Abstract:Peroxynitrite (ONOO-) has been proven to participate in various physiological and pathological processes, and may also be a contributing factor in many diseases. In view of this, there is a need to develop detection tools for unambiguously tracking a small amount of endogenous ONOO- to reveal its exact mechanisms. In this paper, a colorimetric and red-emitting fluorescent probe Red-PN, based on a rhodamine-type fluorophore and hydrazide reactive site is described. The probe Red-PN possesses the advantages of rapid response (within 5 s), visual color change (from colorless to pink), preeminent sensitivity (detection limit = 4.3 nM) and selectivity. Because of these outstanding performances, it was possible to accurately detect endogenous ONOO-. It was encouraging that the probe Red-PN could be used effectively for tracking the relatively low levels of endogenous and exogenous ONOO-in living cells and zebrafish. Thus, it is envisioned that the probe Red-PN would have promising prospects in applications for imaging ONOO- in a variety of biological settings.

Abstract:In this study, we present a novel near-infrared (NIR) fluorescent probe Nile-ONO designed for the selective and sensitive detection of ONOO − . The probe Nile-ONO employed Nile red as the fluorophore, with diphenylphosphinate serving as the reaction site. In the presence of ONOO − , the probe Nile-ONO exhibits remarkable fluorescence enhancement at 659 nm, with a response time of less than 20 min and a low detection limit of 0.32 μM. Importantly, MTT assays demonstrate low cytotoxicity in living cells. Furthermore, Nile-ONO has excellent imaging capabilities for endogenous ONOO − . Overall, this work introduces a valuable new method for the rapid detection of ONOO − in biological systems.

Abstract:Peroxynitrite (ONOO−) has been revealed to play crucial roles in many physiological and pathological processes, and many diseases were proven to be associated with its misregulated production. The development of fluorescent probes meets the need for tracking ONOO− and gives a better understanding of its diverse mechanisms. In this work, a red-emitting fluorescent probe BP-ONOO was synthesized via functionalization of the rhodol-like fluorophore with a reactive site of hydrazide. The probe BP-ONOO exhibited high sensitivity, excellent selectivity, and short response time (less than 4 s) towards ONOO− under neutral or weak alkaline conditions. These attractive properties favor its application in real-time imaging of ONOO− in living cells, and the probe has been successfully applied for imaging the concentration levels of ONOO− in RAW 264.7 macrophage cells under drug stimulation.
Highly selective and sensitive benzopyran-based fluorescent probes for imaging exogenous and endogenous peroxynitrite

Abstract:Peroxynitrite is widely present in organisms and closely related to many pathophysiological functions. Therefore, it is of great physiological significance to develop capable probes for detecting ONOO − . In this work, a novel fluorescent probe B-Ch was designed based on the intramolecular charge transfer (ICT) effect. By means of molecular engineering, the replacement from diethylamine group to hydroxyl group has improved the detection sensitivity of the probe. After the addition of ONOO − , the solution color and fluorescence showed noticeable changes, which were visible to the naked eye. The probe showed excellent advantages: visualization, good selectivity, low sensitivity (22.4 nM), good stability and biocompatibility, exogenous and endogenous imaging of ONOO − in HeLa cells.

Abstract:In- this work, a novel boronate-based fluorescent probe (FAM) for the endogenous detection of peroxynitrite (ONOO-) has been developed by using anthracycline as the fluorophore, arlyboronate as the recognition moiety, lipophilic cation as the mitochondria] targeting moiety. Upon reaction of the probe with ONOO-, the oxidation and subsequent hydrolysis of ONOO- to arlyboronate triggers quite rapid fluorescence off-on response, providing a sensitive and highly selective method for the detection of ONOO-. In addition, probe holds high sensitivity with the detection limit of 32 nM and excellent specificity including a series of biologically relevant reactive oxygen species. Abstract:Peroxynitrite (ONOO−), as one of the important reactive oxygen/nitrogen species, is associated with many physiological and pathological activities. Therefore, effective detection methods with high sensitivity and rapid response time are essential to reveal its functions. A novel Rhodol-based fluorescent probe RN is constructed and synthesized by introducing of 1,1-dimethylhydrazone as a recognition group. Due to the oxidative breakage of the CN bond by ONOO−, the probe has significant fluorescence enhancement change with excellent detection properties such as rapid response time, high sensitivity and selectivity. Moreover, probe RN show low cytotoxicity and is successfully capable of imaging endogenous and endogenous ONOO− in living cells. These properties indicate that RN has great application potential for imaging of ONOO− in biological systems.

