diversity of cancer types. Its content change in the blood serum can be used for early diagnosis of cancer, grading of cancer, and evaluation of therapeutic effect and prognosis. In this paper, we studied and analyzed the terahertz (THz) response of solid sialic acid samples with five concentrations. Based on the characteristics of the peak value among absorption coefficient spectrum, linear fitting is performed to achieve the quantitative detection of sialic acid.

Abstract:In this paper, we propose a discrimination model for abnormal sialic acid content using terahertz time and frequency-domain features of samples in an attempt to explore the significance of human serum sialic acid content in early cancer screening. Specifically, the time-domain spectral features were obtained by applying variational mode decomposition (VMD) for time-domain spectrum preprocessing and composite multi-scale entropy (CMSE) for feature extraction. The frequency-domain spectral features were acquired using principal component analysis (PCA). The model was then constructed on the basis of support vector machine (SVM). To verify the reliability of this discrimination model, we used three different concentrations of albumin solutions to simulate the serum environment in the human body and mixed them with sialic acid solutions of eight different concentrations. The results indicate that our discrimination model achieved an accuracy of 91.01% for the detection of abnormal sialic acid content. This study provides a foundation for the accurate quantitative detection of biomarkers in human serum.

Abstract:The disaccharide galactose-beta 1,3-N-acetylgalactosamine (Gal beta 1,3-GalNAc) attached to serine and/or threonine residues of proteins, also known as the ThomsenFriedenreich (TF) antigen, is highly expressed in various types of human carcinomas. It has been shown to contribute to tumor development, progression, and metastasis. However, current methods have limited power in detecting and imaging TF antigens among a variety of complex cell-surface glycans. Here we describe a tandem enzymatic strategy to detect and label TF antigen disaccharide with high sensitivity and selectivity. We demonstrate that this strategy enables detection of TF antigens on proteins, profiling and identification of unknown TF antigen-modified glycoproteins, and simultaneous labeling of multiple forms of complex glycan motifs on the same cell. This approach expands the capability of glycan labeling to probe the functional role of TF antigens in cancer biology.

Abstract:Tumor genesis is accompanied by glycosylation of related proteins. Glycoprotein is usually regarded as a tumor marker since glycoproteins are consumed remarkably more by the cancer cells than the normal ones. In this paper, the terahertz time-domain attenuated total reflection (ATR) technique is applied to inspect the glycoprotein solution from a concentration gradient of 0.2 mg/ml to 50 mg/ml. A significant nonlinear relationship between the absorption coefficient and the concentrations has been discovered. The influence of the dynamical hydration shell around glycoprotein molecules on the absorption coefficient is discussed and the phenomenon is explained by the concepts of THz excess and THz defect. In order to identify glycoproteins, features are obtained by composite multiscale entropy (CMSE) method and clustered by the K-means algorithm. The results indicate that features extracted by the CMSE method are better than the Principal Component Analysis (PCA) method in both specificity and sensitivity of recognition. Meanwhile, the absorption coefficient and dielectric loss angle tangent are more suitable for qualitative identification. Research shows that the CMSE method has important directive significance for analyzing glycoprotein terahertz spectroscopy. And it has the potential for glycoprotein related tumor markers identification using terahertz technology in medical applications.

Abstract:Human dehydrated normal and cancerous gastric tissues were measured using transmission time-domain terahertz spectroscopy. Based on the obtained terahertz absorption spectra, the contrasts between the two kinds of tissue were investigated and techniques for automatic identification of cancerous tissue were studied. Distinctive differences were demonstrated in both the shape and amplitude of the absorption spectra between normal and tumor tissue. Additionally, some spectral features in the range of 0.2 similar to 0.5 THz and 1 similar to 1.5 THz were revealed for all cancerous gastric tissues. To systematically achieve the identification of gastric cancer, principal component analysis combined with t-test was used to extract valuable information indicating the best distinction between the two types. Two clustering approaches, K-means and support vector machine (SVM), were then performed to classify the processed terahertz data into normal and cancerous groups. SVM presented a satisfactory result with less false classification cases. The results of this study implicate the potential of the terahertz technique to detect gastric cancer. The applied data analysis methodology provides a suggestion for automatic discrimination of terahertz spectra in other applications.

