However, the fundamental limit for phase noise is scarcely discussed in the literature. In a typically off-axis phase microscope system, the phase noise is limited to a few milliradians using a moderate camera. Common-path QPMs offer much reduced phase noise compared to typical Mach-Zehnder-based systems. However, further scaling down the phase noise becomes difficult. Here we propose a high-sensitivity common-path QPM that promises to reduce the phase noise by a factor of 10 (assuming the mechanical noise is negligible). This is achieved by a specifically designed signal filter, leaving only the subtle phase fluctuations coming from the dynamics sample scattering. By working at photon shot-noise limited detection, we can magnify the subtle phase contrast which is proportional to the camera well depth. We expect this system to have the height sensitivity similar to an atomic force microcopy, while measuring biological structures with a full field of view in a single-shot. We plan to use this system to study cell dynamics, particularly lamellipodial height fluctuations as well as stiffer cell membrane fluctuations.

Abstract:A design method for an interferometric phase imaging system integrated with optical scattering information is proposed. Three types of cell models, i. c. , homogeneous cells, monocytes, and binuclear cell models, arc established. The morphological characteristics of a sample can be extracted quickly and quantitatively by analyzing the phase distribution information of the sample. Thus, the substructure characteristics of the sample can be determined accurately. The necessity and advantages of combining scattering information arc demonstrated through experiments on the red blood cells and neutrophils. The research results establish the feasibility of the proposed system. Additionally, the direction for improvement is proposed. The system has a simple structure and is easy to operate, and it can provide a reference for the rapid, non-destructive detection of morphological and structural characteristics of cells.

Abstract:As basic units of structure and function of the biological nervous system, neurons encode, transmit, and integrate information through discharge activities neuron, plays an important role in life activities. Based on the dynamic nature of neuronal discharge activity and phase imaging technique, we discuss nondestructive and label free imaging method for living cells and their inner substructure. According to phase information of neuron model, we study the characteristics of static morphology and dynamic discharge activities. The optical imaging simulation technique is used to establish the neuron phase model and obtain the information of its phase distribution. Starting from the physical meaning of the phase function, the substructure of the model is analyzed. Considering dynamic effect of the change of intracellular ion concentration in discharge activities on refractive index and phase information, the method of visualizing the dynamic process with phase information is discussed preliminarily. For the complex heterogeneous phase volume model, the local static morphological information on the substructure and phase characterization result of dynamic discharge activity on the sample are obtained without phase decoupling by introducing the heterogeneous contrastive compensation idea. The availability of the method is verified by simulation analysis. The results show that the optical phase characterization method for neuronal discharge characteristics and morphology is nondestructive, label-free, and quantifiable.

Abstract:The refractive index (RI) distribution can serve as a natural label for undyed cell imaging. However, the majority of images obtained through quantitative phase microscopy is integrated along the illumination angle and cannot reflect additional information about the refractive map on a certain plane. Herein, a light-field reconstruction method to image the RI map within a depth of 0.2 μm is proposed. It records quantitative phase-delay images using a four-step phase shifting method in different directions and then reconstructs a similar scattered light field for the refractive sample on the focus plane. It can image the RI of samples, transparent cell samples in particular, in a manner similar to the observation of scattering characteristics. The light-field reconstruction method is therefore a powerful tool for use in cytobiology studies.

Abstract:Quantitative phase-contrast microscopy (QPCM) provides an effective approach for label-free detection of transparent samples. In this study, we propose a condenser-free quantitative phase-contrast microscopy (CF-QPCM), in which several light-emitting diodes (LEDs) distributed on a ring are used for direct ultra-oblique illumination. Such condenser-free design greatly simplifies the system's structure and releases the space for installing samples. Quantitative phase maps are reconstructed by retarding the unscattered components of the object waves for a series of phases 0, π/2, π, and 3π/2 through a high-speed spatial light modulator (SLM). With this system, quantitative phase imaging of live cells has been achieved at a spatial resolution of 231 nm (lateral) and a frame rate of 250 Hz. We believe that the proposed CF-QPCM can contribute to biomedical, industrial, chemistry fields, etc.

