Abstract:Titanium dioxide nanoparticles (TiO2 NPs) are among the most widely produced engineered nanoparticles (NPs) and are used in a number of fields. However, the impact of the released TiO2 NPs on nutritional quality of edible vegetable crops and the associated mechanisms responsible for those changes are poorly understood. In the present study, lettuce (Lactuca sativa L.) was exposed to 50% Hoagland's nutrient solution amended with 0, 50, 100, 200, or 400 mg L-1 TiO2 NPs for one week. The results showed that TiO2 NPs exposure at 50-400 mg L-1 negatively impacted the nutritional quality of lettuce. Specifically, 50-400 mg L-1 TiO2 NPs significantly decreased the content of Ca by 33.6-35.7%, Mg by 7.1-27.2%, Mn by 21.0-34.5%, Fe by 8.6-21.0%, Zn by 11.8-22.6%, and B by 27.3-44.1% in lettuce edible tissues, relative to the control. However, 50-200 mg L-1 TiO2 NPs exposure significantly increased the root and shoot dry biomass by 0.69-1.3-fold and 32.0-75.6%, respectively; the contents of chlorophyll a, total chlorophyll, and carotenoid were increased by 14.7-24.5%, 10.3-18.5%, and 12.6-20.5%, respectively, relative to the control. The reductions in nutrient content under 50-200 mg L-1 TiO2 NPs exposure were correlated with the significant increase in plant biomass, suggesting that the changes were a function of the biomass dilution effect. Alternatively, the nutrient content reduction at 400 mg L-1 TiO2 NPs did not result from this mechanism since plant growth (dry biomass) was unchanged. Here, cell structure damage including root cell membrane shrinkage and a disordering of the thylakoid arrangement resulting from strong oxidative stress occurred, which negatively affected nutrient uptake and led to nutrient content decreases. In addition, TiO2 NPs were adsorbed on root surfaces, which could also inhibit both water and nutrient uptake. Importantly, mu-XRF mapping showed that TiO2 NPs exposure did not change the nutrient distribution pattern in exposed leaf tissues. The findings from this work highlight that TiO2 NPs exposure negatively impacts the nutritional quality of lettuce, although the mechanisms of plant response vary with exposure level of TiO2 NPs.
Lettuce (Lactuca sativa L.) alters its metabolite accumulation to cope with CuO nanoparticles by promoting antioxidant production and carbon metabolism

Abstract:Copper-based nanoparticles (NPs) are gradually being introduced as sustainable agricultural nanopesticides. However, the effects of NPs on plants requires carefully evaluation to ensure their safe utilization. In this study, leaves of 2-week-old lettuce (Lactuca sativa L.) were exposed to copper oxide nanoparticles (CuO-NPs, 0 [CK], 100 [T1], and 1000 [T2] mg/L) for 15 days. A significant Cu accumulation (up to 1966 mg/kg) was detected in lettuce leaves. The metabolomics revealed a total of 474 metabolites in lettuce leaves, and clear differences were observed in the metabolite profiles of control and CuO-NPs treated leaves. Generally, phenolic acids and alkaloids, which are important antioxidants, were significantly increased (1.26-4.53 folds) under foliar exposure to NPs; meanwhile, all the significantly affected flavonoids were down-regulated after CuO-NP exposure, indicating these flavonoids were consumed under oxidative stress. Succinic and citric acids, which are key components of the tricarboxylic acid cycle, were especially increased under T2, suggesting the energy and carbohydrate metabolisms were enhanced under high-concentration CuO-NP treatment. There was also both up- and down-regulation of fatty acids, suggesting cell membrane fluidity and function responded to CuO-NPs. Galactinol, which is related to galactose metabolism, and xanthosine, which is crucial in purine and caffeine metabolism, were down-regulated under T2, indicating decreased stress resistance and disturbed nucleotide metabolism under the high CuO-NP dose. Moreover, the differentially accumulated metabolites were significantly associated with plant growth and its antioxidant ability. Future work should focus on controlling the overuse or excessive release of NPs into agricultural ecosystems to limit their adverse effects.
Dose-Dependent Physiological and Transcriptomic Responses of Lettuce (Lactuca sativa L.) to Copper Oxide Nanoparticles—Insights into the Phytotoxicity Mechanisms