Abstract:Peroxynitrite is a short-lived endogenous reactive species and plays important roles in many physiological and pathological processes. In this work, we synthesized a near-infrared probe based on the structure of benzothiazole derived cyanine for determination of peroxynitrite (ONOO-). The designed probe specifically reacted with ONOO- through oxidative cleavage of conjugated C˭C double bonds and generating the non-fluorescent product. Meanwhile, the characteristic absorption of the probe at 630 nm greatly decreased after reaction with ONOO-, accompanied by drastic color change from bright blue to green yellow, which exhibited a distinct visual feature. It was demonstrated that the probe could be used to measure ONOO- in a dose-response manner and had a detection limit lower as 26 nM. Furthermore, the probe Cy-SN was applied for the imaging of endogenous ONOO- in living cells by confocal microscopy, which showed good cell permeability and low cytotoxicity. Successful application of probe for exogenous colorimetric detection and endogenous fluorescence imaging of ONOO- is suggesting its great potential applications in biological analysis.

Abstract:The peroxynitrite ion (ONOO-) has important roles in many biological processes. We have developed a multicolor ONOO--sensing probe (SiONNOH) that undergoes deprotonation and desilylation processes, which result in several changes in the emission wavelengths. In response to different concentrations of ONOO-, the probe exhibits fluorescence changes from pink (595 nm at 2 eq. ONOO-) to green (540 nm at 6 eq. ONOO-) via orange (3 eq. ONOO-) and yellow (4 eq. ONOO-) under physiological conditions until no fluorescence signal is observed after ONOO- is completely eliminated by lipoic acid. The probe shows the high selectivity for ONOO- and the limit of detection is calculated to be 1.27 mu M. Moreover, the probe shows the capacity to monitor the concentration ranges of ONOO- through multicolor fluorescence in living cells, which will greatly facilitate the rapid detection of ONOO- concentration ranges by the naked eye under a UV light without any precision instrumentation.

Abstract:The development of sensitive methods for imaging endogenous peroxynitrite (ONOO-) in the mitochondria of living cells is very important to disclose its diverse physiological and pathological functions. In this paper, we have described a simple, mitochondria-targeted far-red fluorescent probe Mito-PN for selectively detecting ONOO- even though in the presence of higher concentration (1 mM) other biologically important substances. The experimental results indicated that probe Mito-PN could quantitatively determine ONOO- in the range of 0.5-8 mu M with the detection limit of 17 nM. In addition, probe Mito-PN exhibits a fast response for ONOO- (<5 s), which is favorable for monitoring the highly reactive and short-lived ONOO- in the biological systems. The excellent sensing properties of probe Mito-PN enable its applications in the tracking of the fluctuations of endogenous ONOO- levels in the mitochondria of living cells. (c) 2017 Elsevier B.V. Abstract:Peroxynitrite (ONOO-) plays a crucial role in the regulation of diverse pathophysiological processes, and high level of ONOO- is profound association with numerous diseases. Herein, we developed an anthraquinone-based fluorescent probe L for ONOO- determination by a new recognition mechanism: amido oxidized nitroso-group by ONOO-. Probe L with amine-based recognition receptor is more selective to ONOO- than other reactive oxygen species, including H2O2 and ClO-. Abstract:Peroxynitrite (ONOO(-)) is a highly reactive species implicated in the pathology of numerous diseases and there is currently great interest in developing fluorescent probes that can selectively detect ONOO(-) in living cells. Herein, a polymeric micelle-based and cell-penetrating peptide-coated fluorescent nanoprobe that incorporates ONOO(-) indicator dye and reference dye for the ratiometric detection and imaging of ONOO(-) has been developed. The nanoprobe effectively avoids the influences from enzymatic reaction and high-concentration ·OH and ClO(-). The improved ONOO(-) selectivity of the nanoprobe is achieved by a delicate complementarity of properties between the nanomatrix and the embedded molecular probe (BzSe-Cy). This nanoprobe also has other attractive properties, such as good water solubility, photostability, biocompatibility, and near-infrared excitation and emission. Fluorescence imaging experiments by confocal microscopy show that this nanoprobe is capable of visualizing ONOO(-) produced in living cells and it exhibits very low toxicity and good membrane permeability. We anticipate that this technique will be a potential tool for the precise pathological understanding and diagnosis of ONOO(-)-related human diseases.

