Abstract:A novel colorimetric aptasensor based on a Au/Fe3O4 nanohybrid was developed to detect ochratoxin A (OTA). The aptasensor is composed of a free OTA aptamer, a Au/Fe3O4 nanohybrid coated with biotinylated complementary DNA of the OTA aptamer (biotin-cDNA-Au/Fe3O4), and free alkaline-phosphatase-labeled streptavidin (SA-ALP). The Au/Fe3O4 nanohybrid not only immobilizes biotin-cDNA but also magnetically separates SA-ALP from the sample solution. One part of the OTA aptamer sequence hybridizes with biotin-cDNA immobilized on Au/Fe3O4, and the left part of the OTA aptamer sequence covers the biotin and blocks the specific interaction between biotin and SA-ALP. OTA can interrupt the interaction of OTA aptamer binding to biotin-cDNA-Au/Fe3O4 and can inhibit the shielding effect of the OTA aptamer on biotin. The amount of SA-ALP that can be captured by biotin-cDNA-Au/Fe3O4 thus increases with increasing OTA concentration. Through a simple magnetic separation, the collected SA-ALP-linked Au/Fe3O4 can produce a yellow-colored solution in the presence of p-nitrophenyl phosphate (p-NPP). This colorimetric aptasensor can detect OTA as low as 1.15 ng mL(-1) with high specificity.

Abstract:In this study, the aptamer of ochratoxin A (OTA) increased the negative charge density on the surface of gold nanoparticles (AuNPs) and promoted the release of hydroxyl radicals and Au 3+ to enhance the peroxidase-like activity of the AuNPs. The OTA bound only to the aptamer and did not adsorb non-specifically to the AuNPs. Based on these two conclusions, a label-free colorimetric aptasensor was successfully developed, enabling the precise detection of OTA within the concentration range of 10–600 nM, with a remarkably low detection limit of 6.20 nM. The colorimetric aptasensor was applied to detect OTA in oats, corn, soybeans, rice, and glutinous rice. Graphical Abstract

Abstract:A novel colorimetric aptasensor based on charge effect-assisted silver enhancement was developed to detect ochratoxin A (OTA). To achieve this objective, gold nanoparticles (AuNPs), which can catalyze silver reduction and deposition, were used as the carrier of the aptamers tagged with a positively charged tetramethylrhodamine (TAMRA). Due to the mutual attraction of positive and negative charges, the TAMRA attracted and retained the silver lactate around the AuNPs. Thus, the chance of AuNP-catalyzed silver reduction was increased. The charge effect-assisted silver enhancement was verified by tagging different base pair length aptamers with TAMRA. Under optimized conditions, the as-prepared OTA aptasensor had a working range of 1 × 102–1 × 106 pg mL−1. The detection limit was as low as 28.18 pg mL−1. Moreover, the proposed aptasensor has been successfully applied to determine OTA in actual samples with satisfactory results.

Abstract:In this paper, a sensitive and specific fluorescence resonance energy transfer (FRET) aptasensor for the detection of Ochratoxin A (OTA) was developed based on a dye-tagged ssDNA hybridized with aptamer-conjugated Au nanoparticles (Au NPs). The binding between the aptamer-Au NPs conjugate and the dye-labeled ssDNA leads to the fluorescence quenching of FAM due to its close proximity. The addition of OTA results in fluorescence recovery, attributed to the formation of a quadruplex-OTA complex, which detaches from the surface of Au NPs. Under optimal conditions, the relative fluorescence intensity (Delta I) is proportional to the concentration of the OTA in the range of 5 x 10(-12) to 5 x 10(-9) g/mL, with a detection limit of 2 x 10(-12) g/mL. The proposed method was successfully applied to measure the concentration of OTA in naturally contaminated maize samples and validated using a commercially available enzyme-linked immunosorbent assay (ELISA) method. This work demonstrates that the combination of an aptamer that has a high binding affinity for the analyte with highly sensitive Au NPs that undergo FRET is a promising approach for the detection of small molecule toxins.