Abstract:At present, many researchers are exploring biological tissue inspection using terahertz time-domain spectroscopy (THz-TDS) techniques. In this study, based on a modified hard modeling factor analysis method, terahertz spectral unmixing was applied to investigate the relationships between the absorption spectra in THz-TDS and certain biomarkers of gastric cancer in order to systematically identify gastric cancer. A probability distribution and box plot were used to extract the distinctive peaks that indicate carcinogenesis, and the corresponding weight distributions were used to discriminate the tissue types. The results of this work indicate that terahertz techniques have the potential to detect different levels of cancer, including benign tumors and polyps.

Abstract:Terahertz wave technique is very prospective in the biomedical application field, where there exists a practical problem that it is hard to register a terahertz time-domain spectroscopy (THz-TDS) detection point and its accurate corresponding position in pathology image. In this paper, complex human gastric tissue is taken as the experimental subject to search for accurate detection approach. This paper uses tissue microarray technology to process paraffin-embedded gastric tissue, and then contrasts the Terahertz absorption coefficient and refractive index spectrum of detection subjects which are divided into registered experimental group and unregistered control group using principal component analysis method. Meanwhile, this paper contrasts the gastric tubular adenocarcinoma THz-TDS discrimination methods using support vector machine and logistic regression respectively. The research result reveals that the detection accuracy brought by tissue microarray technology helps better discriminate tumor samples and normal samples.

Abstract:Terahertz time-domain spectroscopy (THz-TDS) has been applied to medical applications as THz waves are sensitive to biomolecules. In this paper, terahertz time-domain attenuated total reflection spectroscopy (THz-ATR) was used to identify human colon cancer cell line. The refractive indices of cancerous and normal cell lines with six different concentrations were analyzed in the frequency domain from 0.1-1.2THz. t-Distributed Stochastic Neighbor Embedding (t-SNE) and Principal Components Analysis (PCA) methods were adopted to extract the key features from the THz spectrum. Experimental results showed that the features extracted by t-SNE had better recognition accuracy owing to enhanced clustering efficacy. This work may be helpful for detecting human colon cancer cells by using terahertz techniques.
A simple integrated system for rapid analysis of sialic-acid-containing N-glycopeptides from human serum.

Abstract:Terminal sialylation is very important in cancer biology and has been extensively investigated for the discovery of potential clinical biomarkers of cancers. In this study, we presented a novel approach, by using of Ti(IV)-IMAC, to enrich sialic-acid-containing N-glycopeptides for the analysis of terminal sialylation. Compared with conventional method using TiO2, this approach obtained 2.5 times more glycopeptides and glycosylation sites. Then, a simple integrated system combining filter-aided sample preparation, ACN-improved digestion, and Ti(IV)-IMAC enrichment was established for efficient analysis. In this system, protein digestion, glycopeptide enrichment, and deglycosylation were integrated and were performed sequentially in a single filter unit without any need for desalting, lyophilization, or sample transfer procedures. As a result, the number of identifications was improved by 1.5-fold and the total processing time was drastically reduced to only 78 h. By using this system, fast and efficient analysis of human serum sialylated N-glycoproteome was achieved. From only 1 L of human serum, 217 unique glycopeptides and 194 glycosylation sites were successfully identified.