Abstract:The fluorescent probe, GXY-ADP-2, with xanthene structure as the fluorescent core was designed and prepared for the selective detection of peroxynitrite (ONOO−). ONOO− can be produced endogenously and exogenously and is a strong oxidant with a short half-life. Oxidative modifications of biomolecules, that can be attributed to the formation of ONOO−, occur in the reactions of biomolecules with secondary ONOO−-derived radical oxidants. Therefore, it is very important to develop a specific fluorescent probe for detecting ONOO− to monitor oxidative stress state. The excitation wavelength and emission wavelength of the probe are 689 nm and 739 nm respectively. In the process of co-incubation with ONOO−, generate a new substance with two internal conjugated structures through a special reaction mechanism, one giving the fluorescence with the excitation wavelength of 347 nm and the emission wavelength of 484 nm with the detection limit of 0.12 μM, and the other that with the excitation wavelength of 433 nm and the emission wavelength of 583 nm with the detection limit of 0.077 μM. The linear dynamic range of the probe is 0–5 μM. Its response is not affected by the other reactive oxygen species, thus can sensitively detect ONOO−. In bioimaging experiments with HepG2 cells, the green and blue cell fluorescence signals (583 nm and 433 nm, respectively) were increased, while the red one (739 nm) was significantly reduced, under lipopolysaccharide (LPS) induced oxidative stress, proving that the probe could sensitively detect ONOO− in living cells. This work provides a new tool for the dynamic changes of ONOO− and oxidative stress processes in biological systems.

Abstract:Peroxynitrite (ONOO-), an endogenous reactive oxygen species, acts an important role in various physiological and pathological processes. Herein, we designed a mitochondria targeting fluorescent probe MBTBE for sensitive determination of ONOO-. MBTBE exhibits rapid response ( < 1 min) and high selectivity towards peroxynitrite with distinct color changing (colorless to orange). The fluorescence intensity at 569 rim was linearly correlated with the concentration of ONOO- (0-10 mu M) with a detection limit of 16 nM. Additionally, MBTBE possesses good biocompatibility and mitochondria-targeting. Furthermore, MBTBE was successfully applied to monitor fluctuations in ONOO-levels and H2S-induced ONOO- clearing in HeLa cells, indicating excellent potential applications in bioanalysis analysis.

Abstract:As a member of the reactive nitrogen species (RNS) family, peroxynitrite (ONOO−) as an oxidant and nitrating mediator plays a significant role in some physiopathologic processes. The excessive production of peroxynitrite anion in a drug-damaged liver is a culprit of hepatotoxicity. The detection of peroxynitrite is of vital importance for the treatment of some diseases including cancer and liver injury. In this study, a novel turn-on fluorescent probe IC-ONOO with new chalcone fluorophore was designed and synthesized for the detection of in vitro and in vivo. The probe responded rapidly towards ONOO− (only within 15 min did the fluorescent intensity maximize), and was endowed with high sensitivity and excellent selectivity. Given the fact that the linear correlation between the fluorescent intensity at 560 nm and the concentrations of the probe ranged from 0 to 9 μM, the limit of detection (LOD) was calculated to be 3.1 × 10−8 M. With all the merits, probe IC-ONOO was qualified as a robust tool to monitor peroxynitrite anion under physiopathologic condition. Moreover, it was successfully applied in the imaging of endogenous peroxynitrite in living MCF-7 cells (Human breast carcinoma cells) and mouse drug-damaged liver tissue with low cytotoxicity. Given all the extraordinary merits, great potential has been seen in its application to other peroxynitrite related diseases.

