Abstract:Abstract Respiration is the center of energy metabolism in mammals, and mitochondrial respiratory chain complexes play a major role in energy conversion. According to the high‐resolution structures of individual complexes and the assembled supercomplex resolved recently, we can now try to elucidate the electron transfer and coupled proton pumping mechanism at molecular level. Together with previously accumulated biochemical data, although an overall agreement on the electron pathways was reached, some vital details are still controversial, including the ubiquinone reduction manner in complex I and ubiquinol binding position in CIII. Besides, recently our group reported the first medium‐resolution structure of megacomplex from cultured human cells, showing that two sets of complex I, III, IV and even II may function in a coordinated manner, making the electron pathway even more complex. In this article, we discuss electron transfer between individual complexes in respirasome, describe the megacomplex model, and compare two different electron pathways inside complex III.