Abstract:Understanding the complex mechanisms involved in plant response to nanoparticles (NPs) is indispensable in assessing the environmental impact of nano-pollutants. Plant leaves can directly intercept or absorb NPs deposited on their surface; however, the toxicity mechanisms of NPs to plant leaves are unclear. In this study, lettuce leaves were exposed to copper oxide nanoparticles (CuO-NPs, 0, 100, and 1000 mg/L) for 15 days, then physiological tests and transcriptomic analyses were conducted to evaluate the negative impacts of CuO-NPs. Both physiological and transcriptomic results demonstrated that CuO-NPs adversely affected plant growth, photosynthesis, and enhanced reactive oxygen species (ROS) accumulation and antioxidant system activity. The comparative transcriptome analysis showed that 2270 and 4264 genes were differentially expressed upon exposure to 100 and 1000 mg/L CuO-NPs. Gene expression analysis suggested the ATP-binding cassette (ABC) transporter family, heavy metal-associated isoprenylated plant proteins (HIPPs), endocytosis, and other metal ion binding proteins or channels play significant roles in CuO-NP accumulation by plant leaves. Furthermore, the variation in antioxidant enzyme transcript levels (POD1, MDAR4, APX2, FSDs), flavonoid content, cell wall structure and components, and hormone (auxin) could be essential in regulating CuO-NPs-induced stress. These findings could help understand the toxicity mechanisms of metal NPs on crops, especially NPs resulting from foliar exposure.

Abstract:Engineered nanoparticles (ENPs) and nanoplastics (NPs) are typical nanoparticles in terrestrial environments. Till now, few studies have compared their toxicity and mechanism to plants. Here we investigated the effects of CuO, nZVI ENPs and polystyrene (PS) NPs on lettuce growth, metabolic functions, and microbial community structure. Results showed that low concentrations of nanoparticles decreased root biomass and promoted photosynthetic indicators, whereas increased reactive oxygen species (ROS) were detected in roots exposed to high concentrations of nanoparticles. High-dose CuO ENP exposure significantly raised the MDA content by 124.6 % compared to CK, causing the most severe membrane damage in the roots among the three types of nanoparticles. Although linoleic acid metabolism was down-regulated, the roots alleviated CuO stress by up-regulating galactose metabolism. Uptake of PS by roots similarly caused ROS production and activated the oxidative stress system by altering amino acid and vitamin metabolism. Faster microbial responses to nanoparticles were observed in the nZVI and PS networks. The root toxicity was indirectly mediated by ion release, NP uptake, or ROS generation, ultimately impacting root cell metabolism, rhizospheric microorganism and plant growth. These findings provide theoretical basis for assessing environmental impact of nanoparticles and their possible ecological risks.

Abstract:The increased production and use of nanoparticles (NPs) has generated concerns about their impact on living organisms. In this study, nCu, bulk Cu, nCuO, bulk CuO, Cu(OH)(2) (CuPRO 2005, Kocide 3000), and CuCl2 were exposed for 15 days to 10 days-old hydroponically grown lettuce (Lactuca sativa) and alfalfa (Medicago sativa). Each compound was applied at 0, 5, 10, and 20 mg L-1. At harvest, we measured the size of the plants and determined the concentration of Cu, macro and microelements by using ICP-OES. Catalase and ascorbate peroxidase activity was also determined. Results showed that all Cu NPs/compounds reduced the root length by 49% in both plant species. All Cu NPs/compounds increased Cu, P, and S (>100%, >50%, and >20%, respectively) in alfalfa shoots and decreased P and Fe in lettuce shoot (>50% and >50%, respectively, excluding Fe in CuCl2 treatment). Biochemical assays showed reduced catalase activity in alfalfa (root and shoot) and increased ascorbate peroxidase activity in roots of both plant species. Results suggest that Cu NPs/compounds not only reduced the size of the plants but altered nutrient content and enzyme activity in both plant species.
Application of Metabolomics to Assess the Impact of Cu(OH)2 Nanopesticide on the Nutritional Value of Lettuce (lactuca Sativa): Enhanced Cu Intake and Reduced Antioxidants