Abstract:With the widespread contamination of ochratoxin A (OTA), it is of significant importance for detecting OTA in foods and traditional Chinese medicine (TCM). In this study, a novel label-free fluorescent aptasensor utilizing the interaction between OTA-triggered antiparallel G-quadruplex and (N-methyl-4-pyridy) porphyrin (TMPyP) for the rapid and sensitive determination of OTA was established. The fluorescence of CdTe quantum dots (QDs) could be quenched by TMPyP. In the presence of analyte (OTA), the aptamer could recognize OTA and transform from a random coil to the antiparallel G-quadruplex. The interaction between G-quadruplex and TMPyP could release CdTe QDs from TMPyP, and thus recover the fluorescence of CdTe QDs. Under optimized conditions, the detection limit of the designed aptasensor was 0.16 ng mL-1, with a linear range of 0.2 to 20 ng mL-1. Furthermore, this aptasensor showed high selectivity toward OTA against other structural analogs and other mycotoxins, and was successfully applied in Astragalus membranaceus samples. The presented aptasensor for OTA detection could be a promising tool for the field monitoring of food and TCM.

Abstract:Colorimetric aptasensors have been intensively studied for the ochratoxin A (OTA) detection, but they mostly exhibit just one-color change, resulting in poor visual resolution and limited use for semi-quantitative analysis. Thus, we designed a high-resolution colorimetric assay on the basis of aptamer structural switching and enzyme-induced metallization of gold nanorods (AuNRs). DNA-alkaline phosphatase (ALP)-immobilized magnetic beads were prepared. The aptamer bounded to OTA to form G-quadruplexes, releasing ALP-labelled complementary DNA (cDNA-ALP). After magnetic separation, cDNA-ALP catalyzed the decomposition of ascorbic acid 2-phosphate to ascorbic acid that reduced Ag+, forming an Ag shell on the surface of AuNRs. This caused a blue-shift of the longitudinal local surface plasmon resonance peak of the AuNRs and a naked eye visible multicolor change. Under optimal conditions, the assay exhibited a 9.0 nM detection limit for OTA, with high specificity. This method is promising for the on-site visual semi-quantitative detection of mycotoxins in foods.

Abstract:An innovative colorimetric method based on enzyme-induced gold nanoparticle aggregation was developed to detect the activity of alkaline phosphatase (ALP), and it was further applied to construct an aptasensor to monitor ochratoxin A (OTA) concentrations. In the presence of ALP, the substrate ascorbic acid 2-phosphate was hydrolyzed to generate ascorbic acid (AA). Subsequently, reduction of MnO2 nanosheets by AA produced manganese ions, which mediated gold nanoparticle aggregation. The color of the detection solution changed from brown-red to purple to blue as the ALP concentration increased, and a detection limit of 0.05 U.L-1 was achieved. Furthermore, this strategy was successfully utilized to devise a target-responsive aptasensor for colorimetric detection of an important mycotoxin, OTA, which causes food poisoning and has various toxic effects on humans. The proposed method offers high sensitivity with a detection limit as low as 5.0 nM together with high specificity. When applied to analyze red wine and grape juice samples, no complex sample pretreatment or bulky instruments were required. Overall, a colorimetric platform based on enzyme-induced gold nanoparticle aggregation was successfully established to improve the simplicity and sensitivity of ALP and OTA detection. This platform appears highly promising for mycotoxin-related food safety monitoring.

Abstract:A simple, low-cost, and sensitive liposome-based colorimetric aptasensor has been developed to detect ochratoxin A (OTA). Specifically, a dumbbell-shaped probe was designed, including magnetic beads (MBs), double-stranded DNA (dsDNA), and enzyme-encapsulated liposome. The dsDNA formed by the hybridization between OTA aptamer and its complementary probe. And the dsDNA was used to contact the MBs and the enzyme-encapsulated liposome. In the presence of OTA, the aptamer preferred to combine with OTA to form G-quadruplex, resulting in the release of the detection probe and the enzyme-encapsulated liposome. Each liposome contained a large amount of HRP. Thus, when the liposome was lysed by adding the mixed solution of 3,3',5,5'-tetramethylbenzidine (TMB) and H2O2, a large number of HRP were released. HRP could catalyze the H2O2-mediated oxidation of TMB and hence resulted in the color change from colorless to blue with the OTA concentration varying, and this variation can be observed by naked eyes easily. The result showed that the absorption intensity at 652 nm enhanced with the increase of OTA concentration ranging from 0.05 to 2.0 ng mL-1, and the limit of detection was calculated to be 0.023 ng mL-1 (S/N = 3). The developed colorimetric aptasensor has been applied to detect OTA concentration in corn samples with satisfied results.