Abstract:A dual gold nanoprobe system was designed for in vivo portable Raman detection of sialic acid (SA) for tumor identification. The dual gold nanoprobe system contained two gold nanoprobes, Au10-DTTC/PEG-PBA and Au40-PEG-SA. Au10-DTTC/PEG-PBA was constructed on a 10 nm gold nanoparticle modified with 3,3 '-diethylthia tricarbocyanine iodide (DTTCI) as the Raman reporter and 3-aminophenylboronic acid (APBA) through a thiol PEG succinimidyl carboxymethyl ester (HS-PEG-NHS) linker for specific recognition of SA. Au40-PEG-SA was constructed on a 40 nm gold nanoparticle modified with SA through HS-PEG-NHS. For in vivo detection of SA, Au10-DTTC/PEG-PBA and Au40-PEG-SA were subsequently injected into tumor xenografted mice with optimal interval and retention times. Through the specific recognition between PBA and SA, the conjugates of Au10-DTTC/PEG-PBA and Au40-PEG-SA formed in the tumor region emitted strong SERS signals of DTTC, which could be detected by a portable Raman detector. This work provides a convenient and portable method to detect SA in tumor xenografted mice, which is useful for family-stay identification and clinical cleavage of tumors.

Abstract:Liver cancer usually has a high degree of malignancy and its early symptoms are hidden, therefore, it is of significant research value to develop early-stage detection methods of liver cancer for pathological screening. In this paper, a biometric detection method for living human hepatocytes based on terahertz time-domain spectroscopy was proposed. The difference in terahertz response between normal and cancer cells was analyzed, including five characteristic parameters in the response, namely refractive index, absorption coefficient, dielectric constant, dielectric loss and dielectric loss tangent. Based on class separability and variable correlation, absorption coefficient and dielectric loss were selected to better characterize cellular properties. Maximum information coefficient and principal component analysis were employed for feature extraction, and a cell classification model of support vector machine was constructed. The results showed that the algorithm based on parameter feature fusion can achieve an accuracy of 91.6% for human hepatoma cell lines and one normal cell line. This work provides a promising solution for the qualitative evaluation of living cells in liquid environment.

Abstract:Terahertz (THz) waves are extremely sensitive to water content and biological composition (proteins, genes, and metabolites), which are the key regulator of cancer development. Hence, the application of THz spectroscopy and imaging is promising in the field of biomedicine, especially in cancer diagnosis. Currently, the diagnosis of cancer patients (including tumor borders, tumor stage, etc.) mainly rely on optical microscope observation of pathological slices. The preparation of the slices is complicated and time consuming, and the microscopic field of view is small, which cannot effectively guide the precise removal of the tumor during the operation in time. To solve this problem, a label-free real-time high-resolution near-field THz spectroscopy imaging technique was developed. In this study, we successfully apply this technique to the oral tissue. Compared with conventional point-by-point scanning terahertz time-domain spectroscopy imaging, this technique can exhibit a real-time imaging rate (as high as 20 frames/s at 512 × 1024 pixels) and a field of view approximately 5 mm in diameter with a spatial resolution up to λ /10 at 1 THz in near-field condition. The experimental results show that the terahertz spectroscopic imaging in this paper can clearly detect and identify not only biological tissues with different components, but also normal and cancerous tissues, as well as tissues with the same composition but different slice directions. Abstract:In this paper, the absorption coefficient spectra of trehalose and cellobiose are measured in the frequency range from 0.4 to 1.8 THz by using THz time-domain spectroscopy. The two disaccharide are common disaccharide in agricultural products containing sugar. The experimental results show that trehalose and cellobiose have some characteristic absorption peaks in the range. The structure and vibrational frequencies of trehalose and cellobiose molecule in the THz range are simulated by the B3LYP algorithm of density functional theory, and the THz characteristic absorption peaks of trehalose and cellobiose molecule are identified by using Gaussian View software. The analysis show that the experimental results tally with the theoretical ones well.The observed absorption peaks in the THz range are mainly caused by the intramolecular and intermolecular vibrations jointly. The work of the paper presents promising methods for the identification of disaccharide and the quality assurance of agricultural products.

