Orexinergic Neurons Hyperfunction Causes Sleep Disorders and Hypertension in SHR

Abstract:Background: Epidemiologic studies have shown that sleep disorder may be associated with the development of hypertension. The present study was aimed to clarify the correlations between hypertension and sleep disorder across the age span and to investigate related neural mechanism focusing on orexinergic systems in spontaneously hypertensive rats (SHRs). Methods: Blood pressure was measured by noninvasive blood pressure tail cuff test and sleep was monitored by electroencephalography (EEG) and electromyography (EMG) recordings at the age of 4, 6, 8, 12 and 24 weeks. Anti-OX2r-SAP (200 or 400 ng/each side) was microinjected into the perifornical necleus (Pef) to inhibit orexinergic neurons function. Results: SHR blood pressure was higher than age-matched WKY rats from 6-weeks old, and the differences significantly increased with age. SHR showed sleep disorders, as evidenced by decrease in total sleep time and rapid-eye movement sleep (REMS) time and delta power density during non-REMS (indication of poor sleep quality), and deficiency in homeostatic sleep pressure regulation. These sleep disorders in SHR were observed since 4-weeks old and greatly deteriorated with age, as compared with age-matched WKY rats. Correlation analysis showed that blood pressure was negatively correlated with sleep time and quality across the age span in SHR. Lesion of orexinergic neurons in the Pef partially ameliorated sleep disorders and hypertension in SHR. Conclusions: There results indicate that there is a positive correlation between sleep disorders and hypertension in SHR and orexinergic neuron in the Pef might be one of the common pathogenesis site for the hypertension and sleep disorder.