Abstract:Copper-based nanopesticides are increasingly being used in the agriculture, especially in organic farming. This has triggered some concerns about their risk to environmental and human health. In this study, 24-day-old lettuce plants grown in soil were exposed via the leaves to different concentrations (0, 1050 and 1555mg/L) of Cu(OH)2 nanopesticides for one month. Results showed Cu was mainly localized in lettuce leaves (823–1111 and 1353–2008mg/kg in vascular and photosynthetic tissues), which may potentially increase Cu intake and impact human health. In addition, foliar application of Cu(OH)2 nanopesticide significantly increased potassium concentration in lettuce leaves by 6–7% and 21–28%, in vascular and photosynthetic tissues. A Gas Chromatography-Time-of-Flight Mass Spectrometry (GC–TOF-MS) based metabolomics approach was applied to determine hundreds of organic compounds simultaneously. Using relative quantitation and Partial Least Squares-Discriminant Analysis (PLS-DA) clustering of all compounds, clear differences were observed in the metabolite profiles of control and Cu(OH)2 nanopesticides treated leaves. Discriminating compounds include amino acids, organic acids, polyamines, vitamin C and polyphenols. Dehydroascorbic acid and cis-caffeic acid, which are important antioxidants, were significantly decreased (19–33% and 5–8%) due to foliar exposure to the nanopesticide. Total antioxidant capacity was significantly decreased 20–23% after exposure to Cu(OH)2 nanopesticides. There was also up- and down-regulation of a number of amino acids, particularly 4-hydroxybutytic acid (GABA) which decreased 50% compared to the control, potentially affecting the overall nutritional value of lettuce leaves exposed to the Cu(OH)2 nanopesticides. In future work, determining an appropriate level of nanopesticide will be important to obtain the antifungal benefits without resulting in a significant decrease in nutritional value.
Molecular characterization and transcriptional response of Lactuca sativa seedlings to co-exposure to graphene nanoplatelets and titanium dioxide nanoparticles

Abstract:The widespread use of nanomaterials in agriculture may introduce multiple engineered nanoparticles (ENPs) into the environment, posing a combined risk to crops. However, the precise molecular mechanisms explaining how plant tissues respond to mixtures of individual ENPs remain unclear, despite indications that their combined toxicity differs from the summed toxicity of the individual ENPs. Here, we used a variety of methods including physicochemical, biochemical, and transcriptional analyses to examine the combined effects of graphene nanoplatelets (GNPs) and titanium dioxide nanoparticles (TiO2 NPs) on hydroponically exposed lettuce (Lactuca sativa) seedlings. Results indicated that the presence of GNPs facilitated the accumulation of Ti as TiO2 NPs in the seedling roots. Combined exposure to GNPs and TiO2 NPs caused less severe oxidative damage in the roots compared to individual exposures. Yet, GNPs and TiO2 NPs alone and in combination did not cause oxidative damage in the shoots. RNA sequencing data showed that the mixture of GNPs and TiO2 NPs led to a higher number of differentially expressed genes (DEGs) in the seedlings compared to exposure to the individual ENPs. Moreover, the majority of the DEGs encoding superoxide dismutase displayed heightened expression levels in the seedlings exposed to the combination of GNPs and TiO2 NPs. The level of gene ontology (GO) enrichment in the seedlings exposed to the mixture of GNPs and TiO2 NPs was found to be greater than the level of GO enrichment observed after exposure to isolated GNPs or TiO2 NPs. Furthermore, the signaling pathways, specifically the "MAPK signaling pathway-plant" and "phenylpropanoid biosynthesis," exhibited a close association with oxidative stress. This study has provided valuable insights into the molecular mechanisms underlying plant resistance against multiple ENPs.