Abstract:Ochratoxin A (OTA) is a powerful mycotoxin that can cause severe damage to human health, and its detection has attracted considerable attention in the field of food science. We present a robust and facile label-free colorimetric aptasensor for OTA detection using the aptamer-enhanced oxidase-like activity of MnO2 nanoflowers. The catalytic activities of the nanozymes could be improved by adsorption of the aptamers onto the MnO2 nanoflowers due to the increased affinity of the nanoflowers for the chromogenic substrate. The linear range for OTA detection varied from 0.05 to 33.35 ng/mL with a detection limit of 0.069 ng/mL. The limit of detection of the proposed strategy is equivalent to or even better than those of several previous methods. Moreover, the colorimetric aptasensor exhibited good specificity and stability for the analysis of OTA in wheat flour and red wine samples. Therefore, this method appears to have promising applications in the detection of mycotoxins.
Aptamer-based colorimetric biosensing of Ochratoxin A using unmodified gold nanoparticles indicator.

Abstract:This work presents an aptasensor for Ochratoxin A (OTA) using unmodified gold nanoparticles (AuNPs) indicator. The assay method is based on the conformation change of OTA's aptamer in phosphate buffered saline (PBS) containing Mg2+ and OTA, and the phenomenon of salt-induced AuNPs aggregation. A single measurement took only five minutes. Circular dichroism spectroscopic experiments revealed for the first time that upon the addition of OTA, the conformation of OTA's aptamer in PBS buffer changed from random coil structure to compact rigid antiparallel G-quadruplex structure. This compact rigid G-quadruplex structure could not protect AuNPs against salt-induced aggregation, and thus the color change from red to blue could be observed by the naked eye. The linear range of the colorimetric aptasensor covered a large variation of OTA concentration from 20 to 625 nM and the detection limit of 20 nM (3 sigma) was obtained. (c) 2010 Elsevier B.V. Abstract:Ochratoxin A(OTA), a highly toxic mycotoxin commonly found in food, poses a serious threat to health even at low concentrations. Developing a sensitive, accurate, simple, and cost-effective detection method is of great significance for food safety and quality control. Herein, a triple cascade amplification strategy was used to construct the colorimetric assay for the detection of OTA, where the amplification process consists of an entropy-driven DNA circuit (EDC), a catalytic hairpin assembly (CHA), and Mg2+-assisted DNAzyme catalysis (MNAzyme). Through the specific binding of ochratoxin A (OTA) and its aptamer, an initiator strand is released to initiate upstream EDC and then produce a new trigger unit that motivates downstream CHA to generate MNAzyme, which further cleaves the substrate strand to induce the formation of G-quadruplex/hemin DNAzyme as a signal readout. The aptasensor was shown to detect OTA, with a low detection limit of 8.7 fM and good selectivity. The developed method could be used as a highly colorimetric aptasensor for the detection of OTA in spiked rice samples.

Abstract:Ochratoxin A (OTA) is a toxic pollutant in foods, and its actual detection is crucial. A novel and sensitive dual signaling ratiometric electrochemical aptasensor based on nanoporous gold (NPG) was proposed to determine OTA. NPG, with high specific surface area and conductivity, improved the sensitivity by immobilizing more aptamers. Meanwhile, the dual signaling ratiometric strategy improved the detection reproducibility through self-referencing and built-in correction. NPG and ratiometric strategy multi-amplified the dual signal changes. The sensitivity of OTA was evaluated by the ratio of methylene to ferrocene current values. Under the optimal conditions, the NPG-based aptasensor demonstrated excellent sensitivity with a wide linear range of 1 pg/mL to 2 ng/mL and the limit of detection (LOD) of 0.4 pg/mL for OTA. This developed aptasensor also effectively detected OTA in spiked Cordyceps sinensis and grape juice samples, with recovery values falling in the 98.49-108.0% range.
Facile Synthesis of Fe 3 O 4 /G-C 3 N 4 /HKUST-1 Composites As a Novel Biosensor Platform for Ochratoxin A

Abstract:A fluorescent biosensor for ochratoxin A was fabricated on the basis of a new nanocomposite (Fe3O4/g-C3N4/HKUST-1 composites). Fe3O4/g-C3N4/HKUST-1 was synthesized in this work for the first time, which combined HKUST-1 with g-C3N4 to improve its chemical stability. Fe3O4/g-C3N4/HKUST-1 composites have strong adsorption capacity for dye-labeled aptamer and are able to completely quench the fluorescence of the dye through the photoinduced electron transfer (PET) mechanism. In the presence of ochratoxin A (OTA), it can bind with the aptamer with high affinity, causing the releasing of the dye-labeled aptamer from the Fe3O4/gC(3)N(4)/HKUST-1 and therefore results in the recovery of fluorescence. The fluorescence intensity of the biosensor has a linear relationship with the OTA concentration in the range of 5.0-160.0 ng/mL. The LOD of sensor is 2.57 ng/mL (S/N=3). This fluorescence sensor based on the Fe3O4/g-C3N4/HKUST-1 composites has been applied to detect OTA in corn with satisfying results.

