The Role of Notch Signaling on Nucleus Pulposus Cell Proliferation and Apoptosis

Abstract:Background: Notch signaling regulates cell proliferation, differentiation and apoptosis involved in development of organs and tissues e.g. nerves, cartilage, lungs etc. This study aimed to investigate the role of Notch signaling in nucleus pulposus (NP) cell proliferation, cell cycle arrest and apoptosis, along with the related signaling pathways. Methods: To examine the involvement of Notch signaling on NP cells proliferation and cell cycle arrest, recombinant JAG2 was used to activate Notch signaling, related proteins and mRNA were detected using Western blotting and RT-PCR. Then Notch2, Hes1 and Hey2 siRNA were used to inhibit Notch signaling, NP cells proliferation and cell cycle arrest were determined by CCK8 and Flow cytometric analysis. Cell cycle regulatory factors, Cyclin D1, CDK6 and p21 protein was also detected. Pathways associated with Notch-mediated proliferative effect on NP cells were examined by their specific inhibitors. Finally the role of Notch signaling on NP cells apoptosis was examined by gain and loss of function study using Flow cytometric analysis, and Western blotting and RT-PCR were used to detect the related apoptosis pathway. Results: First, the expression of Notch2 and Hes1/Hey2 was induced by recombinant JAG2 protein. Then, we found recombinant JAG2 protein induces proliferation of NP cells, and depletion of Notch2/Hes1/Hey2 reduces NP cells proliferation. Consistent with cell proliferation, cell cycle regulatory factor Cyclin D1 is critically required for the Notch-mediated effect on NP cells proliferation, besides Notch-mediated proliferative effect required functional PI3K/Akt and Wnt/β‑catenin signaling. Furthermore, flow cytometry showed down-regulation of Notch2/Hes1/Hey2 induces G0/G1 phase cell cycle arrest in NP cells. The role of Notch signal on NP cell apoptosis was also detected, results showed recombinant JAG2 can reverse NP cells apoptosis induced by TNF-α, while Notch2/Hes1/Hey2 siRNA can significantly enhance the apoptotic rate induced by TNF-α. Furthermore, Notch signal inhibit TNF-α-promoted apoptosis by suppressing the formation of RIP1-FADD-caspase-8 complex. Conclusion: The JAG2/Notch2 axial play an important role in NP cell proliferation, and it is via PI3K/Akt and Wnt/β‑catenin signaling. Besides, Notch signal can inhibit TNF-α-induced apoptosis by suppressing the formation of RIP1-FADD-caspase-8 complex. Taken together, current and previous research throw the light on therapeutic implications of targeting Notch signal to inhibit or reverse intervertebral disc degeneration.