Abstract:Manufactured metal oxide nanoparticles (NPs) are being used on a large scale, and these particles will inevitably reach a body of water through wastewater and urban runoff. The ecotoxicological study of these NPs on hydrophyte is limited at present. Lemna minor was exposed to media with different concentrations of titanium dioxide (TiO2) NPs or bulk TiO2 for 7 d. The changes in plant growth, chlorophyll, antioxidant defense enzymes (peroxidase [POD], catalase [CAT], and superoxide dismutase [SOD] activities), and malondialdehyde (MDA) content were measured in the present study. The particle size of TiO2 NPs and the zeta potential of TiO2 NPs and of bulk TiO2 in the culture media were also analyzed to complementally study the toxicity of these materials on duckweed. The results showed that the effect of TiO2 NPs on plant growth was more obvious than bulk TiO2. Titanium dioxide NPs stimulated plant growth in low concentrations, but inhibited plant growth at high concentrations. The POD, SOD, and CAT activity of Lemna minor increased when TiO2 NP concentration was lower than 200?mg/L to eliminate accumulated reactive oxygen species in plant cells. The SOD activity decreased when the TiO2 NP concentration was higher than 200?mg/L, and the plant cell membrane encountered serious damage from 500?mg/L TiO2 NP concentration in the culture media. Abstract:Nanotechnology has attracted the interest of scientists due to its wide range of application specifically in agriculture. Nanoparticles (NPs) may act as a promising materials to alleviate cadmium (Cd) stress in plants. This study aims to assess the impact of multiple nanoparticles including nSiO 2 (50 mg L −1 :100 mg L −1 ), nTiO 2 (20 mg L −1 :60 mg L −1 ), nZnO (50 mg L −1 :100 mg L −1 ), nFe 3 O 4 (100 mg L −1 :200 mg L −1 ), nCuO (50 mg L −1 :100 mg L −1 ), and nCeO 2 (50 mg L −1 :100 mg L −1 ) in combination with CdCl 2 (5 μM) to mitigate Cd toxicity in lettuce through foliar application in hydroponic solution. Current findings indicate that foliar application of nSiL + Cd (50 mg L −1 ), nZnL + Cd (50 mg L −1 ), and nTiL + Cd (20 mg L −1 ) is more effective in improving growth, biomass, root architecture, and elevated photosynthetic efficiency, which might be attributed to the increasing uptake of essential micronutrient (K, Mg, Ca, Fe, Zn) under Cd stress. Similarly, treatment with nanoparticles leads to reduced accumulation of ROS and MDA in lettuce, while enhancing the SOD, POD, CAT, and APX activities. The results showed that nanoparticles have high tolerance against Cd as depicted by the inhibition in Cd accumulation by 3.2–58% and 10–72% in roots as well as edible parts of lettuce, respectively. In addition, Cd alone reduces the morphological traits, antioxidant enzyme activity, and photosynthetic activity, while increasing the ROS, MDA, and Cd accumulation in lettuce. This comprehensive study suggests the role of nanoparticles in reducing Cd toxicity in lettuce, signifying their importance as stress mitigation agents. However, long-term pot, priming, and field trials are needed to identify the optimal nanoparticle for the lettuce under variable environmental conditions.

Abstract:Food grade titanium dioxide (TiO2) containing nanofractions, is commonly applied to whiten and brighten food products, which put consumers under health risks of ingesting TiO(2)nanoparticles (NPs). Although the oral toxicity of TiO2-NPs has been evaluated in several studies, gaps in knowledge exist regarding interactions between NPs and food components. Therefore, this study aimed to estimate the influence of TiO2-NPs on nutrient absorption and metabolism through anin situintestinal loop experiment which conducted on adultSprague Dawley(SD) rats after 30-d gastrointestinal exposure to TiO2-NPs of two different sizes (N-TiO(2)and M-TiO2). Results showed that exposure to TiO2-NPs caused flat apical membranes with sparse and short microvilli and inflammatory infiltration in small intestine. Both particles were absorbed into small intestinal cells, but N-TiO(2)with smaller size could more easily be transported through gut and raise the blood titanium (Ti) levels. Changes in serum levels of amino acid were also different after exposure to these two particles. After injecting mixed solution of nutrients intoin situintestinal loop, the N-TiO(2)exposure groups displayed significant absorption inhibition of the added histidine (His) and metabolism disorder of some non-added amino acid. However, no influence was observed on metal elements or glucose levels. This study identified TiO2-NPs with small sizes could affect nutrient absorption and metabolism by inducing intestinal epithelium injury, and amino acids were more susceptible than metal elements and glucose. These findings suggested that foods supplemented with TiO2-NPs should be carefully consumed by people with high protein requirements, such as children, the elderly, and patients with high metabolic disease or intestinal inflammation.
Impact of Tio2 Nanoparticles on Lead Uptake and Bioaccumulation in Rice (oryza Sativa L.)