Abstract:Ochratoxin A (OTA) is a harmful mycotoxin, which is mainly secreted by Penicillium and Aspergillus species. In this work, an electrochemical aptasensor is presented for OTA detection based on Au nanoparticles (AuNPs) modified zeolite imidazolate frameworks (ZIFs) ZIF-8 platform and duplex-specific nuclease (DSN) triggered hybridization chain reaction (HCR) signal amplification. G-quadruplex-hemin assembled HCR nanowire acted as a nicotinamide adenine dinucleotide (NADH) oxidase and an HRP-mimicking DNAzyme. Besides, thionine (Thi) was enriched as a redox probe for signal amplification in this pseudobienzyme electrocatalytic system. Under the optimal conditions, the analytical response ranged from 1 to 107 fg ml−1 with a detection limit of 0.247 fg ml−1. Furthermore, the aptasensor was proven to be applied in real wheat samples with a recovery between 96.8 and 104.2%, illustrating the potential prospects in practical detection.
A Nanozyme-Based Cascade Colorimetric Aptasensor for Amplified Detection of Ochratoxin A

Abstract:Colorimetric assays have been widely developed for the detection of toxin ochratoxin A (OTA), but most of them suffer from moderate sensitivity when they are adopted for the detection of trace OTA in a complicated food matrix. For the purpose of overcoming this issue, an innovative cascade reaction-based colorimetric aptasensor was developed for the achievement of high sensitivity. The biotin-labelled OTA aptamer was immobilized onto streptavidin magnetic beads by means of the biotin-streptavidin reaction. With OTA binding to its aptamer, the structural switching of the aptamer results in the release of the alkaline phosphatase-labelled oligonucleotide, which is partially complementary to the aptamer. Following the magnetic separation, the cascade reaction is initiated through the enzymatic conversion of ascorbic acid-2-phosphate into ascorbic acid. Subsequent to that, the generated ascorbic acid reduces MnO2 nanosheets to Mn2+ ions, accordingly destroying the oxidase-mimicking activity of MnO2 nanosheets. In consequence, it is not possible to oxidize 3,3',5,5'-tetramethylbenzidine (TMB), a substrate for oxidase, with Mn2+ for the production of the blue colour product (TMB Ox). With the increasing amount of OTA, a colour change occurs from blue to colourless. The cascade reaction has the potential of greatly amplifying the detection signal, together with remarkably improving the sensitivity, making this colorimetric sensor a universal and promising platform for the highly sensitive detection of mycotoxins in the field of public food safety monitoring.

Abstract:A stainless steel mesh (SSM) with the feature of flexibility was employed as the colorimetric biosensor substrate, and aptamer was bond onto the surface of the SSM. Through the cross-linking of ionic liquids (ILs), AuPt nanoparticles were deposited onto the surface of Fe 3 O 4 material to obtain a magnetic nanozyme with high peroxidase catalytic activity and rapid color change. Through the competing interaction of OTA and cDNA with aptamer, AuPt@IL@Fe 3 O 4 signal probe was separated to catalyze the 3,3′,5,5′-tetramethylbenzidine/hydrogen peroxide (TMB/H 2 O 2 ) system to observe the color by bare eye and record the absorbance at 652 nm using a UV-spectrophotometer. Through the study of the catalytic properties on the basis of the Michaelis equation, AuPt@IL@Fe 3 O 4 nanozyme presented a V max of 3.85 × 10 −8 M s −1 and K m of 0.01 mM. Under the optimized conditions, the linear range of the colorimetric biosensor towards OTA was 5–100 ng mL −1 , and the detection limit was 0.078 ng mL –1 . This biosensor was applied to beer and corn samples with recoveries of 70.4–102.6% and 93.3–104.7%, respectively. Results showed that this sensor is a portable, rapid, economical, sensitive visual sensing platform towards mycotoxin in real samples.
An "on-off-on" electrochemiluminescence aptasensor based on a self-enhanced luminophore for ochratoxin A detection