Abstract:Current methods for studying organelle and protein interactions and correlations depend on multiplex fluorescent labeling, which is experimentally complex and harmful to cells. Here we propose to solve this challenge via OS-PCM, where organelles are imaged and segmented without labels, and combined with standard fluorescence microscopy of protein distributions. In this work, we develop new neural networks to obtain unlabeled organelle, nucleus and membrane predictions from a single 2D image. Automated analysis is also implemented to obtain quantitative information regarding the spatial distribution and co-localization of both protein and organelle, as well as their relationship to the landmark structures of nucleus and membrane. Using mitochondria and DRP1 protein as a proof-of-concept, we conducted a correlation study where only DRP1 is labeled, with results consistent with prior reports utilizing multiplex labeling. Thus our work demonstrates that OS-PCM simplifies the correlation study of organelles and proteins.

Abstract:Panoramic and long-term observation of nanosized organelle dynamics and interactions with high spatiotemporal resolution still hold great challenge for current imaging platforms. In this study, we propose a live-organelle imaging platform, where a flat-fielding quantitative phase contrast microscope (FF-QPCM) visualizes all the membrane-bound subcellular organelles, and an intermittent fluorescence channel assists in specific organelle identification. FF-QPCM features a high spatiotemporal resolution of 245 nm and 250 Hz and strong immunity against external disturbance. Thus, we could investigate several important dynamic processes of intracellular organelles from direct perspectives, including chromosome duplication in mitosis, mitochondrial fusion and fission, filaments, and vesicles’ morphologies in apoptosis. Of note, we have captured, for the first time, a new type of mitochondrial fission (entitled mitochondrial disintegration), the generation and fusion process of vesicle-like organelles, as well as the mitochondrial vacuolization during necrosis. All these results bring us new insights into spatiotemporal dynamics and interactions among organelles, and hence aid us in understanding the real behaviors and functional implications of the organelles in cellular activities.

Abstract:Organelles are highly dynamic and fulfill their function by constant motion and cooperation with each other. Current methods rely on fluorescence, leading to short observation time (via photobleaching) and experimental complexity (via multiple labeling). While label-free microscopes promise a paradigm change in this regard, the spatiotemporal resolutions and specificity are still insufficient to study organelle interactions. Using mitochondria and lysosome as examples, we demonstrate that our organelle-specific phase contrast microscopy (OS-PCM) can achieve automatic analysis of dynamic metrics of multiple organelles as well as their interactions from unlabeled cells for the first time. Compared to fluorescence-based methods, our method is gentle and holds great promise for label-free visualization and analysis of pan-organelle dynamics and interactions, with minimum perturbation to the cell.

Abstract:Fourier ptychographic microscopy (FPM), as an emerging computational imaging method, has been applied to quantitative phase imaging with resolution bypassing the physical limit of the detection objective. Due to the weak illumination intensity and long image acquisition time, the achieved imaging speed in current FPM methods is still low, making them unsuitable for real-time imaging applications. We propose and demonstrate a high-speed FPM method based on using laser illumination and digital micro-mirror devices for illumination angle scanning. In this new, to the best of our knowledge, FPM method, we realized quantitative phase imaging and intensity imaging at over 42 frames per second (fps) with around 1 µm lateral resolution. The quantitative phase images have revealed membrane height fluctuations of red blood cells with nanometer-scale sensitivity, while the intensity images have resolved subcellular features in stained cancer tissue slices.

Abstract:Quantitative phase microscopy (QPM) has been emerged as an indispensable diagnostic and characterization tool in biomedical imaging with its characteristic nature of label-free, noninvasive, and real time imaging modality. The integration of holography to the conventional microscopy opens new advancements in QPM featuring high-resolution and quantitative three-dimensional image reconstruction. However, the holography schemes suffer in space-bandwidth and time-bandwidth issues in the off-axis and phase-shifting configuration, respectively. Here, we introduce an on-axis phase-shifting holography based QPM system with single-shot imaging capability. The technique utilizes the Fizeau interferometry scheme in combination with polarization phase-shifting and space-division multiplexing to achieve the single-shot recording of the multiple phase-shifted holograms. Moreover, the high-speed imaging capability with instantaneous recording of spatially phase shifted holograms offers the flexible utilization of the approach in dynamic quantitative phase imaging with robust phase stability. We experimentally demonstrated the validity of the approach by quantitative phase imaging and depth-resolved imaging of paramecium cells. Furthermore, the technique is applied to the phase imaging and quantitative parameter estimation of red blood cells. This integration of a Fizeau-based phase-shifting scheme to the optical microscopy enables a simple and robust tool for the investigations of engineered and biological specimen with real-time quantitative analysis.
Capturing cell morphology dynamics with high temporal resolution using single-shot quantitative phase gradient imaging