Abstract:Titanium dioxide nanoparticles (TiO2 NPs) are among the most widely used metal oxide nanoparticles, and their release into the environment is inevitable. The impact of this NPs release on the fate and toxicity of coexisting heavy metals is largely unknown. In this work, a series of hydroponic experiments were conducted to investigate impact of four types of TiO2 NPs, including one anatase (NAnT), one pristine rutile (NRuT) and two rutiles with hydrophilic and hydrophobic surfaces (NLRuT, NBRuT), as well as the bulk particles (BT), on the bioaccumulation of lead (Pb) by rice (Oryza sativa) seedlings. The results indicate that TiO2 NPs exposure at the levels of 10 and 1000mg/L had no significant adverse effects on the growth and development of rice seedlings. Due to its small crystallite size (252.2Å), only one type of TiO2 NPs (NAnT) entered rice seedling roots through an apoplastic route. However, NAnT particles were not translocated from roots to shoots, likely due to the obstruction effects of casparian strip in the root tissues. Both nano and bulk TiO2 reduced the bioaccumulation of Pb in rice at high TiO2 exposure levels (1000mg/L) but the extent of the reduction was TiO2-type dependent. NAnT, NRuT and NLRuT reduced the Pb concentration in rice roots by >80% and by 77–97% in shoots, likely due to the particles high sorption potential for Pb in the nutrient solution. NBRuT and BT reduced Pb levels by 45–61% in roots and 11–38% in shoots. Although NAnT was able to effectively reduce the bioaccumulation of Pb in rice tissues, this particle did accumulate in rice roots, which poses a potential risk to food safety. Accordingly, NAnT is not recommended for Pb pollution control. NRuT and NLRuT are more appropriate agents to reduce Pb contamination threats to rice plants, which in turn may mitigate health risk to humans through the food chain.
Metabolomics Analysis of TiO 2 Nanoparticles Induced Toxicological Effects on Rice ( Oryza Sativa L.)

Abstract:The wide occurrence and high environmental concentration of titanium dioxide nanoparticles (nano-TiO2) have raised concerns about their potential toxic effects on crops. In this study, we employed a GC-MS-based metabolomic approach to investigate the potential toxicity of nano-TiO2 on hydroponically-cultured rice (Oryza sativa L.) after exposed to 0, 100, 250 or 500 mg/L of nano-TiO2 for fourteen days. Results showed that the biomass of rice was significantly decreased and the antioxidant defense system was significantly disturbed after exposure to nano-TiO2. One hundred and five identified metabolites showed significant difference compared to the control, among which the concentrations of glucose-6-phosphate, glucose-1-phosphate, succinic and isocitric acid were increased most, while the concentrations of sucrose, isomaltulose, and glyoxylic acid were decreased most. Basic energy-generating ways including tricarboxylic acid cycle and the pentose phosphate pathway, were elevated significantly while the carbohydrate synthesis metabolism including starch and sucrose metabolism, and glyoxylate and dicarboxylate metabolism were inhibited. However, the biosynthetic formation of most of the identified fatty acids, amino acids and secondary metabolites which correlated to crop quality, were increased. The results suggest that the metabolism of rice plants is distinctly disturbed after exposure to nano-TiO2, and nano-TiO2 would have a mixed effect on the yield and quality of rice.
Nickel oxide nanoparticles cause substantial physiological, phytochemical, and molecular-level changes in Chinese cabbage seedlings