Abstract:A highly selective and sensitive "on-off-on" electrochemiluminescence (ECL) aptasensor based on a self-enhanced luminophore was developed for the detection of ochratoxin A (OTA). Specifically, polyethyleneimine functionalized multi-walled carbon nanotubes decorated with gold nanoparticles (AuNPs-PEI-MWCNTs) were used as the electrode matrix to accelerate electron transfer and provide a favorable microenvironment for self-enhanced luminophore loading and ECL signal enhancement. In addition, black phosphorus quantum dots (BPQDs) were used as co-reactants of the ECL reagent tris (2,2'-bipyridyl) ruthenium(II) (Ru(bpy)32+) in ECL experiments, and the reaction mechanism was investigated. The self-enhanced luminophore Ru@SiO2-BPQDs was obtained by encapsulating Ru(bpy)32+ in silica (SiO2) nanoparticles and then combining it with BPQDs through electrostatic interaction. In conventional ECL systems, the emitter and its co-reactants reacted via the inter-nanoparticle pathway, leading to long distance electron transfer. However, the electron transfer distance in the self-enhanced luminophore was significantly shortened due to the intra-nanoparticle electron transfer pathway because BPQDs and oxidized Ru(bpy)32+ were bound within one nanoparticle, thereby improving ECL efficiency to achieve the first "switch-on" state. Then, the luminophore was quenched using ferrocenes (Fc) modified on an aptamer to achieve the "switch-off" state. Finally, OTA was specifically identified by the adapter, causing Fc to be released from the sensor interface, restoring the ECL intensity to achieve the second "switch-on" state. Under optimal conditions, the aptasensor exhibited good sensitivity, stability, and reproducibility, with a linear detection range from 0.1 to 320 ng/mL and a detection limit of 0.03 ng/mL. The novel ECL aptasensor provided a common analytical tool for the detection of mycotoxins and other small molecules.

Abstract:Ochratoxin is teratogenic, carcinogenic and immunotoxic to humans. It contains ochratoxin in many foods, so the detection of ochratoxin in food is particularly important. In this paper, gold nanoparticles (Au NPs)@g-C3N4 composite was synthesized by loading Au NPs on carbon nitride material, and it was immobilized on the surface of glassy carbon electrode by chitosan (Chit) as the substrate of electrochemical aptasensors. An ochratomycin A electrochemical aptasensor was constructed by hybridizing DNA1 and ochratoxin A (OTA) aptamers. The resulting hybrid strands were immobilized on the substrate of glassy carbon electrode. Electrochemical alternating current impedance (EIS) was used to detect the impedance value of the aptasensor when incubating different concentrations of OTA. The impedance value is inversely ratio to the concentration of OTA, achieving quantitative detection of OTA. The aptasensor can detect OTA with a linear range of 0.5–100 ng/mL, the linear correlation coefficient is 0.9506, and the detection limit is 0.167 ng/mL. This aptasensor provides a novel and efficient method for detecting OTA.

Abstract:In this paper, an ultrasensitive photoelectrochemical aptasensor based on amplified signal strategy and competitive mechanism for ochratoxin A (OTA) detection was developed. The prepared photoactive composite material (BiFeO3/Bi2S3) possessed excellent separation ability of photogenerated electron-hole pair and effectively inhibited the recombination of hole and pair, resulting in significantly enhanced photocurrent intensity. The amplified signal strategy was achieved by adding methylene blue (MB), which facilitated electron transfer and improved the photoelectric conversion efficiency. In the presence of OTA, the aptamer can be recognized and captured, the complementary DNA (cDNA) and MB were released from the aptasensor electrode, a decreased photocurrent signal appeared, thus achieving quantitative detection for OTA. Under optimal conditions, the aptasensor exhibited a wide linear relationship range from 10 5 to 10 13 g/mL and a low detection limit of 0.0135 fg/mL (3s/S). Simultaneously, the proposed photoelectrochemical aptasensor also showed excellent accuracy, stability, precision and selectivity.

Abstract:In this study, a novel aptasensor based on Aptamer/NH2 Janus particles is developed for the detection of Ochratoxin A(OTA). By coating gold on the hemispherical surface of the aminated polystyrene particles, Ochratoxin A aptamer is immobilized on the surface of the gold layer for selective identification and the other hemispherical able to bind to Glassy carbon electrode via peptide bond. Under optimum conditions, the sensor exhibited a wide dynamic range of OTA concentration from 1 x 10(-5) nM to 10 nM, and the detection limit is 3.3 x 10(-3) pM on condition of acceptable stability and reproducibility. The sensors were showed excellent performance in the detection of OTA in red wine sample with recoveries between 95.7% and 100.18%, which studied by the standard addition spiking technique. This work provides a new idea and method for preparing immune electrochemical sensors and is expected to be used for the OTA detection in red wine sample analysis.