Abstract:Peroxynitrite (ONOO-), a highly reactive oxygen species (ROS), is implicated with many physiological and pathological processes including cancer, neurodegenerative diseases and inflammation. In this regard, developing effective tools for highly selective tracking of ONOO- is urgently needed. Herein, we constructed a concise and specific fluorescent probe NA-ONOO for sensing ONOO by conjugating an ONOO--specific recognition group ((4-methoxyphenylthio)carbonyl, a thiocarbonate derivative) with a naphthalene fluorophore. The probe, NA-ONOO, was in a dark state because the high electrophilicity of (4-methoxyphenylthio)carbonyl disturbs the intramolecular charge transfer (ICT) in the fluorophore. Upon treatment with ONOO-, the fluorescent emission was sharply boosted (quantum yield Phi: 3% to 56.6%) owing to an ONOO- triggered release of (4-methoxyphenylthio)carbonyl from NA-ONOO. Optical analyses showed that NA-ONOO presented high selectivity and sensitivity toward ONOO-. With good cell permeability and biocompatibility, the NA-ONOO probe was successfully applied to imaging and tracing exogenous and endogenous ONOO- in living cells and zebrafish. The probe NA-ONOO presents a new recognition group and a promising method for further investigating ONOO- in living systems.

Abstract:Peroxynitrite (ONOO-), a highly reactive oxygen species generated by the reaction of nitric oxide and superoxide radical anion, is involved in numerous physiological and pathological processes in the human body. To identify important pathogenic mechanisms, it is crucial to develop a reliable tool for detecting peroxynitrite in living systems. In the present study, a new difluoroboron β-diketonate-based fluorescent probe for detecting exogenous and endogenous peroxynitrite in living systems was designed. The red emitting fluorophore can be synthesized in a simple three-step procedure. This probe reacts quickly and selectively with peroxynitrite and its detection limit is determined to be as low as 19.8 nM. It allows for clear imaging of peroxynitrite in RAW 264.7 cells and was successfully applied to visualize changes of intracellular peroxynitrite induced by reactive oxygen species inhibitors. This designed probe is an effective tool for investigating the physiological and pathological role of peroxynitrite in living cells.
Dual NIR-channel Fluorescent Probe for Detecting ONOO− in Vitro and Vivo

Abstract:As one of endogenous reactive oxygen species (ROS), peroxynitrite (ONOO-) performs various functions in both pathological and physiological mechanisms. In this work, an optical and near-infrared (NIR) fluorescent probe (NX), which based on 3-dihydro-1H-xanthene and 2-dicyanomethylene-3-cyano-4,5,5-trimethyl-2,5-dihydrofuran (TCF) group was designed and prepared to detect ONOO-. This probe revealed an obvious optical and a fluorescent response when ONOO- was present and it exhibited higher selectivity over other ROS. Especially, the dual NIR fluorescence changes at 660 and 800 nm allowed quantitative detection of ONOO- in the range of 15-40 mu M, and the detection limit was 82 nM. Finally, the probe was effectively employed to visualize exogenous and endogenous ONOO- in HepG2 cells and zebrafish, respectively. All the results indicated the dual NIRchannel probe could serve as a potent detecting tools in studying ONOO- in vitro and in vivo.

Abstract:Peroxynitrite anion (ONOO-), one of the reactive nitrogen species (RNS), plays momentous roles in physiological and pathological processes especially in a range of oxidative stress-related diseases. Moreover, abundant ONOO- is generated in the liver tissues of drug-induced liver injury. We report herein a novel small molecule fluorescent probe KC-ONOO for monitoring ONOO- based on boronate. The probe displayed high sensitivity and good selectivity towards ONOO-. A good linear relationship was observed between the fluorescent intensity at 530 nm and the concentration of ONOO- ranged 0-10 mu M with a detection limit of 1.5 x 10(-8) M. Furthermore, our probe was successfully applied for imaging ONOO- in living cells and drug-damaged liver tissues with low cytotoxicity, demonstrating the probe KC-ONOO has great potential to further elucidate more biological roles of ONOO-.