However, assessing the toxicity of NiO NPs in major food crops is difficult due to the limited information available on their toxicity. The present investigation was carried out to evaluate how NiO NPs affect plant growth, photosynthetic efficiency, and phytochemical content, as well as changes at the transcriptional level of these phytochemicals in Chinese cabbage seedlings. Chlorophyll, carotenoid, and sugar contents were reduced, while proline and the anthocyanins were significantly upregulated in NiO NPs-treated seedlings. The levels of malondialdehyde, hydrogen peroxide, and reactive oxygen species, as well as peroxidase (POD) enzyme activity, were all enhanced in seedlings exposed to NiO NPs. The levels of glucosinolates and phenolic compounds were also significantly increased in NiO NPs-treated seedlings compared to control seedlings. The expression of genes related to oxidative stress (CAT, POD, and GST), MYB transcription factors (BrMYB28, BrMYB29, BrMYB34, and BrMYB51), and phenolic compounds (ANS, PAP1, and PAL) were significantly upregulated. We suggest that NiO NPs application stimulates toxic effects and enhances the levels of phytochemicals (glucosinolates and phenolic compounds) in Chinese cabbage seedlings.
A multi-omics approach reveals differences in toxicity and mechanisms in rice (Oryza sativa L.) exposed to anatase or rutile TiO2 nanoparticles

Abstract:Titanium dioxide nanoparticles (TiO2 NPs) have been widely used in agriculture, which increased the risk to soil-plant systems. Studies have demonstrated that TiO2 NPs can induce phytotoxicity. However, the toxicity mechanisms, particularly under the stress of TiO2 NPs with different crystalline forms, remain inadequately reported. In this study, we combined transcriptomics and metabolomics to analyze the toxicity mechanisms in rice (Oryza sativa L.) under the stress of anatase (AT) or rutile (RT) TiO2 NPs (50 mg/kg, 40 days). The length (decreased by 1.1-fold, p = 0.021) and malondialdehyde concentration (decreased by 1.4-fold, p = 0.0027) of rice shoots was significantly reduced after AT exposure, while no significant changes were observed following RT exposure. Antioxidant enzyme activities were significantly altered both in the AT and RT groups, indicating TiO2 NPs induced rice oxidative damage (with changes of 1.1 to 1.4-fold, p < 0.05). Additionally, compared to the control, AT exposure altered 3247 differentially expressed genes (DEGs) and 56 significantly differentially metabolites in rice (collectively involved in pyrimidine metabolism, TCA cycle, fatty acid metabolism, and amino acid metabolism). After RT exposure, 2814 DEGs and 55 significantly differentially metabolites were identified, which were collectively involved in fatty acid metabolism and amino acid metabolism. Our results indicated that AT exposure led to more pronounced changes in biological responses related to oxidative stress and had more negative effects on rice growth compared to RT exposure. These findings provide new insights into the phytotoxic mechanisms of TiO2 NPs with different crystalline forms. Based on the observed adverse effects, the study emphasizes that any form of TiO2 NPs should be used with caution in rice ecosystems. This study is the first to demonstrate that AT is more toxic than RT in paddy ecosystems, providing crucial insights into the differential impacts and toxic mechanisms of TiO2 NPs with different crystalline forms. These findings suggest prioritizing the use of RT when TiO2 NPs are necessary in agricultural development to minimize toxicity risks.