Abstract:Terahertz technology is a non-invasive and label-free inspection method, promising for applications in various fields, especially in biomedical diagnosis. Terahertz radiation is capable for cell physiological and pathological status recognition due to its sensitivity to cell composition and structural information of biomacromolecular. In this paper, the transmission properties of glass, polymethyl methacrylate and polypropylene in THz regime were compared and polypropylene was selected as the petri dish material for cell adhesion due to its high transmission of terahertz wave. The time-domain and frequency-domain spectra, refractive index and absorption coefficient of hepatoma cell line Huh7 and normal cell line MIHA were analyzed. The experimental results showed significant distinction between the terahertz characteristic spectra of hepatoma cells and normal cells. This work can be helpful for applying terahertz technology in cancer detection at the cellular level.

Abstract:The rapid and accurate identification of the types and contents of early pathological markers by THz technology are of particular importance for the prevention and treatment of major diseases. Nevertheless, these markers usually contain interference from water and other non-target molecules, resulting in low signal-to-noise ratio (SNR) and making identification and quantitative analysis difficult. Here, based on THz spectroscopy from a high-power THz source radiated by LiNbO3, we perform quantitative and real-time THz detection of mixtures (α-lactose monohydrate and 4-aminobenzoic acid) in liquids. The results demonstrate that the absorption spectra of the aqueous biomolecular mixtures exhibit an accumulation of THz features of each pure product, i.e., the amplitude of the absorption peaks is proportional to the mixing ratio, while the corresponding absorption baseline increases with decreasing concentration. Furthermore, the content of the target substance can be calculated from the linear relationship between the absorption spectra of pure and mixed samples. This technology will support the future application of THz-TDS in early disease diagnosis under complex states and environments.
Terahertz Spectral Analysis of DNA Based Molecule Cytosine and Its Methylated Structure 5-Methylcytosine

Abstract:Most DNA methylation occurs in the cytosine of nucleosides. In this study, the absorption coefficients of cytosine and 5-methylcytosine were measured at 0.1-2.0 THz through terahertz time domain spectroscopy (THz-TDS) without being labeled. Our results showed that these important biomolecules had distinct spectral characteristics within this terahertz band. Moreover, structural optimization and lattice dynamics calculations were performed to determine the lattice vibrational motions of both cytosine crystals and 5-methylcytosine crystals using the density functional theory (DFT) and the properly modeled intermolecular potentials. Typically, the lattice vibrational motion simulated the absorption spectrum which indicated that the terahertz technique was able to detect the absorption spectral features of cytosine and 5-methylcytosine. Hopefully, this study provided the molecular background to detect DNA methylation and lay certain foundation for subsequent cancer detection.

Abstract:Techniques to inspect and analyze human colorectal cancer cell lines by using terahertz time-domain attenuated total reflection spectroscopy (THz TD-ATR) were investigated. The characteristics of THz absorption spectra of two colorectal cancer cell lines DLD-1 and HT-29 in aqueous solutions with different concentrations were studied. Different spectral features were observed compared to normal cell line. Identification results based on different parameters including absorption coefficient, refractive index, real and imaginary parts of complex permittivity, dielectric loss tangent were discussed. This research may be promising for quick and instant inspection of liquid samples by using THz time-domain spectroscopy in medical applications.

Abstract:Detecting cell-surface sialic acids (SAs) is essential for cancer research, as accumulating evidence indicates that SA overexpression is closely related to unusual biopathways, such as tumorigenesis. However, it remains challenging to detect and classify membrane SAs. Here, a highly efficient and convenient two-step tagging strategy is described for live-cell profiling of membrane sialic acids with fluorescence imaging and SERS labelling. The fluorescence of SA-linked dyes indicates the SA distribution, whereas the Raman signals recognize the SA-linked proteins marked by aptamer modified SERS probes. Using the tumor markers MUC-1 and TNC as the model proteins, SAs can be partitioned into distinct space domains via Flu/SERS information on living cell membrane. Importantly, we find that SAs are highly expressed near MUC-1 and TNC on the surface of cancer cells, with subtle differences existing among various carcinoma cell lines, enabling classification of various types of cells. Together, our strategy provides a robust and versatile platform for highly detailed analysis of cell surface saccharide profiles and that it has great potential for optical biosensing and molecular diagnosis.
