P4‐129: CHEMOKINE LIKE RECEPTOR 1 DEFICIENCY IMPROVES COGNITION AND ATTENUATES TAU HYPERPHOSPHORYLATION IN MOUSE MODELS OF ALZHEIMER'S DISEASE

Abstract:Pathological features of Alzheimer's disease (AD) include aggregation of amyloid beta (Aβ) and tau protein. Chemokine-like receptor 1 (CMKLR1) is a G protein-coupled receptor, expressed mainly in mononuclear cells and adipocytes. Our previous study have found that CMKLR1 also expressed in brain and is a functional receptor of Aβ. However, it remains unclear whether CMKLR1 affects cognition and tau phosphorylation. The effects of CMKLR1 in cognition and tau protein were examined in Cmklr1−/- mice receiving intracerebroventricular (ICV) injection of streptozotocin (STZ) and in APP/PS1 transgenic mice mated with Cmklr1−/- mice. Marris water maze, open field test, elevated plus maze, and rotarod test were used to detect the learning and memory, anxiety, space exploration, and motion balance ability respectively. The phosphorylation of tau protein was examined by Western Blotting (WB). The expression of CMKLR1 were determined by immunofluorescence (IF). ICV-TAU mouse model was established to verify the effect of CMKLR1 in tau seeding. CMKLR1 deficiency significantly increased the survival rate of APP/PS1+/- mice. CMKLR1 deficiency improved the cognitive ability of the mice of these two AD models, but had no effect on the anxiety of the mice. In addition, CMKLR1 deficiency alleviated the abnormal hyperphosphorylation of tau in the brains of these AD model mice. Inhibition of CMKLR1 also attenuated hyperphosphorylation of tau protein induced by high glucose treatment in SH-SY5Y cells. CMKLR1 deficiency inhibited hyperphosphorylation of tau protein in hippocampus and cortex of ICV-TAU mice. These results indicate that CMKLR1 deletion can ameliorate cognitive impairment in AD. CMKLR1 mediates the phosphorylation level of tau protein and the transmission of phosphorylate tau by tau seeding. Therefore, inhibition of CMKLR1 may be useful in AD prevention and treatment.