Abstract:Nanoparticle (NP) pollution has negative impacts and is a major global environmental problem. However, the molecular response of alfalfa (Medicago sativa L.) to titanium dioxide nanoparticles (TiO2 NPs) is limited. Herein, the dual effects of TiO2 NPs (0-1000 mg L-1) on carbon (C) and nitrogen (N) metabolisms in alfalfa were investigated. The results showed that 500 mg L-1 TiO2 NPs (Ti-500) had the highest phytotoxicity in the C/N metabolizing enzymes; and it significantly increased total soluble sugar, starch, sucrose, and sucrose-phosphate synthase. Furthermore, obvious photosynthesis responses were found in alfalfa exposed to Ti-500. By contrast, 100 mg L-1 TiO2 NPs (Ti-100) enhanced N metabolizing enzymes. RNA-seq analyses showed 4265 and 2121 differentially expressed genes (DEGs) in Ti-100 and Ti-500, respectively. A total of 904 and 844 differentially expressed proteins (DEPs) were identified in Ti-100 and Ti-500, respectively. Through the physiological, transcriptional, and proteomic analyses, the DEGs and DEPs related to C/N metabolism, photosynthesis, chlorophyll synthesis, starch and sucrose metabolism, and C fixation in photosynthetic organisms were observed. Overall, TiO2 NPs at low doses improve photosynthesis and C/N regulation, but high doses can cause toxicity. It is valuable for the safe application of NPs in agriculture.
Toxicity of TiO2 Nanoparticles to Escherichia Coli: Effects of Particle Size, Crystal Phase and Water Chemistry.

Abstract:Controversial and inconsistent results on the eco-toxicity of TiO2 nanoparticles (NPs) are commonly found in recorded studies and more experimental works are therefore warranted to elucidate the nanotoxicity and its underlying precise mechanisms. Toxicities of five types of TiO2 NPs with different particle sizes (10∼50 nm) and crystal phases were investigated using Escherichia coli as a test organism. The effect of water chemistry on the nanotoxicity was also examined. The antibacterial effects of TiO2 NPs as revealed by dose-effect experiments decreased with increasing particle size and rutile content of the TiO2 NPs. More bacteria could survive at higher solution pH (5.0-10.0) and ionic strength (50-200 mg L(-1) NaCl) as affected by the anatase TiO2 NPs. The TiO2 NPs with anatase crystal structure and smaller particle size produced higher content of intracellular reactive oxygen species and malondialdehyde, in line with their greater antibacterial effect. Transmission electron microscopic observations showed the concentration buildup of the anatase TiO2 NPs especially those with smaller particle sizes on the cell surfaces, leading to membrane damage and internalization. These research results will shed new light on the understanding of ecological effects of TiO2 NPs.
Titanium dioxide nanoparticles affect the growth and microRNA expression of tobacco (Nicotiana tabacum)

Abstract:Titanium dioxide (TiO2) is one of the most widely used pigments in the world. Due to its heavy use in industry and daily life, such as food additives, cosmetics, pharmaceuticals, and paints, many residues are released into the environment and currently TiO2 nanoparticles are considered an emerging environmental contaminant. Although several studies have shown the effect of TiO2 nanoparticles on a wide range of organisms including bacteria, algae, plankton, fish, mice, and rats, little research has been performed on land plants. In this study, we investigated the effect of TiO2 nanoparticles on the growth, development, and gene expression of tobacco, an important economic and agricultural crop in the southeastern USA as well as around the world. We found that TiO2 nanoparticles significantly inhibited the germination rates, root lengths, and biomasses of tobacco seedlings after 3 weeks of exposure to 0.1, 1, 2.5, and 5 % TiO2 nanoparticles and that overall growth and development of the tobacco seedlings significantly decreased as TiO2 nanoparticle concentrations increased. Overall, tobacco roots were the most sensitive to TiO2 nanoparticle exposure. Nano-TiO2 also significantly influenced the expression profiles of microRNAs (miRNAs), a recently discovered class of small endogenous noncoding RNAs (∼20–22 nt) that are considered important gene regulators and have been shown to play an important role in plant development as well as plant tolerance to abiotic stresses such as drought, salinity, cold, and heavy metal. Low concentrations (0.1 and 1 %) of TiO2 nanoparticles dramatically induced miRNA expression in tobacco seedlings with miR395 and miR399 exhibiting the greatest fold changes of 285-fold and 143-fold, respectively. The results of this study show that TiO2 nanoparticles have a negative impact on tobacco growth and development and that miRNAs may play an important role in tobacco response to heavy metals/nanoparticles by regulating gene expression.