Abstract:Significance: Label-free quantitative phase imaging can potentially measure cellular dynamics with minimal perturbation, motivating efforts to develop faster and more sensitive instrumentation. We characterize fast, single-shot quantitative phase gradient microscopy (ss-QPGM) that simultaneously acquires multiple polarization components required to reconstruct phase images. We integrate a computationally efficient least squares algorithm to provide real-time, video-rate imaging (up to 75 frames / s ). The developed instrument was used to observe changes in cellular morphology and correlate these to molecular measures commonly obtained by staining. Aim: We aim to characterize a fast approach to ss-QPGM and record morphological changes in single-cell phase images. We also correlate these with biochemical changes indicating cell death using concurrently acquired fluorescence images. Approach: Here, we examine nutrient deprivation and anticancer drug-induced cell death in two different breast cell lines, viz., M2 and MCF7. Our approach involves in-line measurements of ss-QPGM and fluorescence imaging of the cells biochemically labeled for viability. Results: We validate the accuracy of the phase measurement using a USAF1951 pattern phase target. The ss-QPGM system resolves 912.3 lp / mm , and our analysis scheme accurately retrieves the phase with a high correlation coefficient ( ∼ 0.99 ), as measured by calibrated sample thicknesses. Analyzing the contrast in phase, we estimate the spatial resolution achievable to be 0.55 μ m for this microscope. ss-QPGM time-lapse live-cell imaging reveals multiple intracellular and morphological changes during biochemically induced cell death. Inferences from co-registered images of quantitative phase and fluorescence suggest the possibility of necrosis, which agrees with previous findings. Conclusions: Label-free ss-QPGM with high-temporal resolution and high spatial fidelity is demonstrated. Its application for monitoring dynamic changes in live cells offers promising prospects.

Abstract:We propose a novel, to the best of our knowledge, single-shot quantitative phase imaging (QPI) technique with the phase modulation of a liquid crystal spatial light modulator (LC-SLM) under white light illumination. By studying the phase modulation characteristics of an LC-SLM under white light illumination, images captured at different wavelengths are equivalent to those captured at different defocus distances when loading a Fresnel lens pattern on the LC-SLM. Consequently, a color camera is able to simultaneously acquire multi-intensity images at different defocus distances. Finally, the phase is retrieved from a single-shot color image using the transport of intensity equation. To demonstrate the flexibility and accuracy of our method, a photoetched phase object and human red blood cells are quantitatively measured. An investigation of living yeast cells is conducted to verify the dynamic measurement capability. The proposed method provides a simple, efficient, and flexible means to accomplish real-time high-resolution quantitative phase imaging without sacrificing the field of view (FOV), which can be further integrated into a conventional microscope to achieve real-time microscopic QPI.

Abstract:We propose a novel common-path quantitative phase imaging (QPI) method based on a digital micromirror device (DMD). The DMD is placed in a plane conjugate to the objective back-aperture plane for the purpose of generating two plane waves that illuminate the sample. A pinhole is used in the detection arm to filter one of the beams after sample to create a reference beam. Additionally, a transmission-type liquid crystal device, placed at the objective back-aperture plane, eliminates the specular reflection noise arising from all the “off” state DMD micromirrors, which is common in all DMD-based illuminations. We have demonstrated high sensitivity QPI, which has a measured spatial and temporal noise of 4.92 nm and 2.16 nm, respectively. Experiments with calibrated polystyrene beads illustrate the desired phase measurement accuracy. In addition, we have measured the dynamic height maps of red blood cell membrane fluctuations, showing the efficacy of the proposed system for live cell imaging. Most importantly, the DMD grants the system convenience in varying the interference fringe period on the camera to easily satisfy the pixel sampling conditions. This feature also alleviates the pinhole alignment complexity. We envision that the proposed DMD-based common-path QPI system will allow for system miniaturization and automation for a broader adaption.