Abstract:Applications of TiO2 nanoparticles (NPs) in food, personal care products and industries pose risks on human health, particularly on vulnerable populations including pregnant women and infants. Fetus, deficient in mature defense system, is more susceptible to NPs. Publications on the developmental toxicity of TiO2 NPs on the maternal-exposed progeny have emerged. This review presents the main exposure routes of TiO2 NPs during pregnancy, including skin penetration, ingestion and inhalation, followed by transport of TiO2 NPs to the placenta. Accumulation of TiO2 NPs in placenta may cause dysfunction in nutrient transfer. TiO2 NPs can be even transported to the fetus and generate toxicities, such as impairments of nervous and reproductive system, and failure in lung and cardiovascular development. The toxicities rely on the crystalline phase and concentrations, and the main mechanisms include the accumulation of excessive reactive oxygen species, DNA damage, and over-activation of signaling pathways such as MAPK which impairs neurotransmission. Finally, this review remarks on the significance for identifying TiO2 NPs dosage safe for both mother and fetus, and particular attention should be paid at TiO2 NPs concentrations safe for mother but toxic to fetus. Importantly, research on the epigenetic trans-generational inheritance of TiO2 NPs is urgently needed to provide insights for deciding the prospects of TiO2 NPs applications.

Abstract:As the cadmium-free semiconductor quantum dots, ZnO quantum dots (ZnO QDs) have wide potential applications in agriculture. However, the effects of ZnO quantum dots on crop growth and nutritional quality have not been fully studied. In this work, the lettuce was sprayed with different concentrations of ZnO QDs from 50 to 500 mg·L-1 to evaluate their influence on lettuce antioxidant, biomass, and nutritional quality. The results showed that ZnO QDs existed in the lettuce in the form of Zn2+. Lettuce treated with 500 mg·L-1 ZnO QDs would produce a large amount of reactive oxygen species (ROS), which adversely affected the absorption of nutrients, soluble protein content, and chlorophyll content, thus reducing plant biomass. When the concentrations range from 50 to 200 mg·L-1, the antioxidant enzyme systems of lettuce were triggered to counteract the damage caused by excessive ROS. Moreover, ZnO QDs at this level promoted Ca, Mg, Fe, Mn, Zn, and B absorption and accumulation; increased soluble sugar content; and improved the lettuce biomass and nutritional quality.

Abstract:Copper (Cu) pollution is common in the soil. Due to the widespread application of TiO2 NPs, there is a high propensity for the co-occurrence of TiO2 nanoparticles (NPs) and Cu in agricultural soils. It is therefore imperative to evaluate the joint effects of TiO2 NPs and Cu on crops. In this study, the mutual effects of TiO2 NPs and Cu on their toxicity and accumulation in soybean seedlings and on their fates in a hydroponic system were determined. When Cu was at levels of 1 and 2 mg/L, the co-occurring TiO2 NPs at a non-toxic concentration (10 mg/L) significantly enhanced the toxicity and accumulation of Cu and Ti in soybeans, and inhibited the translocation of Cu from soybean roots to shoots. However, when the Cu concentration for co-exposure was ≥ 5 mg/L, such mutual effects disappeared. The amount of Cu ions adsorbed onto TiO2 NPs after 48 h of co-exposure gradually increased from 31 to 118 mg/g when the Cu concentration was increased from 1 to 20 mg/L. The aggregation and sedimentation of TiO2 NPs were significantly increased after 48 h of co-exposure with the Cu at a concentration higher than 5 mg/L, as compared to the single TiO2 NPs exposure. The increasing aggregation and sedimentation might reduce the bioavailability of TiO2 NPs associated with the adsorbed Cu to soybeans, and consequently alleviate or even neutralize the enhanced toxicity and accumulation of Cu in soybeans exerted by the co-existing TiO2 NPs. Our results thus suggest that consideration of the impact of TiO2 NPs on the phytotoxicity of heavy metals, and specifically Cu, needs to be interpreted with care, and highlight the importance of integrating the interaction and fates of TiO2 NPs and metals into their risk assessment.