Abstract:Understanding how cells respond to external stimuli is crucial. However, there are a lack of inspection systems capable of simultaneously stimulating and imaging cells, especially in their natural states. This study presents a novel microfluidic stimulation and observation system equipped with flat-fielding quantitative phase contrast microscopy (FF-QPCM). This system allowed us to track the behavior of organelles in live cells experiencing controlled microfluidic stimulation. Using this innovative imaging platform, we successfully quantified the cellular response to shear stress including directional cellular shrinkage and mitochondrial distribution change in a label-free manner. Additionally, we detected and characterized the cellular response, particularly mitochondrial behavior, under varying fluidic conditions such as temperature and drug induction time. The proposed imaging platform is highly suitable for various microfluidic applications at the organelle level. We advocate that this platform will significantly facilitate life science research in microfluidic environments.

Quantitative dynamic cellular imaging based on 3D unwrapped optically computed phase microscopy

Abstract:We investigate continuous observation of dynamic phenomena through quantitative phase microscopy. We conduct imaging studies using optically computed phase microscopy, a novel imaging technology developed in our lab, to the best of our knowledge. Inevitably, continuous phase imaging is affected by phase wrapping artifacts, which affects correct quantification of sample dynamics. To address this issue, we develop a 3D unwrapping method that exploits data correlation in space as well as in time. We validate our 3D phase unwrapping method using simulated data. We further validate 3D phase unwrapping using experimental data and demonstrate quantitative phase imaging that accurately characterizes sample dynamics. We image the nanoscale motion of the sample actuated by a piezo transducer (PZT). We calculate the displacement using 3D unwrapped phase, and the result is consistent with the known motion of the PZT. We also image live cells that were detaching from the substrate of the petri dish. The optical path length calculated using 3D unwrapped phase increases as the dry mass of the cell becomes more concentrated during the detachment process.

Abstract:Recent developments in phase contrast microscopy have enabled the label‐free visualization of certain organelles due to their distinct morphological features, making this method an attractive alternative in the study of cellular dynamics. However tubular structures such as ER networks and complex dynamics such as the fusion and fission of mitochondria, due to their low phase contrast, still need fluorescent labeling to be adequately imaged. In this paper, we report a quantitative phase microscope with ultra‐oblique illumination that enables us to see those structures and their dynamics with high contrast for the first time without labeling. The imaging capability was validated through comparison to the fluorescence images with the same field‐of‐view. The high image resolution ( ~270nm) was validated using both beads and cellular structures. Further, we were able to record the vibration of ER networks at a frame rate of 250Hz. We additionally show complex cellular processes such as remodelling of the mitochondria networks through fusion and fission and vesicle transportation along the ER without labels. Our high spatial and temporal resolution allowed us to observe mitochondria "spinning", which has not been reported before, further demonstrating the advantages of the proposed method. Abstract:We design a novel phase real-time microscope camera (PhaseRMiC) for live cell phase imaging. PhaseRMiC has a simple and cost-effective configuration only consisting of a beam splitter and a board-level camera with two CMOS imaging chips. Moreover, integrated with 3-D printed structures, PhaseRMiC has a compact size of 136×91×60 mm3, comparable to many commercial microscope cameras, and can be directly connected to the microscope side port. Additionally, PhaseRMiC can be well adopted in real-time phase imaging proved with satisfied accuracy, good stability and large field of view. Considering its compact and cost-effective device design as well as real-time phase imaging capability, PhaseRMiC is a preferred solution for live cell imaging.

Abstract:Quantitative phase imaging has been an important labeling-free microscopy modality for many biomedical and material science applications. In which, ultra-fast quantitative phase imaging is indispensable for dynamic or transient characteristics analysis. Conventional wide field optical interferometry is a common scheme for quantitative phase imaging, while its data acquisition rate is usually hindered by the frame rate of arrayed detector. By utilizing novel balanced-photo-detector based digital optics coherent detection techniques, we report on a method of constructing ultra-fast quantitative phase microscopy at the line-scan rate of 100 MHz with ~2 μm spatial resolution.
